A multi-laboratory, simulator study investigated the wear of polytetrafluorethylene (PTFE) cups run in bovine serum. Each laboratory used its own test protocol with a variety of simulator types. Our wear model incorporated 32 mm dia CoCr heads matched to PTFE cups run with serum protein-concentrations in the range 17–69 mg/ml. The multi-lab data demonstrated that protein-concentration had the most significant effect on wear performance. Both inverted and anatomical cups followed the same trend with first a rapid increase in wear-rates apparent for the initially low-protein levels and then a wear-rate reduction effect becoming apparent beyond 17 mg/ml of proteins. The results showed that as the protein concentration increased from 17 to 69 mg/ml, the magnitude of the wear-rates increased 200% but the protein wear-rate gradient decreased 24–60% with “inverted” and “anatomical” cups, respectively. This effect was more pronounced with ‘anatomical” than “inverted” cups. Thus, the wear-trends with “inverted” cups were generally the more consistent, particularly at the low-protein levels. Increasing the serum volume by two-fold in one study increased the PTFE wear-magnitudes approximately 40% and the protein-wear gradient by 30%. These PTFE wear phenomena were consistent with the concept that low-concentrations of proteins promoted polymer wear but high-protein concentrations resulted in a protein-degradation phenomenon which progressively masked the actual polymer wear. In the selected protein range 17–69 mg/l, the multi-laboratory simulator data consistently overestimated the average clinical wear-rate by at least 50–100% depending on protein range. It would, therefore, appear clinically relevant to study PTFE wear with an inverted-cup model using a large volume of serum but only in low-protein concentrations. The protein-related wear phenomena observed with PTFE cups in this multi-laboratory project may also have relevance for wear-simulation of UHMWPE cups.  相似文献

The Chediak-Higashi Syndrome (CHS) is a rare inherited disorder occurring in man and several animal homologs including the beige mouse; it is characterized by pigmentary dilution, susceptibility to pyogenic infections, and the presence of enlarged lysosomes in many cell types. Beige mice 6 months of age and older were found to have darkened livers, kidneys, and spleens, accompanied by splenomegaly. A fluorescence microscopic survey of tissues from beige mice revealed marked accumulations of a microscopic survey of tissues from beige mice revealed marked accumulations of a yellow autofluorescent pigment inhepatocytes, renal proximal tubule cells, and splenic macrophages. Additionally, large amounts of hemosiderin were present in the spleen. In beige mice, the pigment was noted in animals as young as 1 to 2 weeks of age and gradually increased in amount as the animals aged. A histochemical investigation of the pigment showed that it was ceroid-like in nature and contained in lysosomes. Ultrastructurally, the pigment was composed of lipid-like droplets embedded in a dense matrix and surrounded by a limiting membrane. The accumulation of ceroid-like material in beige mice may be a reflection of the metabolic disturbance which underlies CHS.  相似文献

Total hip arthroplasty is a highly successful procedure where the hip joint is replaced by an artificial ball and socket joint. Bearing wear continues to be a contributing factor to implant failure. Prosthetic femoral heads roughen in vivo which leads to increased wear. Along with the introduction of improved polyethylene which reduces wear by up to 99%, improved femoral head materials have been introduced to improve resistance to abrasion. The abrasion resistance of two of these improved femoral heads was assessed in this study and compared to a cobalt chromium (CoCr) femoral head. The resulting wear performance against a polyethylene acetabular component was assessed. The bulk ceramic (zirconia toughened alumina) femoral head exhibited superior abrasion resistance compared to CoCr (97% reduction in damage) as well as reduced wear after abrasion (97% reduction in wear). The oxide coated zirconium niobium femoral head showed inferior abrasion resistance compared to CoCr (99% increase in damage) as well as increased wear after abrasion (161% increase in wear). Both femoral head surfaces utilize hard ceramic materials, however, the thin ceramic coating on top of a softer metallic substrate of the oxide coated bearing was unable to withstand aggressive abrasion.  相似文献

Objective: Based on social ecological theory, this study examined the joint relations among adolescents' family, peer, and school contexts and depressive symptoms in youth with spina bifida using cumulative, protective, and specific effects models. Method: Sixty families of adolescents with spina bifida and 65 comparison families reported on adolescents' positive experiences within these contexts and on depressive symptoms when youth were 14–15 and 16–17 years old. Results: Adolescents with spina bifida had fewer total positive contexts and less positive experience within peer and school contexts, as compared to typically developing adolescents. Greater total number of positive contexts and higher levels of positive experiences within family and school contexts were associated with fewer depressive symptoms for both groups; peer positive experiences were related to lower depressive symptoms for typically developing adolescents only. Conclusion: Adolescents with spina bifida have fewer positive contexts, which may place them at risk for higher levels of depressive symptoms. The immutans (im) mutant of Arabidopsis shows a variegated phenotype comprising albino and green somatic sectors. We have cloned the IM gene by transposon tagging and show that even stable null alleles give rise to a variegated phenotype. The gene product has amino acid similarity to the mitochondrial alternative oxidase. We show that the IM protein is synthesized as a precursor polypeptide that is imported into chloroplasts and inserted into the thylakoid membrane. The albino sectors of im plants contain reduced levels of carotenoids and increased levels of the carotenoid precursor phytoene. The data presented here are consistent with a role for the IM protein as a cofactor for carotenoid desaturation. The suggested terminal oxidase function of IM appears to be essential to prevent photooxidative damage during early steps of chloroplast formation. We propose a model in which IM function is linked to phytoene desaturation and, possibly, to the respiratory activity of the chloroplast.  相似文献

Objective: As a follow-up to an earlier cross-sectional study (Holmbeck et al., 2003), the current multimethod, multi-informant investigation examined individual growth in psychosocial adjustment across the adolescent transition in 2 samples: young adolescents with spina bifida (SB) and typically developing adolescents (N = 68 in both groups at Time 1). Method: Growth curve modeling procedures were used to describe the developmental course of psychosocial adjustment across 4 waves of data collection from ages 8 to 15. Child gender was included in the models as a moderator of associations between illness status and adjustment trajectories. Results and Conclusions: Findings revealed that preadolescent differences between groups were maintained for several adjustment variables, indicating that adolescents with SB have enduring academic and attention problems and difficulties with social development (e.g., fewer friends and less influence during family interactions). For other outcomes, trajectories of adjustment levels for adolescents with SB converged on levels observed in comparison adolescents, indicating some areas of resilience. Girls with SB were at risk for increasing levels of social difficulties and negative perceptions of their physical appearance. Clinical implications are discussed. Reduced energy expenditure may predispose children to the development of obesity, but there are limited longitudinal studies to support this theory. We studied 75 white, preadolescent children over 4 y by taking annual measures of body composition and resting energy expenditure (by indirect calorimetry) and two annual measures of total energy expenditure and physical-activity-related energy expenditure (by doubly labeled water). Body composition of parents was assessed at the onset of the study with use of underwater weighing. The major outcome variable was the individual rate of change in fat mass (FM) adjusted for fat-free mass (FFM). The influence of sex, energy expenditure components, initial FM, and parental FM on the rate of change in FM was analyzed by hierarchical linear modeling and analysis of variance. The rate of change in absolute FM was 0.89 +/- 1.08 kg/y (range: -0.44 to 5.6 kg/y). The rate of change in FM adjusted for FFM was 0.08 +/- 0.64 kg/y (range: -1.45 to 2.22 kg/y) and was similar among children of two nonobese parents and children with one nonobese or one obese parent, but was significantly higher in children with two obese parents (0.61 +/- 0.87 kg/y). The major determinants of change in FM adjusted for FFM were sex (greater fat gain in girls), initial fatness, and parental fatness. None of the components of energy expenditure were inversely related to change in FM. The main predictors of change in FM relative to FFM during preadolescent growth are sex, initial fatness, and parental fatness, but not reduced energy expenditure.  相似文献

This paper investigates the tribological performance of a carbon fiber reinforced PEEK composite as a bearing surface for total hip replacement. Extensive hip joint simulator tests were conducted to optimize the microstructure of the composite and the counterface material. A softer and more graphitic carbon fiber is preferred to a harder and more abrasive fiber. A ceramic counterface is preferred to a metal counterface. An excellent wear couple was identified to be a 30 wt% pitch-based carbon fiber reinforced PEEK composite acetabular insert articulating against a zirconia ceramic head. When tested on a hip simulator run for 10 million cycles, a reduction in the wear rate of almost two orders of magnitude was achieved with this wear couple in comparison to a conventional UHMWPE/metal or UHMWPE/ceramic couple.  相似文献
