In order to create a perspective drawing on a curved surface, the quadraturista had to face, in addition to the flat perspective drawing issues, also the ones related to the mapping of the three dimensional surface. In literature we can find two methodologically different approaches related to the flat design transposition procedures onto the curved surface: indirect and direct method. The indirect one, partially described by Vignola, is based on the use of pre-designed cartoons on the ground. The direct one, described by Andrea Pozzo, is based on the direct projection of a flat grid onto the curved surface. Both the ceiling painting of the Church of San Matteo in Pisa and the parietal one of the Church of S. Caterina in Livorno show, even if observed from the privileged viewpoints, some irregularities. These irregularities, as we are going to show, are actually the consequences of the method applied for the transposition of the flat drawings onto the curved surfaces.  相似文献

The effect of a chitosan coating with added essential oil of oregano (Origanum vulgare) on lipid oxidation of dry fermented sausage (Petrovská klobása) was investigated. The aim of this research was to prove the function of the putative opine dehydrogenase from Desulfohalobium retbaense and to characterize the enzyme in terms of functional and kinetic parameters. A putative opine dehydrogenase was identified from a metagenomic library by a sequence-based technique search of the metagenomic library, and afterward was successfully heterologously produced in Escherichia coli. In order to examine its potential for applications in the synthesis of secondary amines, first the substrate specificity of the enzyme towards different amino donors and amino acceptors was determined. The highest affinity was observed towards small amino acids, preferentially L-alanine, and when it comes to α-keto acids, pyruvate proved to be a preferential amino acceptor. The highest activity was observed at pH 6.5 in the absence of salts. The enzyme showed remarkable stability in a wide range of experimental conditions, such as broad pH stability (from 6.0–11.0 after 30 min incubation in buffers at a certain pH), stability in the presence of NaCl up to 3.0 M for 24 h, it retained 80 % of the initial activity after 1 h incubation at 45 °C, and 65 % of the initial activity after 24 h incubation in 30 % dimethyl sulfoxide.  相似文献

Several types of nanostructured hybrid fibrous materials containing poly(3-hydroxybutyrate), nanoparticles from iron oxide (Fe3O4) and titanium dioxide (TiO2), and chitosan or chitosan oligosaccharides (COS) were prepared. The design of the surface of the materials and their magnetic properties were tailored purposefully by conjunction of electrospinning and electrospraying. The surface and bulk morphologies of the obtained nanostructured materials were examined by SEM. Further, the distribution of Fe3O4 and TiO2 nanoparticles was estimated by TEM analyses, as well as their surface chemical composition was determined by XPS. It was found that the simultaneous electrospinning and electrospraying of Fe3O4/chitosan or TiO2/COS dispersions resulted in uniform distribution of the nanoparticles along the length of the fibers, while electrospraying of the mixed Fe3O4/TiO2/chitosan dispersion led to agglomerate formation. Furthermore, the nanostructured hybrid materials preserved the magnetic properties of Fe3O4 embedded therein. It was demonstrated that the hybrid materials of different designs displayed excellent photocatalytic activity under UV light irradiation against a model organic contaminant—methylene blue, even after threefold use of the materials.  相似文献

Our objective is to reveal the molecular mechanism of the anti-inflammatory action of low-molecular-weight heparin (LMWH) based on its influence on the activity of two key cytokines, IFNγ and IL-6. The mechanism of heparin binding to IFNγ and IL-6 and the resulting inhibition of their activity were studied by means of extensive molecular-dynamics simulations. The effect of LMWH on IFNγ signalling inside stimulated WISH cells was investigated by measuring its antiproliferative activity and the translocation of phosphorylated STAT1 in the nucleus. We found that LMWH binds with high affinity to IFNγ and is able to fully inhibit the interaction with its cellular receptor. It also influences the biological activity of IL-6 by binding to either IL-6 or IL-6/IL-6Rα, thus preventing the formation of the IL-6/IL-6Rα/gp130 signalling complex. These findings shed light on the molecular mechanism of the anti-inflammatory action of LMWH and underpin its ability to influence favourably conditions characterised by overexpression of these two cytokines. Such conditions are not only associated with autoimmune diseases, but also with inflammatory processes, in particular with COVID-19. Our results put forward heparin as a promising means for the prevention and suppression of severe CRS and encourage further investigations on its applicability as an anti-inflammatory agent.  相似文献

The degradation of poly(oxyethylene)–poly(ε-caprolactone) (POE–PCL) multiblock copolymers was investigated at 37°C in a 0.13M, pH 7.4 phosphate buffer selected to mimic in vivo conditions. The copolymers were obtained by coupling polycaprolactone diols and poly(ethylene glycol) diacids using dicyclohexylcarbodiimide as coupling agent. Various techniques, such as weighing, size exclusion chromatography, infrared, 1H nuclear magnetic resonance, differential scanning calorimetry, and X-ray diffractometry, were used to monitor changes in total mass, water absorption, molar mass, thermal properties, degree of crystallinity, and composition. The results showed that introduction of POE sequences considerably increased the hydrophilicity of the copolymers as compared with PCL homopolymers. Nevertheless, the degradability of PCL sequences was not enhanced due to the phase separation between the two components. The protective ability of novel arylpiperazine-based dopaminergic ligands against nitric oxide (NO)-mediated neurotoxicity is investigated. The most potent neuroprotective arylpiperazine identified during the study was N-{4-[2-(4-phenyl-piperazin-1-yl)ethyl]-phenyl}picolinamide, which protected SH-SY5Y human neuron-like cells from the proapoptotic effect of NO donor sodium nitroprusside (SNP) by decreasing oxidative stress, mitochondrial membrane depolarization, caspase activation and subsequent phosphatydilserine externalization/DNA fragmentation. The protective effect was associated with the inhibition of proapoptotic (JNK, ERK, AMPK) and activation of antiapoptotic (Akt) signaling pathways, in the absence of interference with intracellular NO accumulation. The neuroprotective action of arylpiperazines was shown to be independent of dopamine receptor binding, as it was not affected by the high-affinity D?/D? receptor blocker butaclamol. These results reported support the further study of arylpiperazines as potential neuroprotective agents.  相似文献

The mechanisms underlying the beneficial effect observed in diabetic rats after treatment with a commercially available β-glucan-enriched extract (BGEE) were examined. Multiple low-dose streptozotocin (STZ) diabetes was used (40 mg STZ/kg) as a model for type 1 diabetes. BGEE was administered daily (80 mg/kg) for 4 weeks, starting from the last day of the STZ treatment. In vitro and in vivo experiments suggest that significant free radical scavenging and antioxidant activities of BGEE were responsible for a systemic adjustment of the redox disturbance and reduction of DNA damage in the liver and kidney of diabetic rats. BGEE-treated diabetic rats also displayed increased Akt kinase activity and decreased pro-caspase-3 degradation, implying that BGEE mediates its beneficial effects through activation of the cellular pro-survival pathway. We conclude that β-glucan administration under diabetic conditions promotes a systemic improvement that can be expected to increase the organism’s resistance to the onset of diabetic complications.  相似文献
