Unscheduled DNA synthesis in lymphocytes of rheumatoid arthritis patients and spleen cells of rats with experimentally induced arthritis

DNA repair is an important factor in the abolition or manifestation of mutations. Since intrinsic somatic mutations may be related to the occurrence of autoimmunity, DNA repair was investigated by measuring unscheduled DNA synthesis in lymphocytes of rheumatoid arthritis patients by autoradiography. Likewise, unscheduled DNA synthesis was investigated in mycoplasma- and Freund's adjuvant-induced arthritis in rats. Both in human rheumatoid arthritis and experimentally-induced arthritis the capacity for repairing lesions in DNA induced by gamma-irradiation of cells was reduced, while an increased rate of thymidine-incorporation into DNA was found after UV-irradiation. The results are discussed in relation to endonucleases of viral or mycoplasmal origin.     

A comparison of the validity of three scales for the assessment of dietary restraint.

The construct validity of Herman and Polivy's Restraint Scale (RS), the restraint factor of the Three Factor Eating Questionnaire ({tfeq-r}), and the restraint scale of the Dutch Eating Behavior Questionnaire ({debq-r}) were investigated by relating these scales to self-reported mean caloric intake per day and to other measures associated with disordered eating and figure consciousness. A factor analysis showed that the three restraint scales measure different components of the restraint construct. A high score on the RS was closely related to consequences of mostly unsuccessful dieting, such as disinhibited eating and weight fluctuations, but not to successful overall caloric restriction in everyday life. High scores on the {tfeq-r} and the {debq-r} represented the more successful dieting behavior component of restraint. The three scales have in common a motivational component of restrained eating, including concerns about shape and weight, and desire for thinness. Hammerhead ribozymes: importance of stem-loop II for activity

The activity of several hammerhead ribozyme constructs with constant lengths of stems I and III of 5 nt each but with variously shortened stems II is reported. Stems with 2 bp rather than the conventional 4 bp show essentially unaltered catalytic activity, independent of the composition of the tetraloop. Further reduction in size to 1 bp or 0 bp decreases activity drastically. Inversion of the G10.1.C11.1 bp next to the invariant core leads to a loss in activity, even when the stem consists of 4 bp. Thus, the minimal structural requirement for stem-loop II is a 2-bp stem with a conserved G.C bp. The reduction in catalytic activity is predominantly a result of a decrease of catalytic constant kcat, whereas Km is only slightly affected. Thus, the structural requirement for optimal activity in these constructs where the chemical-cleavage step is rate limiting is determined by the stabilization of the transition state.     

The specific psychopathology of bulimia nervosa: A comparison with restrained and unrestrained (normal) eaters.

The hypothesis that patients with bulimia nervosa and restrained eaters exist on a simple continuum of psychopathology was tested in 60 Ss (20 bulimics, 20 restrained eaters, 20 unrestrained eaters). Regarding measures related to dieting behavior and physical appearance, the restrained eaters differed significantly from unrestrained eaters and were similar to bulimic patients except for level of psychopathology. The restrained eaters could not be distinguished from the unrestrained eaters with regard to measures representing interoceptive perception, self-esteem, and fears about interpersonal relationships. On these traits, bulimic patients could be clearly distinguished from the normal Ss. The results support a 2-component model of the psychopathology of bulimia nervosa. Oligonucleotide duplexes containing 2''-amino-2''-deoxycytidines: thermal stability and chemical reactivity

Oral iodized oil is the major alternative to iodized salt for correcting endemic iodine deficiency. This study responds to a need for better guidelines in its use. Schoolchildren, aged 6-11 yr, from a severely iodine-deficient area of Algeria received iodized poppy seed oil (Lipiodol) in a single oral dose containing 120, 240, 480, or 960 mg iodine (groups A-D) or in an im injection of 480 mg iodine (group E). Thyroid volume by ultrasonography had not changed 395 days after treatment in groups A, B, and C, had decreased in groups D and E. Urinary iodine concentration rose rapidly from an initial median of 0.21 mumol/L, but fell below 0.79 mumol/L (the currently accepted level for indicating iodine deficiency) by 150 days for groups A and B, and by 395 days for groups C and D. Median serum TSH and T4 levels were normal before and after treatment, whereas high initial serum thyroglobulin values decreased in all groups after iodized oil treatment. For correcting iodine deficiency in children, we recommend single oral doses of Lipiodol containing 240 mg iodine for 6-month coverage or 480 mg for 12 months. These doses may not completely sustain iodine sufficiency, but will prevent the worst of the iodine deficiency disorders. Additionally, we conclude that the urinary iodine concentration is the most useful epidemiological indicator for assessing current iodine status, and thyroid volume and serum thyroglobulin levels are the best markers for assessing chronic effects.
