Systemic sclerosis (SSc) is characterized by excessive collagen deposition in the skin and internal organs. Activated fibroblasts are the key effector cells for the overproduction of type I collagen, which comprises the α1(I) and α2(I) chains encoded by COL1A1 and COL1A2, respectively. In this study, we examined the expression patterns of α1(I) and α2(I) collagen in SSc fibroblasts, as well as their co-regulation with each other. The relative expression ratio of COL1A1 to COL1A2 in SSc fibroblasts was significantly higher than that in control fibroblasts. The same result was observed for type I collagen protein levels, indicating that α2(I) collagen is more elevated than α2(I) collagen. Inhibition or overexpression of α1(I) collagen in control fibroblasts affected the α2(I) collagen levels, suggesting that α1(I) collagen might act as an upstream regulator of α2(I) collagen. The local injection of COL1A1 small interfering RNA in a bleomycin-induced SSc mouse model was found to attenuate skin fibrosis. Overall, our data indicate that α2(I) collagen is a potent regulator of type I collagen in SSc; further investigations of the overall regulatory mechanisms of type I collagen may help understand the aberrant collagen metabolism in SSc.  相似文献

Four benzoic acid derivatives and three phenylpropanoids were tested for their ability to suppress oxidation of dichlorodihydrofluorescein (DCDHF) induced by 3-morpholinosydnonimine (SIN-1, a peroxynitrite donor). Ellagic acid was found to potently inhibit this oxidation and to inhibit oxidation of DCDHF induced by peroxynitrite itself. Moreover, this compound prevented peroxynitrite-induced oxidative single strand breaks in pTZ 18U plasmid DNA and nitration of tyrosyl residues in bovine serum albumin. These results show that ellagic acid protects biomolecules against oxidative and nitrative damage induced by peroxynitrite in vitro.  相似文献

Co-Ni-Al合金不仅可以作为铁磁性形状记忆合金,也是传统和高温形状记忆合金的候选材料,本文通过金相显微分析,DSC和VSM方法,研究了Co40Ni33.5Al26.5合金马氏体相变和Curie点随淬火温度变化的情况,结果发现该合金马氏体相变温度和Curie点与淬火温度成正比关系.马氏体相变的4种温度,即Ms、Mf、As和At基本平行变化,淬火温度每升高10℃,马氏体相变温度升高8~9℃,而Curie点升高6~7℃.其马氏体相变温度和Curie点随淬火温度的变化与基体相β的成分变化有关,β相中Al的含量随淬火温度升高而降低,因而马氏体相变温度和Curie点升高.并且发现随着淬火温度升高,Co40Ni33.5Al26.5合金马氏体相的磁晶各向异性有减弱的倾向.  相似文献

Phase equilibria and thermodynamic properties of the KCl–K2CO3–NaCl–Na2CO3 system were analyzed on the basis of the thermodynamic evaluation of the KCl–NaCl,KCl–K2CO3,NaCl–Na2CO3,K2CO3–Na2CO3 and KCl–K2CO3–NaCl–Na2CO3 systems. The Gibbs energies of individual phases was approximated by two-sublattice models for ionic liquids and crystals. Most of the experimental information was well described by the present set of thermodynamic parameters. The lowest monovariant eutectic temperature in the KCl–NaCl–Na2CO3 system is located at 573 °C, with a composition of XNa2CO3=0.31,XKCl=0.35 and XNaCl=0.34.  相似文献

With a potential-energy function of Co described by the embedded-atom method (EAM), molecular-dynamics (MD) simulations were performed for a series of initial fcc configurations with different types of dislocations or preset hcp embryos. The gliding process of a Shockley dislocation on a closepacked plane has been observed, which starts from the origin of the dislocation and proceeds at a high speed of 280 m/s toward a certain direction. An atom which has been swept by the dislocation line was detected to contribute a displacement close to the Burgers vector of a Shockley dislocation. It is in this way that a new stacking sequence is produced and an hcp lamella grows in the fcc structure. A similar gliding process has been observed in the case where an intrinsic stacking fault is preexisting in the fcc structure. The transformation is, again, toward forming a local hcp region. These results prove that a special dislocation in the fcc structure can act as an embryo of the hcp, as described in many dislocation mechanisms of the martensitic transformation. The fcc → hcp phase-tranformation process of Co has been further reproduced by a simulation initiated from an fcc/hcp two-phase configuration. Phase equilibria and martensitic and magnetic transitions of the β (B2 and L21) phase in the Ni–Fe–Ga system were investigated. The b phase was found to be in equilibrium with the γ (A1 structure) or γ′ (L12 structure) phase. The Curie temperature, T c , equilibrium temperature, T o 5 (Ms + Af)/2, martensitic transition starting temperature, M s , and reverse transition finishing temperature, Af , of the β single–phase alloys were sensitive to the Fe and Ga compositions. The Fe substitution for Ni decreased and increased the T o and T c , respectively. The Ga substitution for Ni or Fe decreased both the T o and T c . The entropy change accompanying the reverse martensitic transition showed compositional dependence due to the magnetic contribution. The saturation magnetization I s of the Ni–Fe–Ga system showed a strong dependence on the magnetic valence Z M . The Is values of the Ni–Fe–Ga alloys annealed at 1023 K showed the same Z m dependence as other ferromagnetic shape memory alloy (FSMA) systems. The antioxidative effects of vegetable extracts were evaluated using linoleic acid emulsion and liposomal phospholipid suspension systems. First, the antioxidative activities of water, ethanol and 2% metaphosphoric acid extracts of the vegetables were investigated by measuring the rate of oxygen consumption in the peroxidation of linoleic acid initiated by a radical generator, 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH), with a Clark electrode in the absence of metal chelators. Ethanol extracts of moroheiya and perilla exhibited the highest antioxidative activities. Without metal chelators, 12 ethanol extracts and one water extract acted as prooxidants, and this phenomenon could be attributed to the prooxidant action of ascorbic acid in the presence of free transition metal ions. Next, peroxyl radical-scavenging activities of vegetable extracts were examined by adding a metal chelator, diethylenetriaminepentaacetic acid (DTPA), to the linoleic acid emulsion system. The results were compared with their antioxidative activities in phospholipid bilayers by measuring the inhibition of lipid peroxidation in large unilamellar vesicles composed of egg yolk phosphatidylcholine (PC). A good correlation was observed between the antioxidative activities obtained from these two systems. The antioxidative activities in the liposomal phospholipid suspension system correlated with total contents of ascorbic acid and polyphenols in vegetable extracts. © 1999 Society of Chemical Industry  相似文献

利用金相显微组织分析技术、示差扫描量热法(DSC)和振动磁力计(VSM),考察了Co41Ni32Al27-xSix合金中Si元素含量x对马氏体相变和铁磁性转变的影响,用X射线衍射方法分析马氏体相的结构类型.增加x能够显著提高合金的马氏体相变温度,并且同时提高铁磁性转变Curie点;在x≤5的范围内,x增加1可以造成马氏体相变温度提高50-60 K,同时Curie点提高大约10 K;马氏体相的晶体结构仍然是L10型有序结构,但是随着x的增加,单胞体积减小.讨论了马氏体相变温度和Curie点同时提高的原因.  相似文献

Phase equilibria and martensitic and magnetic phase transformations of the β phase in the Co−Ni−Ga system have been investigated. It has been shown that the β phase is in equilibrium with the α-phase over a wide range compositions at 600–1100°C. The β phase exhibits both a paramagnetic-ferromagnetic transition and a thermoelastic martensitic transition from B2 to L10 structure. The Curie temperature T C increases with decreases with decreasing Ni content and with increasing Ga content. The composition region of the β phase exhibiting the thermoelatstic martensitic transformation from ferromagnetic austenite is located near the α+β two-phase region. T C and M s of α+β two-phase alloys increase with increasing annealing temperature. 利用金相和SEM,XRD,DSC和VSM研究了Co含量x对Ni52Ga28Fe20-xCox合金马氏体相变和铁磁性的影响。结果表明:x≤6%时(原子分数,下同),Co代替Fe显著提高合金的马氏体相变温度,每1%Co可以使马氏体相变温度提高50K~60K,但对Curie点的影响不大。淬火温度从1423K降低到773K对马氏体相变温度的影响不大,但使Curie点提高20K-30K,其原因是母相的B2→L21转变导致的有序化程度提高。粉末试样X射线衍射发现1423K和773K2种温度处理后都只出现L10(2M)晶体结构类型的马氏体,并且合金粉末化后降低了马氏体相变温度和增加了γ相的析出,但块状Ni52Ga28Fe17Co3合金773K淬火后为14M型马氏体。  相似文献
