OBJECTIVE AND DESIGN: We reported previously that the betamethasone derivative betamethasone dipropionate behaves as an anti-glucocorticoid in rat endotoxin-induced uveitis (EIU). In the present study, we produced EIU in guinea pigs and investigated the effects of betamethasone dipropionate on the EIU. MATERIAL: Male Hartley guinea pigs were used. TREATMENT: Glucocorticoids were instilled into the eye. METHOD: To elicit EIU, lipopolysaccharide (LPS) was injected into the anterior chamber of the eye. Cell numbers in the aqueous humor after LPS injection were determined by flow cytometry. Prostaglandin E2 (PGE2) production after LPS injection into the anterior chamber was also examined. RESULTS: Intracameral injection of LPS (1 microgram/eye) induced cell infiltration into the anterior chamber and PGE2 production. Betamethasone dipropionate inhibited cell infiltration and PGE2 production more strongly than betamethasone. These results suggest that betamethasone dipropionate is a potent glucocorticoid in guinea pigs. CONCLUSIONS: Structure-activity relationships of glucocorticoids in the guinea pig EIL model may differ from those in the rat EIU model.  相似文献

To determine which types of voltage-dependent Ca2+ channels mediate tachykinin release in the isolated rabbit iris sphincter muscle, we examined the effects of several Ca2+ channel modulators on contractions induced by either an elevation of the extracellular KCl concentration or application of the Na+ channel activator veratridine. Contractions caused by either 45.9 mM KCl or veratridine (10 microM) were inhibited by spantide (10 microM), a tachykinin receptor antagonist, and capsaicin (10 microM), a tachykinin-depleting agent, but were not changed by atropine. Nicardipine, an L-type Ca2+ channel blocker, inhibited contractions induced by KCl and veratridine in a concentration-dependent manner. omega-Conotoxin GVIA (1 microM), an N-type Ca2+ channel blocker, inhibited only contractions induced by lower concentrations of KCl, both when applied alone and when combined with nicardipine. Bay K 8644, an L-type Ca2+ channel activator, caused a spantide- and nicardipine-sensitive contraction in muscles partially depolarized with 15.9 mM KCl, and enhanced contractions induced by 15.9 mM KCl and veratridine (2 microM). omega-Agatoxin IVA (0.3 microM), a P-type voltage-dependent Ca2+ channel blocker, did not affect contractions induced by either KCl or veratridine. Contractions induced by exogenous substance P were not modified by any of the Ca2+ channel blockers or by Bay K 8644. These results suggest that, in the rabbit iris sphincter muscle. L- and N-type voltage-dependent Ca2+ channels are involved in neurotransmitter release from tachykininergic nerves elicited by high KCl and by veratridine.  相似文献

Nonpolar polymers such as polyethylene or polypropylene are widely used as insulation materials, often under conditions involving strong electric fields and high temperatures. Under these conditions, the conduction loss due to DC-like carrier transport may give rise to a high-field AC dissipation factor (tan δ). This paper describes the high-field dielectric properties of polyethylene films at frequencies from 50 to 400 Hz and temperatures from room temperature to 100°C, as measured with a new type electrode design developed by the authors. In the high-temperature region, the AC dissipation current waveform at 50 Hz was also measured. At room temperature, the electric-field dependence of tan δ is almost unaffected by the frequency of the applied field. But at high temperatures and strong electric fields, tan δ tends to have a large field dependence and to be almost inversely proportional to the frequency. Thus the AC conductivity becomes independent of the frequency of the applied field in the region of strong fields and high temperatures. A theoretical analysis shows that the high-field dielectric loss is governed by two processes, namely, the dielectric relaxation loss inherent to the AC field and the DC-like carrier transport loss that becomes prominent in the high-temperature region, where AC dissipation current waveform becomes significantly nonsinusoidal.  相似文献

PURPOSE: Several lines of evidence suggest that the infiltration of macrophages and the expression of alpha-smooth muscle actin in myofibroblasts play important roles in the pathogenesis of tubulointerstitial fibrosis. However, the temporal sequence of these pathological changes or the dynamics in the expression of this cytoskeletal molecule in the process of tubulointerstitial fibrosis have not been precisely documented. MATERIALS AND METHODS: We investigated the infiltration of macrophages and the expression of alpha-smooth muscle actin in interstitial fibrosis caused by a unilateral ureteral obstruction (UUO) experimental model. RESULTS: The result showed that the macrophages were immobilized at the interstitium and alpha-smooth muscle actin was up-regulated in myofibroblasts of both cortex and medulla at day 3 when interstitial volume start to increase significantly. The highest expression of alpha-smooth muscle actin was detected at day 5 and the most intense infiltration of macrophages was noted at day 14 while the interstitial volume in renal cortex and medulla continued to increase until day 28. Furthermore, we investigated the effect of mizoribine, an immunosuppressive agent, on the interstitial fibrosis induced by UUO, demonstrating that mizoribine, but not prednisolone, significantly improves the tubulointerstitial fibrosis by suppressing the macrophage infiltration and the expression of alpha-smooth muscle actin. CONCLUSIONS: We discuss the pathological roles of macrophages and alpha-smooth muscle actin in tubulointerstitial fibrosis induced by UUO treatment. We also emphasize the pharmacological basis and clinical relevance of mizoribine in the treatment of interstitial fibrosis caused by obstructive nephropathy.  相似文献

It is known that lymphocyte function is impaired after cardiopulmonary bypass (CPB). In this study, the lymphocyte stimulation test (LST) with PHA was used before and after CPB in 28 adult patients, and compared with the surgical parameters and serum cytokine (IL-6, IL-8) levels. LST was impaired after CPB in all patients. Although this value usually recovered by the third postoperative day (POD); (normal group, n = 16), some patients showed prolonged duration of the impaired LST (delayed group, n = 12). Therefore, the parameters of surgery, white blood cell (WBC) count, lymphocytes and subsets, and serum cytokine levels were compared between the normal and the delayed groups. There was no significant difference in the number of WBCs or lymphocytes between these two groups. OKT4-positive cells were reduced on the first POD in both groups, and in the normal group, the number of OKT4-positive cells recovered more quickly than in the delayed group. Serum IL-6 and IL-8 levels in the delayed group were elevated after CPB, and were significantly higher in the delayed group than in the normal group. In conclusion, patients who showed prolonged impairment of lymphocyte function may be partly due to prolonged CPB.  相似文献

Hybrid plasma spraying combined with yttrium-aluminum-garnet laser irradiation was studied to obtain optimum zirconia coatings for thermal barrier use. Zirconia coatings of approximately 150 μm thickness were formed on NiCrAlY bond coated steel substrates both by means of conventional plasma spraying and hybrid plasma spraying under a variety of conditions. Post-laser irradiation was also conducted on the plasma as-sprayed coating. The microstructure of each coating was studied and, for some representative coatings, thermal barrier properties were evaluated by hot erosion and hot oxidation tests. With hybrid spraying, performed under optimum conditions, it was found that a microstructure with appropriate partial densification and without connected porosity was formed and that cracks, which are generally produced in the post-laser irradiation treatment, were completely inhibited. In addition, hybrid spraying formed a smooth coating surface. These microstructural changes resulted in improved coating properties with regard to hardness, high temperature erosion resistance, and oxidation resistance. Cyclic plastic deformation of stainless steel SUS 304 is experimentally investigated at low temperature of liquid nitrogen (77 K) under various cyclic loading conditions Thin walled tubular specimens are subjected to cyclic loading under constant strain ranges. At low temperature, the material shows remarkable hardening by cyclic loading comparing with cyclic loading at room temperature. The hysteresis curves of stress-strain relations by cyclic loading are saturated by increasing the cycle numbers. The saturation tendency depends on loading direction. The saturated stress values are related with cumulative plastic strain of cyclic loading. The prestraining is given at 77 K by axial and torsional loadings, and subsequent cyclic loading under constant strain range is conducted at 77 K. The cyclic stress-strain curves are saturated by increasing cyclic numbers. At small cyclic numbers, cyclic plastic deformation depends on the prestrain direction. The directional effect of pre-strain on cyclic loading becomes small with increasing number of cycles.  相似文献
