Accuracy of patient positioning and dose delivery in mantle field irradiations was investigated on 29 patients treated between August 1990 and December 1991. Patients were treated in two different centers, University Hospital St. Rafa?l in Leuven and Institut Gustave Roussy in Villejuif, where different techniques and procedures were used. Measurements were performed on 341 portal films and entrance doses were measured in 518 treatment set-ups. The impact of systematic errors occurring during treatment preparation and day-to-day variations on the accuracy of treatment execution were separately analysed. Daily reproducibility, defined as the deviation from the respective mean measured value for a treatment was demonstrated to be good for both the treated volume and the delivered dose and no difference between the two techniques was shown. Comparing the successive portal films of individual patients (reproducibility of a treatment, once it has started), only small day-to-day variations are found: the SD is 3.4 mm for craniocaudal movements and 2.6 mm for lateral movements. For dose delivery very narrow distributions are obtained with SDs of, respectively, 1.5% and 1.85% for the Leuven and the Villejuif group. This suggests that the position of the patient, which is often thought as the critical point in this complex set-up can be done in a very accurate way, regardless of the position used. To assess the global accuracy of the treatment, the actually treated volume and delivered dose were compared with the planned values. The feasibility of quantifying the perceived active ingredient (P57) in Hoodia gordonii raw material using Fourier transform near- and mid-infrared spectroscopy combined with chemometric techniques was investigated. The concentration of P57 (a triterpene glycoside) was determined in 146 plant samples with liquid chromatography coupled to mass spectrometry and these values were used to develop a calibration model based on the partial least squares projections to latent structures (PLS) and orthogonal projections to latent structures (O-PLS) regression algorithms. The performance of each calibration model was evaluated according to the root mean square error of prediction (RMSEP) and correlation coefficient (R2). The PLS model with 2nd derivative pre-processing predicted P57 content based on the FT-NIR spectra with the best accuracy and a correlation coefficient (R2) value of 0.9629 and the lowest RMSEP of 0.03%. These results demonstrated that FT-NIR spectroscopy can be used to rapidly quantify P57 in H. gordonii raw material with high accuracy.  相似文献

Secretory proteins are cotranslationally translocated across the mammalian ER membrane through an aqueous pore in the translocon while the permeability barrier is maintained by a tight ribosome-membrane junction. The lumenal end of the pore is also blocked early in translocation. Extraction of soluble lumenal proteins from microsomes and reconstitution with purified proteins demonstrate, by fluorescence collisional quenching, that BiP seals the lumenal end of this pore. BiP also seals translocons that are assembled but are not engaged in translocation. These ribosome-free translocons have smaller pores (9-15 A diameter versus 40-60 A in functioning translocons) and are generated when ribosomes dissociate from functioning translocons with large pores. BiP therefore maintains the permeability barrier by sealing both nontranslocating and newly targeted translocons.  相似文献

This paper illustrates a comparative study to analyze the physical differences between numerical simulations obtained with both the conservation and incompressible forms of the Navier-Stokes equations for natural convection flows in simple geometries. The purpose of this study is to quantify how the incompressible flow assumption (which is based upon constant density advection, divergence-free flow, and the Boussinesq gravitational body force approximation) differs from the conservation form (which only assumes that the fluid is a continuum) when solving flows driven by gravity acting upon density variations resulting from local temperature gradients. Driving this study is the common use of the incompressible flow assumption in fluid flow simulations for nuclear power applications in natural convection flows subjected to a high heat flux (large temperature differences). A series of simulations were conducted on two-dimensional, differentially heated rectangular geometries and modeled with both hydrodynamic formulations. From these simulations, the selected characterization parameters of maximum Nusselt number, average Nusselt number, and normalized pressure reduction were calculated. Comparisons of these parameters were made with available benchmark solutions for air with the ideal gas assumption at both low and high heat fluxes. Additionally, we generated specific force quantities and velocity and temperature distributions to provide a basis for further analysis. The simulations and analysis were then extended to include helium at the Very High Temperature gas-cooled Reactor (VHTR) normal operating conditions. Our results show that the consequences of incorporating the incompressible flow assumption in high heat flux situations may lead to unrepresentative results. The results question the use of the incompressible flow assumption for simulating fluid flow in an operating nuclear reactor, where large temperature variations are present.  相似文献

OBJECTIVE: Diabetic retinopathy (DR) is a major cause of visual impairment and blindness in adults. Antioxidant nutrients, such as vitamins C and E and beta-carotene, may be protective of some eye disorders, such as cataract and age-related macular degeneration, but a relationship between these nutrients and DR has yet to be defined. The purpose of this study was to examine the relation between dietary and supplement intakes of vitamins C, E, and beta-carotene and the risk of DR. DESIGN: Both cross-sectional and longitudinal data were collected from participants in the San Luis Valley Diabetes Study, including non-Hispanic white and Hispanic adults in southern Colorado. PARTICIPANTS: A total of 387 participants with type 2 diabetes completed at least 1 complete retinal examination and 24-hour dietary recall (including vitamin supplement use). MAIN OUTCOME MEASURES: Type 2 diabetes was defined according to World Health Organization criteria. DR was assessed by retinal photographs, using the Airlie House criteria to classify DR as none, background, preproliferative, or proliferative. Data for both eyes, from up to three clinic visits per participant, were used for analysis. Ordinal logistic regression analysis was used, taking advantage of multiple clinic visits by individual participants and observations from both eyes, to assess the risk for increased DR severity over time as a function of changes in intake of vitamin C, vitamin E, and beta-carotene. Six categories of intake for each nutrient (first to fourth quintiles and ninth and tenth deciles) were considered to ascertain any potential threshold effect. Analyses accounted for age, duration of diabetes, insulin use, ethnicity, glycated hemoglobin, hypertension, gender, and caloric intake. RESULTS: An increase over time in vitamin C intake from the first to ninth deciles was associated with a risk for increased severity of DR (odds ratio = 2.21, P = 0.01), although excess risk was not observed for the tenth decile or the second through fourth quintiles compared to the first quintile. Increased intake of vitamin E was associated with increased severity of DR among those not taking insulin (odds ratios = 2.69, 2.59, 3.33, 5.65, 3.79; P < 0.02, for an increase over time from the first to the second through fourth quintiles and ninth and tenth deciles, respectively). Among those taking insulin, increased intake of beta-carotene was associated with a risk for severity of DR (odds ratio = 3.31, P = 0.003, and 2.99, P = 0.002, respectively, for the ninth and tenth deciles compared to the first quintile). CONCLUSIONS: No protective effect was observed between antioxidant nutrients and DR. Depending on insulin use, there appeared to be a potential for deleterious effects of nutrient antioxidants. Further research is needed to confirm associations of nutrient antioxidant intake and DR.  相似文献

Contents The voltage-frequency relationship and its influence on the losses in the inverter fed induction motor drive is investigated. It is found that the control condition yielding minimum losses very nearly coincides with the control condition requiring minimum motor current for a particular load and speed, and it is shown that there exists an optimum voltage vs torque relationship applicable to all frequencies.

Übersicht Der Spannungs-Frequenz-Zusammenhang und sein Einfluß auf die Verluste eines wechselrichtergespeisten Induktionsmotor-Antriebs wird untersucht. Es wird gefunden, daß der Regelungszustand, der minimale Verluste ermöglicht, näherungsweise mit einem minimalen Effektivstrom für eine beliebige Last und Geschwindigkeit übereinstimmt. Es wird auch gezeigt, daß ein optimales Spannungs-Drehmoment-Verhältnis besteht, das für alle Frequenzen gilt.

Although many absorption enhancers have been investigated, very few are used clinically. A need exists therefore for more effective absorption enhancers. The drug-absorption-enhancing effects of combinations of N-trimethyl chitosan chloride (TMC) with degrees of quaternization of 48 and 64%, dicarboxymethyl chitosan oligosaccharide, and chitosan lactate oligomer with monocaprin and melittin were compared to their individual performances using the in vitro Caco-2 cell model. Combining the absorption enhancers showed synergism in both the reduction of the transepithelial electrical resistance (TEER) and the enhancement of the transport of a macromolecular model compound across this intestinal epithelial cell layer. Lower concentrations of the absorption enhancers in the combination groups exhibited greater effects on the epithelial cells compared with the individual absorption enhancers.  相似文献

Human telomerase, a ribonucleoprotein enzyme is known to be associated with immortalized cancer cells but is absent in most normal tissues. Thus, telomerase appears to be an attractive new target for anticancer agents and an important diagnostic marker of human cancers. Here, we describe an improved telomerase assay method based on the Dynabead biomagnetic separation theory. In this method, 5'-biotinylated (TTAGGG)3 was used as a primer for the telomerase reaction. Telomerase reaction products were then immobilized on streptavidin-coated Dynabeads and washed intensively to eliminate excess [alpha-32P]dGTP. Using this method, without the amplification of telomerase reaction products by the PCR, we were able to quantitatively detect telomerase activity in human HeLa cell extracts equivalent to between 200-500 cells. This method is anticipated to be useful for the measurement of telomerase activity in various tumor cells, for assessing potential telomerase and for understanding the biochemical aspects of the telomerase reaction.  相似文献

This study examined types and frequency of coaching of learning at the middle school level and its relation to students' strategic learning. Teachers (n?=?11) were videotaped during daily instruction on 3 separate occasions, and students (n?=?235) responded to a self-report questionnaire about learning-strategy use. Consistent with previous research at the elementary level (E. B. Moely et al., 1992), average instances of teachers' coaching of learning (e.g., describing cognitive processes, suggesting strategy use) occurred in only 9% of the instruction segments. Teachers most frequently recommended use of learning aids, engagement in metacognitive activity, and use of elaboration strategies. Results from a multiple regression analysis indicate that students' strategic-learning activity was significantly related to teachers' coaching of learning (R2?=?.18). Evaluation of the mucoadhesive properties of N-trimethyl chitosan chloride   总被引:5,自引:0,他引:5  
Previous studies have established that N-trimethyl chitosan chloride (TMC) is a potent absorption enhancer for peptides and large hydrophilic compounds across mucosal surfaces, especially in neutral and basic environments where chitosan is ineffective as an absorption enhancer. The degree of quaternization of TMC plays an important role on its absorption-enhancing properties. Several TMC polymers with different degrees of quaternization were synthesized and the molecular mass of the polymers was determined by SEC/MALLS. The mucoadhesive properties of the TMC polymers were measured with a modified tensiometer based on the Willhelmy plate method. The effect of the TMC polymers on the surface tension of a mixture of polymer and mucus was measured with a Du Noüy tensiometer. The degrees of quaternization of the synthesized TMC polymers were between 22.1% and 48.8% and the molecular mass was above 100,000 g/mole for all the polymers. A decrease in mucoadhesivity with an increase in the degree of quaternization of the TMC polymers was found. Surface-tension analysis of a mixture of polymer and mucus showed the effect of excessive polymer hydration on mucoadhesion. The results show that the degree of quaternization of TMC had a pronounced effect on the mucoadhesive properties of this polymer. Although the mucoadhesive profiles for the TMC polymers were lower than the original chitosan, they still retained sufficient mucoadhesive properties for successful inclusion into mucoadhesive dosage forms.  相似文献
