Individual differences in prepulse inhibition of startle as a measure of individual dopamine function.

This study investigated whether individual differences in prepulse inhibition (PPI) of the acoustic startle reflex reflect meaningful trait differences in the function of dopaminergic substrates that regulate it. Baseline PPI of individual rats showed strong test-retest reliability across 3 consecutive test days, and there was a significant negative correlation between individual baseline PPI and both disruption of PPI produced by apomorphine and facilitation of PPI by haloperidol. The test-retest reliability and the inverse association between baseline PPI and drug-induced effects were stronger with 8–10 dB prepulses compared with less intense prepulses. These results demonstrate that individual differences in baseline PPI predict individual differences in sensitivity of PPI to drugs that affect the dopamine system and that PPI produced by more intense prepulses may be more representative of these individual differences. Versatile high-repetition-rate phase-locked chopper system for fast timing experiments in the vacuum ultraviolet and x-ray spectral region

A novel light chopper system for fast timing experiments in the vacuum-ultraviolet (VUV) and x-ray spectral region has been developed. It can be phase-locked and synchronized with a synchrotron radiation storage ring, accommodating repetition rates in the range of ~8 to ~120 kHz by choosing different sets of apertures and subharmonics of the ring frequency (MHz range). Also the opening time of the system can be varied from some nanoseconds to several microseconds to meet the needs of a broad range of applications. Adjusting these parameters, the device can be used either for the generation of single light pulses or pulse packages from a microwave driven, continuous He gas discharge lamp or from storage rings which are otherwise often considered as quasi-continuous light sources. This chopper can be utilized for many different kinds of experiments enabling, for example, unambiguous time-of-flight (TOF) multi-electron coincidence studies of atoms and molecules excited by a single light pulse as well as time-resolved visible laser pump x-ray probe electron spectroscopy of condensed matter in the valence and core level region.     

Feeding effects of growth hormone-releasing factor in rats are photoperiod sensitive.

By examining the influence of photoperiod, this study extended previous findings showing that centrally administered growth hormone-releasing factor (GRF) increases feeding. Rats received icv injections of GRF (0, 0.4, 4.0, and 40.0 pmol) during either the light or dark phase of the photocycle. GRF produced a dose-dependent increase in 90-min food intake during the light photoperiod but a dose-dependent suppression during the dark photoperiod. GRF did not significantly alter 24-hr feeding or locomotor activity, and a biologically inactive analog of GRF had no effect on feeding. Possible mechanisms underlying GRF's photoperiod dependency are discussed, including a suggestion that endogenous GRF may play a role in the regulation of circadian feeding. Coping with conflict situations: Middle-aged and elderly men.

In this study we investigated the coping responses of fairly healthy, middle-aged (40–64 years, n?=?76) and elderly (65–92 years, n?=?106) men to five conflict situations (i.e., decision making, defeat in a competitive circumstance, frustration, authority conflict, and peer disagreement). Coping responses were measured by the Life Situations Inventory, developed to assess three forms of coping: problem-solving, avoidance, and resignation. Scales were based on a 28-item questionnaire and were derived rationally through item analysis. Alpha coefficients ranged from .75 to .82. Elderly subjects used avoidance significantly less often than did middle-aged subjects in handling decision-making and authority-conflict situations. No differences were noted between the age groups in use of problem solving or resignation. Both middle-age and elderly persons favored use of problem solving in managing all conflicts. Results suggest that studies that do not address potential interactions between age and stress situation may be missing an important element in the age–coping relation. Elective lymph node dissection in malignant melanoma--status of color Doppler findings

Since there is still a controversial discussion about the ELND in melanoma patients, the purpose of this prospective study was to optimize the indication concerning ELND by ultrasound examinations. 144 patients with primary melanomas were checked every 3 months after excision. Echomorphologic pattern and intranodal vascularisation in the color-flow Doppler modus provide essential information for differential diagnosis. We found 47 patients (32.6%) with suspicious ultrasound lesions. 50% of these patients had no pathological clinical findings, the histological findings of excised lymph nodes were positive in all cases. It must be emphasized, however, that the group with lymph node metastases included 12 patients with low-risk-melanomas (2 x Tis, 10 x T1 < 1 mm tumor thickness). In comparison with a historical control group (141 patients), where ELND was performed routinely in high-risk-patients ( > T2), the incidence of ELND in our hospital decreased more than 50%; at the same time the percentage of detected lymph node metastases increased (twice).     

Changes in colitis surgery. I: Is there a standard in primary surgical treatment?

In a 15 years period surgery of ulcerative colitis expanded from ileostomy (IS) via Kock's pouch (KP) and ileoanal pouch (IAP) to ileorectal anastomosis (IRA). Interactions between availability of methods and frequency of operations are investigated retrospectively in order to establish an optimal primary procedure. With an overall amount of 80 operations the yearly operative frequency raised in correlation to the introduction of continence reconstructive procedures. As a consequence of this fact history of disease was shortened to less than 8 year and global colitis-associated morbidity markedly decreased. For patients readiness to undergo operation Kock's pouch was only important at the beginning (n = 9/11.3%). Most decisive was IAP (n = 49/61.3%) which could be realized last even in an one-stage-procedure with better early results due to improved patients conditions and simplified technical modifications. IRA (n = 7/8.8%) played only a limited role in the last years for selected patients, whereas IS (n = 15/18.8%) kept reserved for contraindications to reconstructive surgery. For all procedures operative complications decreased from 46.1% (12/26) to 11.0% (6/54) and lethality to 0%. Late complications were related to proctectomy (nerve damage) and construction of IAP (pouchitis in 34.8% and defunctioning of the pouch in 10.4%), whereas IRA was free of specific morbidity so far. Surgery of ulcerative colitis is characterized today by restoration of anal continence. The advantage of the changed surgical concept lies within the ability to perform colectomy at an earlier stage of the disease. Safe construction of IAP is the most important technical progress. Early operation of colitis and late morbidity of pouch justify (preliminary) IRA. Thus, surgical standard in colitis-surgery is defined more individually.

A novel, complement factor H-related regulatory protein expressed on the surface of human B cell lines

Complement regulatory proteins present on the surface of various mammalian cells play an important role in controlling homologous lysis, by interacting with C3 (and usually C4). These proteins have a similar structural motif ("short consensus repeat") (Reid, K.B.M., Bentley, R.D., Campbell, R.D., Chung, L.P., Sim, R.B., Kristensen, T. and Tack, B.F., Immunol. Today 1986. 7:230), and the genes encoding them are members of the family of regulators of complement activation. Here we describe a hitherto unknown member of this family, a molecule expressed by B lymphoblastoid cells. This protein is recognized by polyclonal antibodies to factor H and by MAH4, a monoclonal antibody reacting with the N-terminal portion of factor H. The cell surface protein is built up of two disulfide-linked chains of approximately 68 and 75 kDa. Biosynthetic labeling studies confirmed that it is synthesized by B cells only, but not by the investigated lines of other origin. When tested for its functional activity, this molecule was shown to act as cofactor for factor I-mediated cleavage of fluid-phase C3b to C3bi. The protein appears to be encoded by a 3.5-kb mRNA, hybridizing with a cDNA probe coding for the N-terminal portion of factor H. Due to its cross-reactivity with anti-H antibodies, cofactor activity for factor I and hybridization with factor H cDNA, despite its two-chain composition, it is considered a factor H-like protein.     

Another look at fear of death.

There is increasing recognition of the need for multilevel measures when dealing with the fear of death (FOD) construct. The present study addressed itself to determining degree of FOD where categorization was based on a consistent positioning of the individual on both direct and indirect measures of FOD. Additionally, FOD was examined in connection with other death dimensions and behaviors. 212 adult males with a high degree of FOD (as measured by the Collett-Lester Fear of Death of Self subscale) when compared with 208 adult males low in FOD significantly viewed death in more negative terms, reported an increase in their personal FOD as they became older, and were less religious in both conduct and creed. Further, when a fantasy level measure (metaphors) was combined with a conscious level assessment of FOD, prediction of degree of FOD was enhanced significantly. The study emphasizes the importance of using multilevel facets of FOD and the need for a variety of outcome measures in appraising the construct.
