Investigated rationale content, an assumed cognitive determinant of resistance to deviation in children, using a standard punishment paradigm with 120 7-, 9-, and 11-yr-old children. Sex of child, age of child, rationale focus, and rationale orientation were combined in a 2 * 3 * 2 * 2 factorial design with a female as a punitive agent. Maturity of moral judgment, sex of child, rationale focus, and rationale orientation were used as factors in another series of analyses. Rationales were significantly more effective with girls than boys, and response inhibition was greatest for intention-focused and person-oriented rationales. However, sex and age of the child were interacting factors. The data suggest that level of moral judgment influences moral behavior. The consequences of glucocorticoid receptor (GR) dysfunction for neuroimmunoendocrine responses to an inflammatory challenge were studied in transgenic mice expressing antisense RNA directed against the GR [GR-impaired (GR-i) mice]. Mice were implanted intraperitoneally with a biotelemetry transmitter to monitor body temperature and locomotion. GR-i mice showed decreased locomotion and body temperature during the dark phase of the diurnal cycle. Intraperitoneal administration of saline caused a rapid increase in body temperature in control mice, which was terminated within 90 min. In GR-i mice, however, body temperature remained elevated for about 6 h. Intraperitoneal injection of endotoxin (10 micrograms/mouse) produced a biphasic fever in control mice. However, in endotoxin-injected GR-i mice, body temperature was not significantly different from their saline-injected controls during the first 6 h. Body temperature then increased and remained elevated during the night period. Both strains showed hypolocomotion after endotoxin. In a second experiment, mice were injected intraperitoneally with saline or endotoxin and killed after 1, 3, 6 or 24 h. In GR-i mice, endotoxin caused an augmented rise in plasma ACTH, but not in corticosterone levels. The endotoxin-induced increase in serum levels of interleukin-1 beta and interleukin-6 was not different between the strains. However, whereas in control mice tumour necrosis factor-alpha levels were below detection at the time points studied, substantial levels of this cytokine were found in the serum of GR-i mice 1 h after endotoxin administration. It may be concluded that life-long impairment of GR evolves in aberrant physiological and humoral responses to an acute inflammatory challenge. These findings expand our understanding about the neuroendocrine and physiological disturbances associated with stress-related disorders.  相似文献

School psychology is not a recognized field in the People's Republic of China, therefore, most of the appropriate research has been conducted by educational or developmental psychologists. These researchers, like those in the US, are not practitioners in the field, and are located at universities. The focus of many research articles is on assessment or consultation-related areas. Research related to intervention is an infrequent topic. Both insufficient resources and trained professionals are 2 of the major problems facing applied research in China. Compared the effectiveness of an aversive stimulus, withholding of resources, withdrawal of love, and reasoning, when used alone and when combined with praise, in a standard laboratory punishment paradigm using 120 1st- and 2nd-grade boys and girls as Ss. Resistance to deviation and stability of resistance to deviation were used as the measures of punishment effectiveness. Sex of child, use of praise, and type of punisher were combined in a 2 * 2 * 5 factorial design, with a female as the punitive agent. An aversive stimulus appeared to produce more suppression of deviant behavior, but the effect was not significant for all response measures. The stability of the deviant response pattern varied for each punisher, but the most stable response pattern resulted from the use of an aversive stimulus. Neither use of praise nor sex of child significantly influenced punisher effectiveness. Results are discussed within the framework of anxiety arousal and information content of the punishers, and the practice of grouping various punishers under the categories of sensitization and induction measures is considered. Modulators of P-glycoprotein (P-gp) are often themselves transported out of cells, thereby limiting their effectiveness. It may be possible to develop more effective modulators of multidrug resistance by designing drugs that irreversibly block the function of P-gp. Therefore, we studied the effect of the mustard derivatives of fluphenazine (FPN) and trans-flupenthixol (FPT) on P-gp function. Both fluphenazine-mustard (FPN-M) and trans-flupenthixol-mustard (FPT-M) possessed alkylating activity, as assayed using 4-(p-nitrobenzyl) pyridine. Multidrug-resistant MCF-7/AdrR cells were incubated with FPN or FPN-M, or FPT or FPT-M for 1 h, washed for varying number of times in phosphate-buffered saline (PBS), then resuspended in medium containing [3H]vinblastine (VBL), and assayed for steady-state accumulation of the drug. Washing had far less of an effect on the ability of FPN-M and FPT-M to increase VBL accumulation compared to their parent compounds. After eight washes in excess PBS, the cells initially exposed to FPN or FPT accumulated only 30% and 50% of the initially accumulated drug, whereas the FPN-M- or FPT-M-treated cells accumulated approximately 75% and 90% of the control, respectively. FPN-M and FPT-M also increased the uptake and decreased the efflux of VBL from MDR cells despite repeated washing. We also examined the effects of these modulators on sensitivity of MDR cells to cytotoxic agents. FPN-M and FPT-M sensitized MCF-7/AdrR cells to VBL and doxorubicin to a greater extent than their parent compounds. These studies point out the potential of "irreversible" P-gp modulators to produce prolonged chemosensitization.  相似文献

In 2 experiments, young and older adults demonstrated modality effects of similar magnitude in perceptual identification tasks. That is, both young and older adults demonstrated more repetition priming when study and test modalities matched than when they were different, suggesting that contextual information was equally available across age. However, when asked explicitly to retrieve modality information, older adults were less accurate than young adults. These results constitute evidence for a dissociation between direct and indirect measures of memory for modality information. They call into question hypotheses that memory impairment in old age is due to deficient encoding of contextual information and challenge current accounts of modality effects in repetition priming. Spectroscopic, calorimetric, DNA cleavage, electrophoretic, and computer modeling techniques have been employed to characterize the DNA binding and topoisomerase poisoning properties of three protoberberine analogs, 8-desmethylcoralyne (DMC), 5,6-dihydro-8-desmethylcoralyne (DHDMC), and palmatine, which differ in the chemical structures of their B- and/or D-rings. DNA topoisomerase-mediated cleavage assays revealed that these compounds were unable to poison mammalian type II topoisomerase. By contrast, the three protoberberine analogs poisoned human topoisomerase I according to the following hierarchy: DHDMC > DMC > palmatine. DNA binding by all three protoberberine analogs induced negative flow linear dichroism signals as well as unwinding of the host duplex. These two observations are consistent with an intercalative mode of protoberberine binding to duplex DNA. However, a comparison of the DNA binding properties for DMC and DHDMC, which differ only by the state of saturation at the 5,6 positions of the B-ring, revealed that the protoberberine analogs do not "behave" like classic DNA intercalators. Specifically, saturation of the 5-6 double bond in the B-ring of DMC, thereby converting it to the DHDMC molecule, was associated with enhanced DNA unwinding as well as a reversal of DNA binding preference from a DNA duplex with an inaccessible or occluded minor groove {poly[d(G-C)]2} to DNA duplexes with accessible or unobstructed minor grooves {poly[d(A-T)]2 and poly[d(I-C)]2}. In addition, a comparison of the DNA binding properties for DHDMC and palmatine revealed that transferring the 11-methoxy moiety on the D-ring of DHDMC to the 9 position, thereby converting it to palmatine, was associated with a reduction in binding affinity for both duplexes with unobstructed minor grooves as well as for duplexes with occluded minor grooves. These DNA binding properties are consistent with a "mixed-mode" DNA binding model for protoberberines in which a portion of the ligand molecule intercalates into the double helix, while the nonintercalated portion of the ligand molecule protrudes into the minor groove of the host duplex, where it is thereby available for interactions with atoms lining the floor and/or walls of the minor groove. Furthermore, saturation at the 5,6 positions of the B-ring, which causes the A-ring to be tilted relative to the plane formed by rings C and D, appears to stabilize the interaction between the host duplex and the minor groove-directed portion of the protoberberine ligand. Computer modeling studies on the DHDMC-poly[d(A-T)]2 complex suggest that this interaction may involve van der Waals contacts between the ligand A-ring and backbone sugar atoms lining the minor groove of the host duplex. The hierarchy of topoisomerase I poisoning noted above suggests that this minor groove-directed interaction may play an important role in topoisomerase I poisoning by protoberberine analogs. In the aggregate, our results presented here, coupled with the recent demonstration of topoisomerase I poisoning by minor groove-binding terbenzimidazoles [Sun, Q., Gatto, B., Yu, C., Liu, A. , Liu, L. F., & LaVoie, E. J. (1995) J. Med. Chem. 38, 3638-3644], suggest that minor groove-directed ligand-DNA interactions may be of general importance in the poisoning of topoisomerase I.  相似文献

Examined the effects of facial attractiveness and physique on sex-role identity and constancy in 74 middle-class 3-9 yr olds. Measures included Ss' sex-role orientation, sex-role preference, sex-role adoption, parent salience, gender recognition, and sex-role constancy. Ratings for facial attractiveness and the ponderal index for body type were completed for each S. Analysis of variance using age, sex, and facial attractiveness as factors revealed no significant main effects for facial attractiveness on any of the measures. Interactions did appear, however, for Sex * Attractiveness * Age on sex-role adoption and for Facial Attractiveness * Sex on gender recognition. Analyses of variance for physique effects resulted in significant interactions of Physique Type * Age on both gender recognition and sex-role preference. Sex differences were present for sex-role orientation, preference, adoption, and parent salience, but not for gender recognition or sex-role constancy. Age differences appeared for sex-role orientation, preference, adoption, constancy, and gender recognition. Overall results fail to support the hypothesis that physical attractiveness and body stereotypes significantly influence sex-role development. It is concluded that these attributes may influence a child's perception of others, but they do not necessarily affect the child's gender assignment of others. N. R. Brown and R. S. Siegler (1996) found that training participants on a subset of country populations improved estimations for novel transfer country populations, an effect called seeding that remained intact over time. They attributed this effect to the abstraction by participants of a general metric framework for estimating populations not dependent on specific country anchors. In a series of 3 follow-up experiments, the authors found that training on seed populations produces both general metric information and durable specific country information. Moreover, minimal amounts of general (mean or range of populations) or specific (1 or 3 countries) information made available for inspection while estimating produced a significant seeding effect. Retention over long intervals was facilitated by both presenting 3 seed countries as opposed to 1 and providing names for the seed countries.
