The global market for liquid mixed‐metal stabilizers is migrating toward heavy‐metal‐free products, Historically, these systems have not been performance and cost competitive. High efficiency calcium‐zinc stabilizers have the potential to replace heavy‐metal‐based products. An assay was developed for the specific detection of Salmonella enterica serotype Enteritidis, using a novel application of the polymerase chain reaction (PCR). This PCR assay is based on the mismatch amplification mutation assay, an allele-specific reaction, and can discriminate Enteritidis from all other salmonella. PCR primers were selected to amplify a 351-base pair (bp) DNA fragment from the salmonella plasmid virulence A (spv A) gene of Enteritidis. A single base difference at position 272 is present between the nucleotide sequence of the spvA gene of Enteritidis and other salmonellae. The downstream PCR primer, that encompasses position 272 of the Enteritidis spvA gene, was designed to contain a single base mismatch at the penultimate position, resulting in a 1-base mismatch with Enteritidis and a 2-base mismatch with other salmonellae that harbour the virulence plasmid. The upstream primer was completely homologous with the region immediately 5' to the spvA gene. When these primers were used and the annealing and extension reactions were performed at the same temperature, the PCR assay was specific for Enteritidis; no PCR product was detected for 40 other serotypes and 28 different genera examined. In pure culture, 120 colony forming units (c.f.u.) could be detected; a PCR product was observed from template derived from a 5 h enrichment broth culture of chicken seeded with 1 c.f.u. per gram of Enteritidis. This PCR assay is specific, reproducible, and less time consuming than the standard bacteriological methods used to detect Enteritidis.  相似文献

Senescent foliage from pines is potentially a large contributor to the total monoterpene content of the litter layer, and the availability of these compounds as phytotoxins may result from release of these compounds into the vapor phase. In order to determine the fate of several monoterpene hydrocarbons in the natural environment, we examined their concentrations in fresh, senescent, and decaying needles from 32 single-leaf pinyon pine (Pinus monophylla Torr. & Frem.: Pinaceae) trees growing at two different locations. Total monoterpene content was highest in the fresh needles (mean=5.6 ± 2.2 mg/g extracted air dry weight), but also remained relatively high in senescent needles (mean=3.6 ±1.8 mg/g extracted air dry weight), either still attached to the tree or forming the freshest layer of understory litter. Decaying needles within a dark decomposing layer of litter material 5–20 cm from the surface were found to contain much lower amounts of total monoterpenes (average: =0.12 ±0.06 mg/g extracted air dry weight). Further investigation of the fate of these compounds in the pinyon understory is required to determine if these hydrocarbons are indeed exerting phytotoxic characteristics.  相似文献

An experimental study of the gas-dynamic aspects of the high-velocity oxyfuel (HVOF) thermal spray process has been performed using commercially available HVOF equipment (Hobart-Tafa JP-5000, Ho-bart-Tafa Technologies, Inc., Concord, NH). Optical diagnostic techniques, including microsecond-expo-sure schlieren and shadowgraph imaging, were applied to visualize the hot supersonic jet produced by this equipment without particle injection. Rapid turbulent mixing of the jet with the surrounding atmos-phere was observed, which is an issue of concern in coating quality due to the possibility of oxidation of sprayed particles. This mixing appears to be a function of the ratio of densities of the hot jet and the cold atmosphere as well as a function of the velocity of the jet, rather than one of combustion-chamber pres-sure or barrel length. The supersonic core of the HVOF jet dissipates rapidly due to the mixing, so that the jet is no longer supersonic when it impinges on the target surface being sprayed. Secondary issues also observed in this study include strong jet-noise radiation from the HVOF plume and the entrainment and induced bulk motion of the surrounding air.  相似文献

Zirconium or hafnium additions to austenitic stainless steels caused a reduction in grain boundary Cr depletion after proton irradiations for up to 3 dpa at 400 °C and 1 dpa at 500 °C. The predictions of a radiation-induced segregation (RIS) model were also consistent with experiments in showing greater effectiveness of Zr relative to Hf due to a larger binding energy. However, the experiments showed that the effectiveness of the solute additions disappeared above 3 dpa at 400 °C and above 1 dpa at 500 °C. The loss of solute effectiveness with increasing dose is attributed to a reduction in the amount of oversized solute from the matrix due to growth of carbide precipitates. Atom probe tomography measurements indicated a reduction in amount of oversized solute in solution as a function of irradiation dose. The observations were supported by diffusion analysis suggesting that significant solute diffusion by the vacancy flux to precipitate surfaces occurs on the time scales of proton irradiations. With a decrease in available solute in solution, improved agreement between the predictions of the RIS model and measurements were consistent with the solute-vacancy trapping process, as the mechanism for enhanced recombination and suppression of RIS.  相似文献

Rate theory modeling was used to simulate the effects of oversized solute additions on radiation-induced segregation in austenitic stainless steels. The purpose was to understand the effects of a solute-vacancy trapping mechanism on radiation-induced segregation and to define key parameters that most affect segregation behavior. Sensitivity analysis of the model showed the solute-vacancy binding energy to be the most important model parameter. Binding energies from ab initio first principles were calculated for oversized solutes of Pt, Ti, Hf and Zr, with energies of 0.31, 0.39, 0.71 and 1.08 eV, respectively. Differences in binding energies, despite similar sizes of the atoms, suggests that the short-range electronic interactions play an important role in determining binding energy. The model results show oversized solutes to be most effective at reducing grain boundary Cr depletion at temperatures of 450-500 °C for a dose rate applicable to proton irradiations. The reduction increases with increasing oversized solute concentration, where it saturates at approximately 0.1 at.%.  相似文献

This paper presents results obtained by the experimental technique of photoelastic stress analysis and by finite or discrete element analysis, applied to the case of buckling of a single, axially-loaded metal fiber embedded in a plastic matrix. The program was conducted utilizing a mild steel fiber embedded in an epoxy resin system. The fiber was loaded axially with an increasing compressive force until buckling occurred. Along with determination of the critical load for the fiber, the maximum shearing stress at any point in the epoxy matrix was determined using two-dimension photoelastic stress analysis. In the analytical study, the fiber was modeled by a series of bending elements and the matrix by an assemblage of constant-strain triangular elements. The total system was thus modeled by the two types of elements connected at common nodal points. The instability was solved as a standard eigenvalue problem and the stress tensor was obtained by finite element stress analysis.  相似文献

Although rapid smoking has proved to be a successful treatment strategy for smokers, its hazard potential has been recently debated. In the present study, 6 Ss (mean age 30.8 yrs) were monitored throughout 8 standard sessions of rapid smoking. Heart rate, blood pressure, and carboxyhemoglobin increases were generally higher than those reported in the few extant physiological studies of rapid smoking, and EKG abnormalities occurred in several Ss during rapid smoking. The present study investigated the tolerability, safety profile, and anti-obsessional efficacy of sertraline, a selective serotonin reuptake inhibitor, during long-term treatment of patients with obsessive-compulsive disorder (OCD). Fifty-nine OCD patients who had completed a 1 year double-blind, fixed dose study comparing sertraline and placebo subsequently entered a 1-year open extension. Among the 51 patients who had been treated with sertraline during the double-blind phase, the mean total duration of sertraline treatment was 690 days. Only treatment responders who completed the 52-week double-blind treatment phase were permitted to enter the open extension. The higher rate (p < 0.02) of sertraline patients (51 out of 241) than of placebo patients (eight out of 84), who responded to treatment and entered the open-label phase is therefore consistent with the greater mean improvement observed in the sertraline group during double-blind treatment. Placebo responders differed from sertraline responders in that they were less impaired at baseline of the double-blind study [Yale-Brown Obsessive-Compulsive Scale (Y-BOCS) of 18.5 versus 23.4] and they exhibited less improvement during double-blind treatment (-6.1 versus -11.4). In the open-label phase all patients received sertraline at a starting dose of 50 mg once a day, titrated in 50 mg increments to a maximum dose of 200 mg according to clinical response. At end-point the mean Y-BOCS score for all patients decreased by a further 3.6 points. Patients previously treated with placebo showed greater improvement after being switched to sertraline than those who received continued sertraline treatment. Patients who completed the study and received 2 full years of sertraline treatment (n = 38) exhibited a mean improvement of 15.6 points using the Y-BOCS. Sertraline was well tolerated during both the double-blind phase and the open extension, and the incidence of adverse experiences was generally reduced during the second year of treatment. Three patients discontinued open treatment because of adverse experiences. Long-term sertraline treatment did not appear to be associated with the emergence, increased incidence, or increased severity of adverse experiences or clinically significant abnormalities in laboratory tests, vital signs, or the electrocardiogram. The study supports the long-term safety and tolerability of sertraline over a 2-year treatment course and the sustained efficacy of sertraline in patients with OCD.  相似文献

To test a hypothesis from self-efficacy theory, we randomly assigned 149 subjects to verbal or mathematics and success or failure conditions in which they attempted to solve easy or difficult anagram or number series tasks. Changes in task self-efficacy and task interest as a result of task success or failure were in accordance with predictions from self-efficacy theory. We also examined the generalizability of the effects of task performance. The results indicated that task performance effects generalized to self-efficacy and interest ratings on an irrelevant task and to global ratings of math and verbal ability. Task performance effects did not generalize to career self-efficacy and career interest measures but consistent gender differences in self-efficacy emerged as a result of both math and verbal task performance.
