Maturation of Peyer's patch dendritic cells in vitro upon stimulation via cytokines or CD40 triggering

In mouse Peyer's patches (PP), dendritic cells (DC) are localized in T cell areas as NLDC145+ CD11c+ cells, and in the dome and corona region of the follicle as NLDC145- CD11c+ cells, respectively, suggesting the presence of two different DC populations with distinct roles in antigen uptake, processing, and presentation. However, it is not clear how this relates to DC maturation. In this report, we demonstrate that freshly-isolated CD11c+ DC have the properties of immature DC since they endocytose soluble antigens, phagocytose particulate material such as latex beads, synthetize major histocompatibility complex (MHC) class II and invariant chain, but, at the same time, display low stimulatory activity for resting T cells, as shown in mixed-lymphocyte reaction and oxidative mitogenesis assays. When cultured for 24 h in the presence of the cytokines granulocyte-macrophage colony-stimulating factor and tumor necrosis factor or anti-CD40, the cells undergo dramatic phenotypic and functional changes characteristic of DC maturation. After 24 h stimulation in vitro, CD11c+ cells lose the ability to take up proteins such as ovalbumin, and in parallel with this decline, the biosynthesis of MHC class II and invariant chain is dramatically down-regulated or eliminated. On the other hand cells treated in vitro exhibit on the cell surface higher levels of MHC class II, of co-stimulatory molecules (CD80, CD86), of adhesion molecules (CD44, intercellular adhesion molecule-1), and acquire expression of the interdigitating DC surface marker NLDC145. Concomitantly, the ability to stimulate naive T cells drastically increased after in vitro treatment with both stimuli. Taken together, our results indicate that the majority of DC in the PP are immature in terms of their antigen-uptake capacity. These sentinel antigen presenting cells are strategically positioned at the dome region of PP, where antigens are transcytosed via the M cells from the gut lumen. A second population of mature interdigitating NLDC145+ CD11c+ DC stimulates naive unprimed T cells in interfollicular areas by up-regulation of surface ligands and accessory signals.     

Nutritional response of mature rats to kidney bean (Phaseolus vulgaris) lectins

In a series of 10-day pair feeding experiments it was found that the nutritional value of diets containing beans was essentially the same for rats aged between 30 and 123 days. Thus net protein utilisation (NPU) values of 25–39 on diets containing Processor bean (35 g protein kg^?1) + egg albumin (65g protein kg^?1) were obtained. As food intakes were considerably reduced when rats were fed diets containing more than 35g protein kg^?1 of Processor bean, the measurement of protein utilisation became increasingly more difficult. The severe disruption of the brush borders of duodenal and jejunal enterocytes, originally observed when bean-containing diets were fed to young (30-day-old) rats was also found with rats up to the age of 120 days on similar diets. Similarly, the development of circulating anti-lectin antibodies in the animals showed no age dependence within the age limits investigated. It was also shown that oral immunisation did not protect the rats from the effects of toxicity and that the immune response was a result of continuous absorption of lectin throughout the feeding period. Thus the extent and the mechanism of toxicity of Phaseolus vulgaris bean lectins were found not to be dependent on the age or maturity of the animal.     

Potential applications of poly(vinyl alcohol)‐congo red aqueous solutions and hydrogels as liquids for hydraulic fracturing

The authors report on the viscoelastic characterization of guar hydrogels obtained through complexation reactions with borax ions. These gels are compared with hydrogels obtained from poly(vinyl alcohol) of different degree of hydrolysis through complexation reactions with congo red. The effect of the degree of hydrolysis and both, the concentration of PVA and the concentration of congo red, on the viscoelastic properties of the hydrogels is analyzed. The potential use of the PVA‐based hydrogels as hydraulic fracturing liquids is discussed in relation to the commonly used fracturing liquid based on the guar–borax system. © 2008 Wiley Periodicals, Inc. Putative liver progenitor cells: conditions for long-term survival in culture

BACKGROUND AND OBJECTIVES: We operated on three patients with leiomyosarcoma of the inferior vena cava. METHODS: Complete excision was possible in all three patients. RESULTS: One patient developed widespread metastasis at 23 months, one patient is alive with no evidence of disease at 70 months, and one patient is alive at 15 months. The third patient had subcutaneous and pulmonary metastases at the time of presentation, which are radiologically nondetectable at present following postoperative chemotherapy. CONCLUSIONS: The clinicopathologic features, prognostic factors, and treatment of 130 cases found in a comprehensive literature search and our three cases are reported.     

Cpk index estimation using data transformation

Process capability indices (PCIs) are used in industry to assess percentages of nonconforming parts. An underlying assumption is that the output process measurements are distributed as normal random variables. When normal distributions are assumed, but different distributions are present - such as skew, heavy-tailed, and short-tailed distributions - the percentages of nonconforming parts are significantly different than the computed PCIs indicate. Data arising from nonnormal distributions can sometimes be transformed to conform to the normality assumption and the PCI's computed for the transformed data. In this paper, the effect of the transformation on the estimate of nonconforming parts is examined for three examples of nonnormal distributions - gamma, lognormal, and Weibull. The results of this experimental analysis suggest that data transformation can be useful for estimating an interval for C_pk values and the number of nonconforming parts.     

Generalized thermoelastic diffusion in a nanoscale beam using eigenvalue approach

In this paper, the effect of mass diffusion in a thermoelastic nanoscale beam in context Lord and Shulman theory is studied. The analytical solution in the Laplace domain is obtained for lateral deflection, temperature, displacement, concentration, stress and chemical potential. The both ends of the nanoscale beam are simply supported. The basic equations have been written in the form of a vector-matrix differential equation in the Laplace transform domain, which is then solved by an eigenvalue approach. The results obtained are presented graphically for the effect of time and mass diffusion to display the phenomena physical meaning.     

Practical entanglement concentration of nonlocal polarization-spatial hyperentangled states with linear optics

We present some different hyperentanglement concentration protocols (hyper-ECPs) for nonlocal N-photon systems in partially polarization-spatial hyperentangled states with known parameters, resorting to linear optical elements only, including those for hyperentangled Greenberger–Horne–Zeilinger-class states and the ones for hyperentangled cluster-class states. Our hyper-ECPs have some interesting features. First, they require only one copy of nonlocal N-photon systems and do not resort to ancillary photons. Second, they work with linear optical elements, neither Bell-state measurement nor two-qubit entangling gates. Third, they have the maximal success probability with only a round of entanglement concentration, not repeating the concentration process some times. Fourth, they resort to some polarizing beam splitters and wave plates, not unbalanced beam splitters, which make them more convenient in experiment.
