Inbred mouse strains vary in sensitivity to a number of behavioral and physiological effects produced by nicotine. Differences in sensitivity to nicotine are correlated with variance in the number of brain nicotinic receptors as measured in regionally dissected brain tissue. The studies reported here used quantitative autoradiography and in-situ hybridization methods to measure regional levels of alpha-bungarotoxin (alpha BTX) binding and alpha 7 mRNA levels. Two inbred mouse strains, ST/b and DBA/2, were compared because these strains differ maximally in sensitivity to nicotine-induced seizures and in alpha BTX binding measured in regional brain homogenates. The binding of alpha BTX was significantly greater in the St/b strain in 42 of 127 brain regions that were analyzed, and a trend towards increased binding was seen in many additional brain regions. The most consistent strain differences were found in hippocampal, thalamic and pontine nuclei. Strain differences in alpha 7 mRNA levels were also detected, but these were not as widespread as were the alpha BTX binding differences. The alpha 7 mRNA levels were significantly correlated with alpha BTX binding in both mouse strains which suggests that the strain differences in binding are related, in part, to the levels of alpha 7 mRNA.  相似文献

The regulation of conventional protein kinase Cs by Ca2+ was examined by determining how this cation affects the enzyme's 1) membrane binding and catalytic function and 2) conformation. In the first part, we show that significantly lower concentrations of Ca2+ are required to effect half-maximal membrane binding than to half-maximally activate the enzyme. The disparity between binding and activation kinetics is most striking for protein kinase C betaII, where the concentration of Ca2+ promoting half-maximal membrane binding is approximately 40-fold higher than the apparent Km for Ca2+ for activation. In addition, the Ca2+ requirement for activation of protein kinase C betaII is an order of magnitude greater than that for the alternatively spliced protein kinase C betaI; these isozymes differ only in 50 amino acids at the carboxyl terminus, revealing that residues in the carboxyl terminus influence the enzyme's Ca2+ regulation. In the second part, we use proteases as conformational probes to show that Ca2+dependent membrane binding and Ca2+-dependent activation involve two distinct sets of structural changes in protein kinase C betaII. Three separate domains spanning the entire protein participate in these conformational changes, suggesting significant interdomain interactions. A highly localized hinge motion between the regulatory and catalytic halves of the protein accompanies membrane binding; release of the carboxyl terminus accompanies the low affinity membrane binding mediated by concentrations of Ca2+ too low to promote catalysis; and exposure of the amino-terminal pseudosubstrate and masking of the carboxyl terminus accompany catalysis. In summary, these data reveal that structural determinants unique to each isozyme of protein kinase C dictate the enzyme's Ca2+-dependent affinity for acidic membranes and show that, surprisingly, some of these determinants are in the carboxyl terminus of the enzyme, distal from the Ca2+-binding site in the amino-terminal regulatory domain.  相似文献

The relationship between cow evaluations from a 305-d lactation yield animal model [i.e., lactation model (LM)] and a random regression model (RRM) were studied using the first-lactation milk yield of 2,477,807 Holstein heifers. In the LM analysis, 2 values of heritability were used, 0.35 (LM1-H) or 0.57 (LM2-H), the latter being equal to that used in the random regression model for the analysis of the Holstein test-day records (RRM-H). The relative weights on parent average (PA) and yield deviations (YD) were computed and studied to understand factors contributing to reranking of cows’ predicted transmitting abilities (PTA) from the various models. The degree of relatedness and inbreeding were calculated for the top 2,000 cows from the various models. Analyses of Jersey milk yield in the first 3 parities was implemented using 305-d lactation yield multivariate animal (MLM-J) and random regression models (MRRM-J). The ability of both models using only first-parity yield records to predict evaluations in second and third parities when records for these later parities were excluded was studied in a sample of cows. The correlations of cow PTA between LM1-H or LM2-H and RRM-H were 0.91 and 0.92, respectively, in the Holstein data. The data sets used were identical in this case for all models in terms of number of cows and yield records. The correlations were slightly lower at 0.89, 0.87, and 0.88 for parities 1, 2, and 3 in the Jersey analyses, where the data sets were not identical. The relative weights on PA and YD were 0.28 (0.11) and 0.72 (0.89), respectively, from the LM2-H (RRM-H). The RRM-H placed more emphasis on YD and therefore on Mendelian sampling deviations. Thus, the top 2,000 cows from the RRM-H were less related and inbred. The average additive genetic relationship was 22% greater in the LM2-H and average inbreeding coefficients were 0.68 and 0.43% for the LM2-H and RRM-H, respectively. When records were initially available in the first parity, the MRRM-J predicted PTA in parities 2 and 3 with about 2 to 7% greater accuracy compared with the MLM-J.  相似文献

Wether lambs (n=20) were used to compare the effect of dietary magnesium oxide (MgO), unweathered Magnesium Mica® (UMM) and weathered Magnesium Mica® (WMM) on muscle quality. Lambs were fed a corn-based, control diet (Ctrl), or the Ctrl supplemented with either MgO, UMM, or WMM for 95 days before harvest. Following a 24-h chill, carcasses were fabricated, and L*, a* and b* values were determined on the triceps brachii (TB), longissimus thoracis (LT), semimembranosus (SM), and semitendinosus (ST) muscles. Supplemental magnesium had no (P > 0.10) effect on live and carcass weights, fat thickness, loin eye area, and USDA yield grade. Carcasses from lambs supplemented with MgO had higher (P < 0.10) flank streaking scores and USDA quality grades than those from lambs fed diets containing UMM. Although magnesium-supplementation had no (P > 0.10) effect on marbling scores, the LT from Ctrl-fed lambs had more (P < 0.05) intramuscular lipid than lambs fed diets containing UMM or WMM. Lambs supplemented with UMM had greater (P < 0.05) LT shear force values than lambs fed MgO or WMM. Magnesium-supplementation had no (P > 0.10) effect on muscle color; however, supplementing finishing diets with UMM may result in less palatable lamb.  相似文献
