The NRC's Research Program on Core-Debris/Cavity Interactions comprises two principal elements: (1) an analytical effort focused primarily on development of computer codes needed to predict the potential consequences of risk-significant severe-accident scenarios; and (2) an experimental component to provide insights into the relevant phenomenological processes and to develop the experimental data base necessary for validation of the codes. The analytical activities at Sandia National Laboratories (SNL) focus primarily on refinement and validation of the CORCON and VANESA codes. Two major experimental activities are also based at SNL: (1) the large-scale SURC tests address the thermal-hydraulic phenomena in the cavity as well as aerosol release associated with prototypical core-melt materials in various types of concrete crucibles, while (2) the WITCH and GHOST experiments are concerned with aerosol generation and radionuclide release phenomena. A program of small-scale special-effects tests at Brookhaven National Laboratory (BNL) is coupled to a concomitant model-development and code-validation activity. In addition, measurements are being made at Battelle Columbus Laboratory (BCL) to augment the thermochemical data base needed in the VANESA code to permit refined radiological source-term predictions. In June of 2000, Maine DEP (in company with the other five New England states) found itself under EPA pressure to develop and document a quality management system by the end of the calendar year. In the frenzy that followed, the Department decided to use a private sector model for system development that called for a prospectively-focused QM plan that would be implemented through formal auditing. That is, instead of trying in advance to identify all the deficits in departmental quality management, and then assigning individuals and task groups to create structures to fill the gaps, Maine's QMP specifies the future desired system in broad terms. ME-DEP then uses its own cadre of trained auditors to assess current practice against the "condition expected" in the QMP, knowing that in many cases basic quality management practices will need to be developed. This approach assists program managers, particularly (but not exclusively) in areas sensitive to environmental data standards, in creating systems and practices that are rooted in reality, and that are perceived to add value to the Department's core work instead of just additional paperwork. Audit reports create a continuous feedback loop assuring that written procedures document actual operations. Finally, the results of auditing identify areas in which the QM system (and its plan) should be elaborated or refined, which leads to an iterative process by which quality approaches are infused in all areas of DEP operations. In the twelve months following EPA-Region I's initial approval of Maine's QMP, a total of seven audits were completed at various program levels, including two focused on critical QMP elements: Documents and Records; and Computer Hardware/Software. This paper will briefly describe the background and implementation of this approach; identify some of the factors which led to success; and describe, using selected examples, some of the early outcomes of the program.  相似文献

Abstract: Inhibition of Clostridium perfringens spore germination and outgrowth in reduced sodium roast beef by a blend of buffered lemon juice concentrate and vinegar (MoStatin LV1) during abusive exponential cooling was evaluated. Roast beef containing salt (NaCl; 1%, 1.5%, or 2%, w/w), blend of sodium pyro‐ and poly‐phosphates (0.3%), and MoStatin LV1 (0%, 2%, or 2.5%) was inoculated with a 3‐strain C. perfringens spore cocktail to achieve final spore population of 2.5 to 3.0 log CFU/g. The inoculated products were heat treated and cooled exponentially from 54.4 to 4.4 °C within 6.5, 9, 12, 15, 18, or 21 h. Cooling of roast beef (2.0% NaCl) within 6.5 and 9 h resulted in <1.0 log CFU/g increase in C. perfringens spore germination and outgrowth, whereas reducing the salt concentration to 1.5% and 1.0% resulted in >1.0 log CFU/g increase for cooling times longer than 9 h (1.1 and 2.2 log CFU/g, respectively). Incorporation of MoStatin LV1 into the roast beef formulation minimized the C. perfringens spore germination and outgrowth to <1.0 log CFU/g, regardless of the salt concentration and the cooling time. Practical Application: Cooked, ready‐to‐eat meat products should be cooled rapidly to reduce the risk of Clostridium perfringens spore germination and outgrowth. Meat processors are reducing the sodium chloride content of the processed meats as a consequence of the dietary recommendations. Sodium chloride reduces the risk of C. perfringens spore germination and outgrowth in meat products. Antimicrobials that contribute minimally to the sodium content of the product should be incorporated into processed meats to assure food safety. Buffered lemon juice and vinegar can be incorporated into meat product formulations to reduce the risk of C. perfringens spore germination and outgrowth during abusive cooling.  相似文献

Intramuscular collagen (IMC) was isolated from the Longissimus dorsi of six Simmental bulls, 17 months of age, to evaluate the effect of heating on the proportion of types I and III collagen. Cyanogen bromide (CNBr) peptides were prepared from unheated IMC and the soluble and insoluble fractions of IMC heated to 70°C for 70 min or 90°C for 140 min. Percentage of type III collagen was determined by densitometric scans of the CNBr peptides, αl(I)CB8 and αl(III)CB8, as resolved by SDS-PAGE. Percentage of collagen solubilized was greater (P < 0·05) at 90°C than at 70°C. The 70°C and 90°C insoluble IMCs were similar (P > 0·05) for percentage of type III, but both had a greater (P < 0·05) percentage of type III than unheated IMC, indicating that type I is more heat labile than type III. Heat-soluble IMC contained both α and β components and the CNBr peptides of 70°C soluble IMC were mostly type I. These results indicated that heating intramuscular collagen from bulls mainly solubilized type I collagen. Improved tenderness associated with increased heat solubility of collagen may be more closely related to heat-induced solubilization of type I than of type III collagen.  相似文献

OBJECTIVE AND IMPORTANCE: The endovascular treatment of carotid-cavernous dural fistulae is becoming the prominent treatment modality for these lesions. The intractability of these lesions and their tendency to recur, especially after previous endovascular treatment sessions, exhausts the available routes and tends to present a difficulty in accessing the cavernous sinus. To avoid the risks associated with a direct surgical approach, an alternative, less invasive route to the cavernous sinus using a pretemporal extradural approach is combined with a direct endovascular approach. CLINICAL PRESENTATION: A 38-year-old woman presented with a history of right visual and ocular symptoms related to a Type D cavernous carotid dural fistula, which was fed by internal carotid and external carotid branches. The fistula was initially treated with embolization of the external carotid arterial supply. After a transient improvement, the patient's visual acuity worsened. A follow-up angiogram showed the major supply from the intracavernous internal carotid branches and draining through the inferior ophthalmic vein. The transvenous route was not accessible. An attempt to cannulate the intracavernous branches was not successful. The combined pretemporal and endovascular approach was then used. INTERVENTION: The pretemporal extradural region of the superior orbital fissure was exposed. Using microsurgical techniques and Doppler flow guidance, the anterior cavernous sinus was cannulated through the orbital venous drainage channels. Using intraoperative angiography, thrombogenic coils were deployed at the level of the fistula. Intraoperative angiography confirmed complete obliteration of the fistula. CONCLUSION: The combined pretemporal (extradural) and endovascular approach to the cavernous sinus is a less invasive alternative for the treatment of intractable carotid-cavernous dural fistulae.  相似文献

The effect of nitrite and erythorbate on Clostridium perfringens spore germination and outgrowth in ham during abusive cooling (15 h) was evaluated. Ham was formulated with ground pork, NaNO2 (0, 50, 100, 150 or 200 ppm) and sodium erythorbate (0 or 547 ppm). Ten grams of meat (stored at 5 °C for 3 or 24 h after preparation) were transferred to a vacuum bag and inoculated with a three-strain C. perfringens spore cocktail to obtain an inoculum of ca. 2.5 log spores/g. The bags were vacuum-sealed, and the meat was heat treated (75 °C, 20 min) and cooled within 15 h from 54.4 to 7.2 °C. Residual nitrite was determined before and after heat treatment using ion chromatography with colorimetric detection. Cooling of ham (control) stored for 3 and 24 h, resulted in C. perfringens population increases of 1.46 and 4.20 log CFU/g, respectively. For samples that contained low NaNO2 concentrations and were stored for 3 h, C. perfringens populations of 5.22 and 2.83 log CFU/g were observed with or without sodium erythorbate, respectively. Residual nitrite was stable (p > 0.05) for both storage times. Meat processing ingredients (sodium nitrite and sodium erythorbate) and their concentrations, and storage time subsequent to preparation of meat (oxygen content) affect C. perfringens spore germination and outgrowth during abusive cooling of ham.  相似文献

Spores of foodborne pathogens can survive traditional thermal processing schedules used in the manufacturing of processed meat products. Heat-activated spores can germinate and grow to hazardous levels when these products are improperly chilled. Germination and outgrowth of Clostridium perfringens spores in roast beef during chilling was studied following simulated cooling schedules normally used in the processed-meat industry. Inhibitory effects of organic acid salts on germination and outgrowth of C. perfringens spores during chilling and the survival of vegetative cells and spores under abusive refrigerated storage was also evaluated. Beef top rounds were formulated to contain a marinade (finished product concentrations: 1% salt, 0.2% potassium tetrapyrophosphate, and 0.2% starch) and then ground and mixed with antimicrobials (sodium lactate and sodium lactate plus 2.5% sodium diacetate and buffered sodium citrate and buffered sodium citrate plus 1.3% sodium diacetate). The ground product was inoculated with a three-strain cocktail of C. perfringens spores (NCTC 8238, NCTC 8239, and ATCC 10388), mixed, vacuum packaged, heat shocked for 20 min at 75 degrees C, and chilled exponentially from 54.5 to 7.2 degrees C in 9, 12, 15, 18, or 21 h. C. perfringens populations (total and spore) were enumerated after heat shock, during chilling, and during storage for up to 60 days at 10 degrees C using tryptose-sulfite-cycloserine agar. C. perfringens spores were able to germinate and grow in roast beef (control, without any antimicrobials) from an initial population of ca. 3.1 log CFU/g by 2.00, 3.44, 4.04, 4.86, and 5.72 log CFU/g after 9, 12, 15, 18, and 21 h of exponential chilling. A predictive model was developed to describe sigmoidal C. perfringens growth curves during cooling of roast beef from 54.5 to 7.2 degrees C within 9, 12, 15, 18, and 21 h. Addition of antimicrobials prevented germination and outgrowth of C. perfringens regardless of the chill times. C. perfringens spores could be recovered from samples containing organic acid salts that were stored up to 60 days at 10 degrees C. Extension of chilling time to > or =9 h resulted in >1 log CFU/g growth of C. perfringens under anaerobic conditions in roast beef. Organic acid salts inhibited outgrowth of C. perfringens spores during chilling of roast beef when extended chill rates were followed. Although C. perfringens spore germination is inhibited by the antimicrobials, this inhibition may represent a hazard when such products are incorporated into new products, such as soups and chili, that do not contain these antimicrobials, thus allowing spore germination and outgrowth under conditions of temperature abuse.  相似文献

Recent assessments of risk due to severe accidents in light water reactors indicate that molten core-concrete interactions (MCCI) may dominate containment loads and performance, as well as the release of non-volatile fission product aerosols to the containment building. This program has investigated several important aspects of MCCIs in an effort to support the USNRC integral melt-concrete programs as well as the CORCON and VANESA computer code development and verification program. Among these are interlayer heat and mass transfer and liquid-liquid boiling processes.The issues of interlayer heat and mass transfer address phenomena that occur primarily interior to the molten core debris itself. Models have been developed to predict the onset of entrainment between liquid layers, the rate of entrainment, and the rate of heat transfer, both with and without entrainment. Application to the reactor case indicates that entrainment and mixing may or may not occur, depending upon the prevailing thermal hydraulic conditions in the melt.The issues of liquid-liquid film boiling address phenomena that occur primarily at the melt-coolant interface. Results from melt-water film boiling studies have suggested that the coolant heat flux increases above the flat plate limit with non-condensable gas injection from below. Steam explosions occur frequently; the frequency and magnitude vary with gas injection rate.  相似文献

People are inaccurate judges of how their abilities compare to others'. J. Kruger and D. Dunning (1999, 2002) argued that unskilled performers in particular lack metacognitive insight about their relative performance and disproportionately account for better-than-average effects. The unskilled overestimate their actual percentile of performance, whereas skilled performers more accurately predict theirs. However, not all tasks show this bias. In a series of 12 tasks across 3 studies, the authors show that on moderately difficult tasks, best and worst performers differ very little in accuracy, and on more difficult tasks, best performers are less accurate than worst performers in their judgments. This pattern suggests that judges at all skill levels are subject to similar degrees of error. The authors propose that a noise-plus-bias model of judgment is sufficient to explain the relation between skill level and accuracy of judgments of relative standing.
