Aspiration biopsy of tumors and transplanted organs is a popular atraumatic method for diagnosis and also for the management of patients. Apart from the cytological analyses, it is clear that large aggregates of cells are obtained and these may be examined by electron microscopy. A study was carried out with dogs to assess the yield of intact glomeruli and tubules, resulting from a standard fine-needle aspiration procedure. Employing a 20-gauge needle, yields of approximately ten intact glomeruli were obtained with a tissue processing protocol that utilized filtration through a nylon mesh and backwashing, to resuspend the trapped aggregates. Processing for electron microscopy was carried out in microcentrifuge tubes. The ultrastructural preservation was excellent and the extension of these studies to investigations of not only renal transplantation, but also for diagnosis in human renal diseases of a broad spectrum, is suggested.  相似文献

The goal of this study was to develop a mathematical model which incorporates heat flow, phase transformation kinetics, and property-structure-composition relationships to predict the mechanical properties of steel rod being control cooled under industrial conditions. Thus, the principles of microstructural engineering have been brought to bear on this interdisciplinary problem by combining computer modeling with laboratory measurements of heat flow, austenite decomposition kinetics, microstructure and mechanical properties, and industrial trials to determine heat transfer and obtain rod samples under known conditions. Owing to the length and diversity of the study, it is reported in three parts,[8191]the first of which is concerned with the heat flow measurements. A relatively simple and reliable technique, involving a preheated steel rod instrumented with a thermocouple secured at its centerline, has been devised to determine the cooling rate in different regions of the moving bed of rod loops on an operating Stelmor line. The measured thermal response of the rod has been analyzed by two transient conduction models (lumped and distributed parameter, respectively) to yield overall heat-transfer coefficients for radiation and convection. The adequacy of the technique has been checked by cooling instrumented rods under well-defined, air crossflow conditions in the laboratory and comparing measured heat-transfer coefficients to values predicted from well-established equations. The industrial thermal measurements have permitted the characterization of a coefficient to account for radiative interaction among adjacent rod loops near the edge and at the center of the bed. Total phosphorus (TP) inputs to Lake Simcoe have led to hypolimnetic dissolved oxygen (DO) depletion and loss of cold water fish habitat. Since 1990, efforts have been made to reduce the total TP input to the lake below a defined target of 75 t/year, which was predicted to lead to reductions in spring TP concentration and improvements in end-of-summer hypolimnetic DO concentrations. The total TP load to the lake during the most recent period of record (1998/99-2003/04) ranged from 53 to 76 t/yr and averaged 67 t/yr, compared to an average of 114 t/yr estimated between 1990/91 and 1997/98 (range 85-157 t/yr). Reductions in TP loads from the catchment via tributary discharge (∼26 t) accounted for the majority of the decrease in total load between the two time periods. Total P concentrations decreased significantly in four out of six long-term monitored tributaries; however, concentrations in all six tributaries remain above the level recommended to avoid nuisance plant growth (30 μg/L). Although TP loads to the lake are currently below the target 75 t/yr, excessive growths of filamentous algae and macrophytes continue to be a problem in the nearshore zone. End-of-summer minimum hypolimnetic DO concentrations (average 4.3 mg/L, 1998/99-2003/04) remain substantially below the level (7 mg/L) that is considered protective of lake trout. Efforts to reduce TP loads to the lake therefore need to continue.  相似文献

Luteinizing hormone releasing hormone (LHRH) stimulates the development of cellular FSH immunoreactivity in the perinatal hamster adenohypophysis. Because neuropeptide Y (NPY) can act directly on rat adenohypophysial cells to stimulate FSH and LH release and potentiate the stimulatory effect of LHRH on FSH and LH release, we investigated the effects of NPY alone and in combination with a low, ineffective dose of LHRH on inducing cellular FSH immunoreactivity in the neonatal hamster adenohypophysis. Neonatal female pituitary glands were grafted beneath the right renal capsules of hypophysectomized-ovariectomized adult hamster hosts with a catheter implanted in the external jugular vein. After treatment, hosts were decapitated and graft tissue was stained for FSH and LH immunoreactivity. The mean percentage of adenohypophysial cells that stained for FSH was low (2.8%) in grafts in hosts infused continuously with heparinized saline vehicle for 7 days. In other hosts, peptides were pulsed through the catheter every 12 h for 7 days. The mean percentage of FSH cells also was low after pulsing 6 ng LHRH or 2 micrograms NPY but increased substantially when the two peptides were pulsed simultaneously. No differences in the mean percentage of LH cells existed between any of the groups. The results demonstrate that NPY and LHRH can synergize to induce cellular FSH immunoreactivity in the neonatal female hamster.  相似文献

Existing NASA supported scientific databases are usually developed and managed by a team of database administrators whose main concern is the efficiency of the databases in terms of normalization and data search constructs. The populating of the database is usually done in a manual fashion by row and column as the data become available, and the data dictionary is usually defined by the same team (at times with little input from the end science user). This process is tedious, error prone and self-limiting in terms of what can be described in a relational Data Base Management System (DBMS). The next generation Earth remote sensing platforms [i.e., Earth Observing System (EOS)] will be capable of generating data at a rate of over 300 Megabits per second from a suite of instruments designed for different applications. What is needed is an innovative approach that creates object-oriented data-bases that segment, characterize, and catalog, and are manageable in a domain-specific context, and whose contents are available interactively and in near-real-time to the user community. This paper describes work in progress that utilizes an artificial neural net approach to characterize satellite imagery of undefined objects into high-level data objects. The characterized data is then dynamically allocated to an object-oriented database where it can be reviewed and accessed by a user. The definition, development, and evolution of the overall data system model are steps in the creation of an application-driven knowledge-based scientific information system.  相似文献

Is there a relationship between personality and criminal behavior? We addressed this question in a representative birth cohort of 862 male and female 18-yr-olds. Personality was assessed with the Multidimensional Personality Questionnaire (MPQ). The MPQ measures 10 relatively independent personality traits and was not designed to identify offenders. Delinquency was assessed via 3 data sources: self-reports, informant reports, and official records. Variable-centered analyses revealed that MPQ scales indexing negative emotionality and behavioral constraint were consistent predictors of delinquency across the 3 data sources. Person-centered analyses revealed that youths abstaining from delinquency were uniquely characterized by low interpersonal potency. Youths involved in extensive delinquency were uniquely characterized by feelings of alienation, lack of social closeness and risk taking. Advances in understanding criminal behavior can be made through research that places the personality-delinquency link in a developmental context. OBJECTIVE: The effect of cigarette smoking on gallbladder (GB) emptying and refilling after a fatty meal was examined in 10 healthy volunteers (four women and six men, mean age 27.6 yr). METHODS: On three different days, the subjects underwent in randomized order: a control test without smoking (C), or they smoked two cigarettes during the early (0-20 min; S0-20), or late (20-40 min; S20-40) phase of the meal-induced GB emptying. GB volumes were measured ultrasonographically before the meal and at 10, 20, 30, 40, 60, 90, 120, 150, and 180 min postprandially. Two-way ANOVA was applied for statistical assessment of the results. RESULTS: The fasted GB volumes amounted to 15.7 +/- 1.8 cm3 (C), 15.0 +/- 1.7 cm3 (S0-20), and 18.4 +/- 2.3 cm3 (S20-40), F2;18 = 1.524, NS. Maximum GB emptying was observed until 60 min after the meal, with a nadir of the GB volume amounting to 7.3 +/- 1.3 cm3 (C), 6.6 +/- 1.2 cm3 (S0-20), and 7.1 +/- 1.1 cm3 (S20-40). No significant difference was found between the stimuli tested when absolute GB volumes were considered: F2;180 = 2.725, NS. Analysis of the GB emptying-refilling curves normalized for the fasted GB volume revealed that a significant inhibitory effect was produced by smoking two cigarettes during the late phase of GB emptying on the subsequent GB refilling: F2;162 = 11.066, p < 0.001 for the whole curve, and F2;72 = 7.126, p < 0.005 for the refilling phase. A significant contrast was found next between S20-40 and the control day (p < 0.001 whole curve; p < 0.005 refilling phase only), as well as between S20-40 and S0-20 (p < 0.001 whole curve; p < 0.025 refilling phase only). CONCLUSION: We conclude that smoking two cigarettes does not disturb the fatty meal-induced GB contraction in healthy humans. Subsequent GB refilling is delayed if smoking takes place during the late phase of the postprandial GB contraction.  相似文献

This article presents a two-dimensional transient model for gas-solids flow and heat transfer through pipes using the coupled Computational Fluid Dynamics and Discrete Element Method approach. Numerical simulations have been conducted to examine the modification of fluid thermal structure due to the presence of particles in a pneumatic transport pipeline. Modeled results have demonstrated the key role of transversal motion of rebounding particles in the pipe cross section in altering fluid temperature. Further implementation of this modeling technique in air-drying processes is discussed and possible experimental methods for the measurement of in situ particle and fluid motion and temperature profile are cited.  相似文献
