A probabilistic model for cleavage fracture with a length scale--parameter estimation and predictions of stationary crack experiments 总被引:1,自引:0,他引:1
This study presents a large experimental investigation in the transition temperature region on a modified A508 steel. Tests were carried out on single-edge-notch-bend specimens with three different crack depth over specimen width ratios to capture the strong constraint effect on fracture toughness. Three test temperatures were considered, covering a range of 85 °C. All specimens failed by cleavage fracture prior to ductile tearing. A recently proposed probabilistic model for the cumulative failure by cleavage was applied to the comprehensive sets of experimental data. This modified weakest link model incorporates a length scale, which together with a threshold stress reduce the scatter in predicted toughness distributions as well as introduces a fracture toughness threshold value. Model parameters were estimated by a robust procedure, which is crucial in applications of probabilistic models to real structures. The conformity between predicted and experimental toughness distributions, respectively, were notable at all the test temperatures. 相似文献

In this report, we demonstrate the utility of interleukin-2 (IL-2) stimulation of spleen cell cultures and bivariate flow cytometry in the analysis and purification of the C57BL/6J mouse Y Chromosome. We determined that the DNA content of the C57BL/6J Y Chromosome is approximately 94.7 Mb, making it similar in size to human Chromosome 16 and significantly larger than previous estimates. In addition, we describe the bulk isolation of mouse Y Chromosomes and demonstrate enrichment of the isolated material using a fluorescence in situ hybridization strategy. We detail the construction of two small insert Y Chromosome-specific libraries, ideal for sampling Y Chromosome sequences. From these libraries 1566 clones were analyzed. We provide a detailed characterization of 103 clones, generating nearly 50 kb of sequence. For 30 of these clones, we identify regions of homology to known Y chromosomal sequences, confirming the enrichment of the sorted DNA. From the remaining characterized clones, we describe the development of 15 male-specific PCR assays and 19 male-female PCR assays potentially originating from the pseudoautosomal region or other areas of X-Y or autosome-Y homology. 相似文献

Relying on SIDE theory, this 2 × 2 experimental design tested the effects of adherence to organizational norms and interpersonal familiarity in e‐mail messages from both superior and subordinate perspectives. Results reveal that using norm‐congruent e‐mail messages account for over 50% of the variance in both superior and subordinate attitudes toward the message and between 30% and 56% of the variance in perceptions of source credibility. Data from the superiors indicate that norm congruent e‐mail messages account for 14.4% of the variance in message compliance. Results from subordinates reveal an interaction effect that is consistent with SIDE‐based predictions. Subordinates believe superiors are most likely to comply with e‐mail requests from unfamiliar subordinates that are crafted in a manner congruent with organizational norms. 相似文献

A new plasticity model with a yield criterion that depends on the second and third invariants of the stress deviator is proposed. The model is intended to bridge the gap between von Mises’ and Tresca’s yield criteria. An associative flow rule is employed. The proposed model contains one new non-dimensional key material parameter, that quantifies the relative difference in yield strength between uniaxial tension and pure shear. The yield surface is smooth and convex. Material strain hardening can be ascertained by a standard uniaxial tensile test, whereas the new material parameter can be determined by a test in pure shear. A fully implicit backward Euler method is developed and presented for the integration of stresses with a tangent operator consistent with the stress updating scheme. The stress updating method utilizes a spectral decomposition of the deviatoric stress tensor, which leads to a stable and robust updating scheme for a yield surface that exhibits strong and rapidly changing curvature in the synoptic plane. The proposed constitutive theory is implemented in a finite element program, and the influence of the new material parameter is demonstrated in two numerical examples. 相似文献

Inspired by the incorporation of metallocene functionalities into a variety of bioactive structures, particularly antimicrobial peptides, we endeavored to broaden the structural variety of quaternary ammonium compounds (QACs) by the incorporation of the ferrocene moiety. Accordingly, 23 ferrocene-containing mono- and bisQACs were prepared in high yields and tested for activity against a variety of bacteria, including Gram-negative strains and a panel of clinically isolated MRSA strains. Ferrocene QACs were shown to be effective antiseptics with some displaying single-digit micromolar activity against all bacteria tested, demonstrating yet another step in the expansion of structural variety of antiseptic QACs. 相似文献

Peanut allergy represents one of the most severe IgE-mediated reactions with food, but to date, the only effective way to prevent peanut allergy is total avoidance. If allergens could be mitigated during food processing before a product reaches the consumer, this would substantially lessen the food allergy problem. The efficacy of pulsed ultraviolet light (PUV), a novel food processing technology, on reducing peanut allergens, was examined. This study revealed for the first time that PUV was also capable of deactivating Ara h 2, the most potent allergenic protein of peanut. Protein extracts from raw and roasted peanuts were treated for 2, 4, and 6?min and peanut butter slurry was treated for 1, 2, and 3?min in a Xenon Steripulse XL 3000? PUV system. The distance from the central axis of the lamp was varied at 10.8, 14.6, and 18.2?cm. The SDS?CPAGE showed a reduction in the protein band intensity for Ara h 1, Ara h 2, and Ara h 3 at the energy levels ranging from 111.6 to 223.2?J/cm2. Reduction of the protein band intensity for peanut allergens increased with treatment time but decreased with increased distance from the PUV lamp. The ELISA for peanut extracts and peanut butter slurry showed a reduction in IgE binding of up to 12.9- and 6.7-folds, respectively, compared to control. 相似文献

The profiles of tocopherol (T) and tocotrienol (T3) homologues in 37 samples of seven different types of bran (rye, wheat, oat, spelt, buckwheat, rice, and corn), available on the Polish market, were studied. Tocochromanols were identified and quantified by reverse phase-high-performance liquid chromatograph/fluorescence detector and reverse phase-ultra performance liquid chromatography-electrospray ionization/mass spectrometry. Only rice bran contained all eight tocochromanol types. Corn bran lacked β-T3; rye, wheat, oat, and spelt bran lacked γ-T3 and δ-T3; and buckwheat bran lacked β-T3, γ-T3, and δ-T3. In buckwheat and corn bran tocopherols predominated (98 and 78%, respectively); whereas rye, wheat, oat, spelt, and rice bran were rich in tocotrienols (78, 76, 66, 87, and 66%, respectively). The average total tocochromanol contents in the oat, corn, spelt, buckwheat, wheat, rye, and rice bran were 5.5, 16.2, 15.8, 14.7, 12.8, 10.7, and 9.1 mg/100 g of dry weight, respectively. Tocochromanol concentrations in samples of the same type bran from different sources varied considerably. Better labeling of bran products to reflect this variation would assist with control of vitamin E daily dietary requirements. 相似文献
