We present a novel strategy for sparse direct factorizations that is geared towards the matrices that arise from hp-adaptive Finite Element Methods. In that context, a sequence of linear systems derived by successive local refinement of the problem domain needs to be solved. Thus, there is an opportunity for a factorization strategy that proceeds by updating (and possibly downdating) the factorization. Our scheme consists of storing the matrix as unassembled element matrices, hierarchically ordered to mirror the refinement history of the domain. The factorization of such an ‘unassembled hyper-matrix’ proceeds in terms of element matrices, only assembling nodes when they need to be eliminated. The main benefits are efficiency from the fact that only updates to the factorization are made, high scalar efficiency since the factorization process uses dense matrices throughout, and a workflow that integrates naturally with the application.  相似文献

In response to the concern that Minnesota Multiphasic Personality Inventory-2 (MMPI-2; J. N. Butcher, W. Dahlstrom, J. R. Graham, A. Tellegen, & B. Kaemmer, 1989; J. N. Butcher et al., 2001) Variable Response Inconsistency (VRIN) and True Response Inconsistency (TRIN) score invalidity criteria recommended for use with American samples results in an excessive number of exclusions in Asian samples (F. M. Cheung, W. Z. Song, & J. X. Zhang, 1996), we examined the cross cultural equivalence of the original VRIN and TRIN scales, and developed and validated Korean-specific VRIN and TRIN scales with Korean adult normative, clinical, and college samples. Although the results from item pair correlation analyses suggested the superiority of the Korean VRIN and TRIN over the original VRIN and TRIN, the mean comparison results and classification accuracy statistics using data with varying degrees of randomly inserted true and/or false responses did not reveal a strong advantage of one version over the other. Cholangiocarcinoma (CCC) is the second most primary liver cancer with an aggressive biological behavior, and its incidence increases steadily. An aberrant up-regulation of the sonic hedgehog signaling pathway has been reported in a variety of hepatic diseases including hepatic inflammation, fibrosis, as well as cancer. In this study, we determined the effect of a sonic hedgehog inhibitor, vismodegib, on the development of CCC. Through database analyses, we found sonic hedgehog signaling was up-regulated in human CCC, based on overexpression of its target genes, GLI1 and GLI2. Further, human CCC cells were highly sensitive to the treatment with vismodegib in vitro. Based on the data, we investigated the in vivo anti-cancer efficacy of vismodegib in CCC employing a murine model of CCC developed by hydrodynamic tail vein injection method. In the murine model, CCC induced by constitutively active forms of TAZ and PI3K exhibited up-regulated sonic hedgehog signaling. Treatment of vismodegib significantly suppressed tumor development in the murine CCC model, based on comparison of gross morphologies and liver weight/body weight. It is expected that pharmacological inhibition of sonic hedgehog signaling would be an effective molecular target therapy for CCC.  相似文献
