Previous studies have shown that Tetrahymena citrate synthase and the Tetrahymena 14-nm filament protein are encoded by a single gene and translated from one species of mRNA, and that they are identical in terms of molecular weight, antigenicity, and some enzymatic properties. In this study, using two-dimensional gel electrophoresis, we demonstrated that the citrate synthase comprised pI 7.7 and 8.0 isoforms, while the 14-nm filament protein comprised three isoforms with isoelectric points of 7.7, 8.0, and 8.4. The amino acid sequences of the NH2-terminal portions of all isoforms were identical and the peptide maps with V8 protease were almost the same. In addition, when the citrate synthase activity of each isoform was measured after separation by non-urea isoelectric focusing without denaturing treatment, the pI 7.7 and/or pI 8.0 isoforms exhibited the citrate synthase activity, but the pI 8.4 isoform only found for the 14-nm filament protein did not possess this activity. These results suggest that the polymorphism of these isoforms is caused by some posttranslational modifications, and that it may have resulted in the different compartmentalization and functions of Tetrahymena citrate synthase and the 14-nm filament protein.  相似文献

The autoimmune-prone MRL-lpr substrain of mice develop an autoimmunity-associated behavioral syndrome (AABS) which resembles in many respects the behavior of animals exposed to chronic stress. The present study examined whether these mice show changes in the morphology of neuronal dendrites, as found in animals exposed to chronic stress. A modified Golgi-Cox procedure was used to visualize the dendrites of pyramidal neurons in the parietal cortex and in the CA1 hippocampal field of 5-week and 14-week old MRL-lpr mice and MRL + / + controls. Reduced dendritic branching and length, and an up to 20% loss of dendritic spines were observed in parietal and hippocampal pyramidal neurons of MRL-lpr mice at both ages. In the parietal cortex, there was an age-dependent potentiation in the reduction of basilar, but not apical, dendrite branching and length, as well as in the loss of spines on basilar segments. Loss of spines in the hippocampus followed an age-related course for apical but not basilar dendrites. Moreover, compared to age-matched controls, brain weight was smaller in MRL-lpr mice at 14 but not 5 weeks of age. Considering that dendritic atrophy becomes more extensive when autoimmune disease is florid in MRL-lpr mice, it is proposed that immune/inflammatory factor(s) produce dendritic loss. Reduced dendritic complexity may represent, at least in part, a structural basis for the altered behavioral profile of MRL-lpr mice.  相似文献

Reviews the perspectives of the drive/structural, object relations, and self-psychology models as to premature termination. A case history is presented of a man in his late twenties who ended therapy after 2.5 yrs. Examining the commonalities in the application of the 3 models, a central issue is apparent: the man's conflicts over dependency. Whether these conflicts are construed as resistance to experiencing passive feminine desires, as separation–individuation conflicts, or as fears of retraumatization by an unattuned self-object, issues of dependence are central to the patient's leaving treatment prematurely. Clinicians sometimes worry that the administration of intentional memory tests early in a clinical battery might artificially enhance performance on subsequent incidental memory tests, with possible differential effects for younger vs. older adults. Eighty-one healthy adults (ages 30–80) were blindly assigned to 1 of 2 testing conditions (incidental followed by intentional and vice versa). Contrary to the concerns of some, administration of intentional tests prior to incidental tests did not affect performance on any measures, and there was no interaction with age. Age differences in processing resources seem salient to age-related declines in secondary (or "recent") memory. Community-dwelling adults (N?=?90, ages 30–80) completed 4 memory tests: Wechsler Memory Scale—Revised (WMS-R), Logical Memory (LM), Cowboy Story (CS), WMS-R Visual Reproductions (VR), and Extended Complex Figure Test (ECFT; Fastenau, in press). Two space-capacity measures (WMS-R Digit Span and Visual Memory Span) and 4 processing speed measures (cancellation and mental-tracking tasks) assessed processing resources. A statistical control procedure was used to isolate retrieval efficiency and measure contributions of age and processing resources to retrieval. In humans, the emotional nature of stimuli appears to have a complex influence on long-term declarative memory for those stimuli: Whereas emotion enhances memory for gist, it may suppress memory for detail. On the basis of prior studies, the authors hypothesized that the amygdala helps mediate the above 2 effects. Long-term memory for gist and for visual detail of aversive and neutral scenes was assessed in 20 subjects with unilateral amygdala damage and 1 rare subject with bilateral amygdala damage. Comparisons with 2 control groups (15 brain-damaged and 47 healthy) provided evidence that bilateral, but not unilateral, damage to the amygdala results in poorer memory for gist but superior memory for visual details. The pattern of findings provides preliminary support for the idea that the amygdala may help filter the encoding of relevant information from stimuli that signal threat or danger. Informant reporting is important in the assessment of depression and anxiety among individuals with cognitive impairment. The authors examined the influence of the visibility effect on the ease of rating depression and anxiety symptoms. Fifty-three family members of dementia patients and 65 staff members working with cognitively impaired adults judged the ratability of the Inventory of Depression and Anxiety Symptoms item pool. Results indicated that Appetite Loss, Lassitude, and Insomnia scales were easiest to rate; Suicidality and Traumatic Intrusions scales were most difficult to rate.
