Genes that encode enzymes that convert inactive "prodrugs" into anticancer metabolites may be therapeutically useful against brain tumors. Unlike other genes tested to date in brain tumor models, the Escherichia coli gpt gene is unique in that it not only sensitizes cells to the prodrug 6-thioxanthine (6TX) but also encodes resistance to a different regimen (mycophenolic acid, xanthine, and hypoxanthine), thus providing a means to select for gpt-positive cells. In the present study, rat C6 glioma cells were infected with a retrovirus vector that transduces this gene. A clonal line (C6GPT-7) was derived that exhibited significant 6TX susceptibility in vitro with an ID50 of 2.5 mumol/L, whereas 50% growth inhibition of parental C6 cells was not achieved at concentrations tested (up to 50 mumol/L). This line also exhibited significant sensitivity to 6-thioguanine (6TG), with an ID50 of 0.05 mumol/L, whereas 50% growth inhibition of parental C6 cells was achieved at 0.5 mumol/L. In a "bystander" assay, C6GPT-7 tumor cells efficiently transferred 6TX sensitivity to C6 cells at ratios as low as 1:9 (C6GPT-7:C6). This in vitro bystander effect was abrogated when C6GPT-7 and C6 cells were separated by a microporous membrane, suggesting that it was not mediated by highly diffusible metabolites. In vivo both 6TX and 6TG significantly inhibited the growth of subcutaneously transplanted C6GPT-7 cells but not that of C6 cells in athymic mice. In an intracerebral model, both 6TX and 6TG exhibited significant antiproliferative effects against tumors formed by C6GPT-7 cells. Conducted 2 experiments with a total of 140 male and 140 female hooded rats. In Exp. I, frequencies of upright postural responses displayed by Ss when shocked alone were predictive of upright postural response frequencies during shock-elicited aggression. These differences in upright postural frequencies were associated with differences in frequencies of aggression in male Ss only. Exp. II manipulated upright postural responses. Results suggest that the upright behavior of Ss could be controlled in the shock-elicited-aggression paradigm. Only a slight influence on the aggression of female Ss was obtained. The need for reliable simulation systems has been discussed and recognized by many researchers. At the same time researchers have recognized that the quality of a simulation model's results are strictly related to the quality of the input probability distribution functions. The data used in this research were acquired from the Atkinson-Washington-Zachry joint venture on the Eastside Reservoir Project in California. The data were analyzed using BestFit software to obtain the parameters of the theoretical distribution functions that best described the field data set. The research validated previous warnings about the influence of the class interval decision on the selection of a distribution function when the chi-square fitting test is utilized. To enable a better characterization of pathophysiologic processes in colon and rectum, we have developed a perfusion technique for collection of soluble substances and cells from standardized intestinal segments. A tube with balloons attached to its outer wall was endoscopically introduced into the rectum and sigmoid colon, and its position ascertained fluoroscopically. The balloons delimited two segments, one in the sigmoid colon and one in the rectum. The segments were simultaneously perfused by a buffer at 37 degrees C. After a 30-min rinsing period, perfusate and cells were collected at 20-min intervals. Of 51 attempted perfusions, 45 were successfully completed. Recovered volumes equaled those infused. Leakage from the proximal intestine to the segments was negligible. In 18 healthy volunteers, the mean perfusate concentration from the rectal segment was 57.5 (27.5-120.2) mg/L for albumin, 1.3 (1.0-1.7) micrograms/L for eosinophil cationic protein, 5.1 (2.8-9.5) ng/L for prostaglandin E2, and 61.7 (41.7-89.1) micrograms/L for hyaluronan, and all values were lower in the sigmoid segment. Steady state conditions were achieved from the second perfusion period. The perfusate contained 4-80 x 10(4) cells, more than 95% of which were epithelial cells. A three-dimensional steady-state model of the industry-standard AG Associates 4108 Heatpulse Rapid Thermal Processing system has been developed for the study of thermal uniformity across 8 inch wafers. The model combines radiation energy transfer among all solid surfaces in the chamber with energy transfer among the chamber materials and to the process ambient. Surfaces included are those of the tungsten filaments of the lamps, the silicon wafer, the polysilicon annular thermal guard ring, the quartz process tube, and the gold reflectors which surround the lamps and process tube. These surfaces are divided into approximately 4800 individual surface elements for the radiation transfer allowing very accurate thermal analysis. The model has previously been shown to provide very good agreement with experiment for temperature distributions across an 8 inch wafer. The model is presently used to make quantitative examinations of asymmetric effects occurring in a RTP chamber which cannot be examined by 2-dimensional models.
