The molecular events involved in apoptosis induced by ionizing radiation remain unresolved. In this paper we show that the cleavage of fodrin to a 150 kDa fragment is an early proteolytic event in radiation-induced apoptosis in the Burkitts' Lymphoma cell line BL30A and requires 100 microM zVAD-fmk for inhibition. Caspases-1, -3, -6 and -7 were shown to cleave fodrin to the 150 kDa fragment in vitro and all were inhibited by 10 microM zVAD-fmk. We also show that the in vitro cleavage of fodrin by calpain is inhibited by 100 microM zVAD-fmk as was the calpain-mediated hydrolysis of casein. We demonstrate that calpain is activated within 15 min after radiation exposure, concomitant with the cleavage of fodrin to the 150 kDa fragment whereas caspase-3 is activated at 2 h correlating with the cleavage of fodrin to the 120 kDa fragment. These results support a role for calpain in the early phases of the radiation-induced apoptosis pathway, upstream of the caspases.  相似文献

The authors evaluated the reliability and validity of a tool for measuring older adults' decision-making competence (DMC). A sample of 205 younger adults (25–45 years), 208 young-older adults (65–74 years), and 198 old-older adults (75–97 years) made judgments and decisions related to health, finance, and nutrition. Reliable indices of comprehension, dimension weighting, and cognitive reflection were developed. Comparison of the performance of old-older and young-older adults was possible in this study, unlike previous research. As hypothesized, old-older adults performed more poorly than young-older adults; both groups of older adults performed more poorly than younger adults. Hierarchical regression analyses showed that a large amount of variance in decision performance across age groups (including mean trends) could be accounted for by social variables, health measures, basic cognitive skills, attitudinal measures, and numeracy. Structural equation modeling revealed significant pathways from 3 exogenous latent factors (crystallized intelligence, other cognitive abilities, and age) to the endogenous DMC latent factor. Parametric modeling strategies are explored in conjunction with linear discriminant analysis for use in an electroencephalogram (EEG)-based brain-computer interface (BCI). A left/right self-paced typing exercise is analyzed by extending the usual autoregressive (AR) model for EEG feature extraction with an AR with exogenous input (ARX) model for combined filtering and feature extraction. The ensemble averaged Bereitschafts potential (an event related potential preceding the onset of movement) forms the exogenous signal input to the ARX model. Based on trials with six subjects, the ARX case of modeling both the signal and noise was found to be considerably more effective than modeling the noise alone (common in BCI systems) with the AR method yielding a classification accuracy of 52.8+/-4.8% and the ARX method an accuracy of 79.1+/-3.9 % across subjects. The results suggest a role for ARX-based feature extraction in BCIs based on evoked and event-related potentials.  相似文献

The goal of the work was to study female tolerance of g-loads before and after simulated weightlessness, and evaluate the effectiveness of countermeasures and anti-g means proposed for space flight. Centrifuge with a 7.25 m arm was used for 29 runs of 8 female subjects. The subjects were exposed to the transverse loads (+GX) at 8.2 g on a special regimen with the total run time of 9 min to simulate the ballistic descent from orbit in a SOYUZ-TM vehicle. Baseline tolerance of the females to transverse loads up to +8GX was good and satisfactory. Following 120 days of bed rest without the countermeasures or the anti-g suit g-tolerance was significantly lowered as evidenced by visual disorders in 67% of cases and exaggerated, if compared with the pre-hypokinesia levels, heart and respiration rates, i.e. 20% and 23.5%, respectively. The countermeasures and anti-g suit Centaur maintained g-tolerance at essentially the baseline level and precluded visual disorders. Results of the study speak in favor of the countermeasures and anti-g means used by the females in simulated long-duration weightlessness which can be recommended for the final phase of extended space mission.  相似文献

This article reports progress in an ongoing research program examining older adults' decision-making competence (DMC). Using a theoretical framework that emphasizes the person-task fit in assessing DMC, the authors report the results of a study comparing older versus younger adults' decision performance on simple and complex tasks about health, finance, and nutrition. The authors hypothesized and found that increasing age and task complexity were related to greater comprehension errors and inconsistency in decision making. Hierarchical regression analyses showed that a large amount of age-related variance in performance on the decision tasks could be accounted for by exogenous social variables, health measures, basic cognitive skills, and attitudinal measures. The discussion emphasizes several directions for future research, including the need to validate the meaning of performance on these tasks for real-life decision processes.
