The short-term impacts of atrazine (herbicide), tributyltin (organometal) and copper on the behaviour of Euglena gracilis Klebs (Euglenophyta) were assessed. First, the ECOTOX automated image analysis system was used, which measured swimming velocity, cell shape, percentage of cells swimming upwards, and randomness of swimming. Next, visual observation by microscopy was used to measure percentage of cell motility and cell shape. Behavioural changes can be used as an indicator of stress in less than 24 h, potentially making them suitable for inclusion in early-warning systems for water quality. Findings indicate that E. gracilis is a very sensitive organism to copper, showing inhibition of motility with visual observation at 0.8 μmol/L within 1 h. The image analysis system was in general less sensitive than visual observation for detecting behavioural changes after incubation in copper. In contrast, after exposure to organic contaminants atrazine and tributyltin, the ECOTOX system detected small changes in the number of cells swimming upwards (antigravitactic behaviour) at higher concentrations.  相似文献

Thresholds for detecting the presence of amplitude modulation in a noise carrier were determined for rats using conditioned avoidance procedures. There was a progressive increase in threshold with modulation rates between 5 Hz and 2 kHz. Further tests were conducted to determine difference thresholds for detecting an increase in modulation rate for standard rates of 10, 50, and 100 Hz. The size of the difference threshold increased progressively as the standard rate increased. In addition, thresholds for detecting an increase in the duration of a noise burst were determined for various standard durations. The difference thresholds were constant for values between 10 and 50 ms but increased progressively, with standard durations between 0.1 and 1.0 s. As building owners, designers, and operators aim to achieve significant reductions in overall energy consumption, understanding and evaluating the probable impacts of occupant behavior becomes a critical component of a holistic energy conservation strategy. This becomes significantly more pronounced in ultra-efficient buildings, where system loads such as heating, cooling, lighting, and ventilation are reduced or eliminated through high-performance building design and where occupant behavior-driven impacts reflect a large portion of end-use energy. Further, variation in behavior patterns can substantially impact the persistence of any performance gains. This paper describes a methodology of building occupant behavior modeling using simulation methods developed by the Building Energy Research Center (BERC) at Tsinghua University using measured energy consumption data collected by the University of Washington Integrated Design Lab (UW IDL). The Bullitt Center, a six-story 4831 m2 (52,000 ft2) net-positive-energy urban office building in Seattle, WA, USA, is one of the most energy-efficient buildings in the world (2013 WAN Sustainable Building of the Year Winner). Its measured energy consumption in 2015 was approximately 34.8 kWh/(m2?yr) (11 kBtu/(ft2?yr)). Occupant behavior exerts an out-sized influence on the energy performance of the building. Nearly 33% of the end-use energy consumption at the Bullitt Center consists of unregulated miscellaneous electrical loads (plug-loads), which are directly attributable to occupant behavior and equipment procurement choices. Approximately 16% of end-use energy is attributable to electric lighting which is also largely determined by occupant behavior. Key to the building’s energy efficiency is employment of lighting controls and daylighting strategies to minimize the lighting load. This paper uses measured energy use in a 330 m2 (3550 ft2) open office space in this building to inform occupant profiles that are then modified to create four scenarios to model the impact of behavior on lighting use. By using measured energy consumption and an energy model to simulate the energy performance of this space, this paper evaluates the potential energy savings based on different occupant behavior. This paper describes occupant behavior simulation methods and evaluates them using a robust dataset of 15 minute interval sub-metered energy consumption data. Lighting control strategies are compared via simulation results, in order to achieve the best match between occupant schedules, controls, and energy savings. Using these findings, we propose a simulation methodology that incorporates measured energy use data to generate occupant schedules and control schemes with the ultimate aim of using simulation results to evaluate energy saving measures that target occupant behavior.  相似文献

Administered a biographic inventory, Holland's Vocational Preference Inventory, and the Adjective Check List (ACL) to 50 active and 50 resigned priests. Resigned Ss could not be differentiated from those who remained based on personality variables and person-environment interaction factors as expressed in J. Holland's theory of vocational choice or by demographic data. On the ACL, resigned priests scored significantly higher on confidence, achievement, dominance, endurance, and order. The reduction and degradation of total organic carbon (TOC) and bacteria from a secondary effluent of synthetic slaughterhouse wastewater using vacuum-ultraviolet (VUV) and ultraviolet-C (UV-C) processes and their combination (UV-C/VUV and VUV/UV-C) were investigated. The TOC removal rates under continuous mode operation ranged from 5.5 to 6.2%. In addition, the treatment with the UV-C/H2O2 and VUV/H2O2 processes under continuous mode operation doubled the TOC removal rates 10.8 and 12.2%, respectively. The optimum molar ratio of H2O2/TOC was found to be 2.5 and 1.5 for the UV-C and VUV processes, respectively. It was observed that all photochemical processes were able to totally inactivate different strains of bacteria with concentrations up to 105 CFU/mL within 27.6 s. Finally, a kinetic model was developed to simulate the TOC degradation from a secondary effluent of synthetic slaughterhouse wastewater.  相似文献

Recent developments in water quality research have highlighted difficulties in accurately predicting the incidence of pathogens within freshwater based on the viability, culturability and metabolic activity of indicator organisms. QPCR-driven assays are candidates to replace standard culture-based methods, however, protocols suitable for routine use have yet to be sufficiently validated. The objective of this study was to evaluate five oligonucleotide primers sets (ETIR, SINV, exoT, VS1 and ipaH2) for their potential applicability in qPCR assays to detect contamination from five waterborne bacterial pathogens (Escherichia coli O157:H7, Salmonella Typhimurium, Campylobacter jejuni, Pseudomonas aeruginosa, and Shigella flexneri). An enrichment-free qPCR protocol was also tested using S. Typhimurium-seeded source water, combining membrane filtration and mechanical, chemical and enzymatic lysis techniques to recover the bacterial cells. All five primer sets were found to have high specificity and sensitivity for the tested organisms. Four of the primers were able to detect pathogen loads as low as 10 cells/mL while 200 cells/mL of C. jejuni were detectable in pure culture. Although sensitivity decreased in an artificially contaminated environmental matrix, it was still possible to detect as few as 10 S. Typhimurium cells without enrichment. The primers and protocols evaluated in this study have demonstrated potential for further validation for possible application alongside traditional indicator techniques.  相似文献

The bacterial community composition in an activated sludge plant treatment from a bleached kraft pulp mill was monitored over a period of 209 days. Using DGGE and terminal-Restriction Fragment Length Polymorphism (t-RFLP) analysis we generated community DNA fingerprints over the time period. Both methods produce fingerprints that can be used to monitor stability in the system and generate fragments that can be associated with bacterial taxa. Chemical and physical parameters were also collected during that same time frame. We found a number of significant correlations with influent variables such as temperature, chemical oxygen demand (COD), Biochemical oxygen demand (BOD) and chloroform concentrations suggesting that these were the most likely parameters to influence the bacterial community structure. In addition several taxa correlated to important performance indicators such as COD/BOD removals and SVI. Multivariate analysis also confirmed the strong links between taxa variation and temperature, nutrient loads, chloroform and also one class of filaments. Establishing the identity of these taxa and their ecological preferences will greatly enhance our understanding and management of biological treatment systems.  相似文献

The dependence of the mechanical properties such as strength, modulus and fracture toughness on the volume fraction of the reinforcing glass fibres and glass beads in polyoxymethylene (POM) matrix was studied. The majority of the measured quantities in tension or flexure tests, seemed to be linear functions of either the volume fraction of the glass fibres or the glass beads. The relationship between some individual mechanical properties seemed to be linear as well. Consequently, many of the mechanical properties of these POM composites (POM/GF and POM/GB) could be estimated from one measured property using the relationships presented. Also, the same property measured for the two composite systems was found to be linearly related. Consequently, the mechanical properties of one composite system, (i.e. POM/GB) could be used to determine that of the other system (i.e. POM/GF) at the same filler concentration.  相似文献

The multitude of food recalls in 2007 clearly demonstrated that total nitrogen-content (ΣN) determination by means of near-infrared spectroscopy (NIRS) and Kjeldahl-based measurements can be deceived, and should no longer be regarded as a complete quality assurance program for nutritive-protein evaluations. Furthermore, contemporary Canadian-employed analytical tools are precariously limited in their ability to effectively assure a product where there is no a priori knowledge of the environmental toxin(s) involved. In light of these challenges, this study explored a number of analytical techniques used to assess and furthermore assure the quality of vegetable protein products (VPPs). Using liquid chromatography with tandem mass spectrometry (LC/MS/MS) technologies, a combination of VPP-based samples was analyzed for the presence of nitrogen-bearing environmental toxicants. Of the 52 samples tested, involving an assortment of matrices, melamine and cyanuric acid were positively identified (>1 ng/mL) in 22 and 17 samples, respectively. Subsequent high pressure liquid chromatography with ultraviolet/visible (HPLC-UV) amino acid profiling further confirmed the adulteration of those materials contaminated with melamine and melamine-related compounds. Based on the evidence presented herein, LC/MS/MS in combination with HPLC-UV provides for a reliable food safety detection system as applied to VPPs. Moreover, HPLC-UV is indispensable as a stand-alone 1st level of screening to assess the integrity of a VPP or any nutritive protein-based sample.  相似文献
