Stimulus control of ring swimming was studied with male Siamese fighting fish (Betta splendens) using 2-component multiple schedules in which the components were correlated with the presence or absence of air bubbles in the water. In Experiment 1, either response-independent mirror presentations or extinction was juxtaposed with immediate response-dependent mirror presentations. Rates of ring swimming generally were higher with immediate reinforcement than with either response-independent mirror presentations or extinction. In Experiment 2, different durations of response-dependent mirror presentations were juxtaposed. Generally, higher rates of ring swimming occurred with 15-s than with 0-, 1-, or 3-s durations. Results demonstrate that stimulus control of responding can be established with these fish under several conditions of differential reinforcement. An issue of increasing interest in Pavlovian conditioning is to identify ways to facilitate the development and persistence of extinction. Both behavioral and molecular lines of evidence demonstrate that learning during extinction can be enhanced. Similar evidence has been offered to support the idea that extinction causes the original association to be unlearned, or erased. Differentiating between extinction and erasure accounts is extremely difficult and requires many assumptions about the fundamental nature of how memory storage maps into memory expression. In this issue of Behavioral Neuroscience, Norrholm et al. (see record 2008-13280-002) describe a study of extinction with humans that has the potential to serve as a translational bridge between rodent work and clinical applications. They find less recovery of a conditioned fear response when extinction occurs 10 min compared with 72 hr after conditioning; however, the recovery of subjects' expectancies of the fearful stimulus is independent of when extinction occurred. These findings and others discussed in this article demonstrate some of the challenges in making inferences about memory erasure during extinction. Four experiments with C57BL/6 mice investigated extinction of a spatial preference in the Morris water maze. In Experiment 1, a spatial preference was extinguished by exposing mice to the water maze in the absence of a platform but in the presence of the distal spatial cues. In Experiment 2, extinction occurred when the platform was removed from the pool, when it was presented in random locations, or when it was presented consistently in the opposite location. Contextual renewal (Experiment 3) and spontaneous recovery (Experiment 4) of spatial preferences argue against an interpretation of extinction in terms of unlearning and instead suggest that extinction in the water maze, like extinction in Pavlovian conditioning, suppresses the original association. Studies of contextual fear conditioning have found that ethanol administered prior to a conditioning session impairs the conditioned freezing response during a test session the next day. The present experiments examined the effects of ethanol on extinction, the loss of conditioned responding that occurs as the animal learns that a previously conditioned context no longer signals shock. Ethanol (1.5 g/kg) administered prior to single (Experiment 1) or multiple (Experiment 2) extinction sessions impaired extinction. Ethanol administered prior to a test session disrupted the expression of freezing after extinction (Experiments 3-5). There was some evidence that ethanol served as an internal stimulus signaling the operation of conditioning or extinction contingencies (Experiments 4-5). In Experiment 6, postsession injections of 1.5 g/kg ethanol had no effect on extinction with brief (3 min) or long (24 min) exposures to the context, but injections of 3 g/kg after long exposures impaired extinction. Together, these results indicate that ethanol affects extinction by acting on multiple learning and performance processes, including attention, memory encoding, and memory expression. Five experiments with C57BL/6 mice (Mus musculus) investigated whether failures in shock processing might contribute to deficits in freezing that occur after an animal receives a shock immediately on exposure to a conditioning context. Experiment 1 found that more contextual freezing resulted from delayed shocks than from immediate shocks across 4 shock intensities. Experiment 2 extended the immediate-shock freezing deficit to discrete stimuli. Experiment 3 found that preexposure to the to-be-conditioned cue did not facilitate immediate cued conditioning. Experiment 4 found that context preexposure enhanced context-evoked fear after an immediate shock. Experiment 5 found that context preexposure also enhanced immediate cued conditioning. These findings are problematic for current theories of the immediate-shock freezing deficit that focus exclusively on processing of the conditioned stimulus, and they suggest that failures in shock processing may contribute to the deficit. In experiments examining the potential reconsolidation of drug-associated contextual memories, rats were given a single pairing of cocaine with a specific context, and the ability of the protein synthesis inhibitor anisomycin administered following a context-only memory retrieval trial to impair conditioned locomotor sensitization was tested. Rats receiving 150 mg/kg anisomycin immediately following a 5-min reexposure to the cocaine-conditioned context showed decreased activity compared with the vehicle control group in response to a low-dose cocaine challenge during a subsequent test for conditioned sensitization. This effect was not seen when anisomycin was administered following a 30-min reexposure to the context or when anisomycin was administered 25 min after a 5-min reexposure. These results are consistent with a growing literature suggesting that following retrieval, associative contextual memories may undergo a transient protein synthesis-dependent reconsolidation phase that normally serves to maintain memory. The metaphor of the behavior stream provides a framework for studying the effects of response-independent food presentations intruded into an environment in which operant responding of pigeons was maintained by variable-interval schedules. In the first two experiments, response rates were reduced when response-independent food was intruded during the variable-interval schedule according to a concomitantly present fixed-time schedule. These reductions were not always an orderly function of the percentage of response-dependent food. Negatively accelerated patterns of key pecking across the fixed-time period occurred in Experiment 1 under the concomitant fixed-time variable-interval schedules. In Experiment 2, positively and negatively accelerated and linear response patterns occurred even though the schedules were similar to those used in Experiment 1. The variable findings in the first two experiments led to three subsequent experiments that were designed to further illuminate the controlling variables of the effects of intruded response-independent events. When the fixed and variable schedules were correlated with distinct operanda by employing a concurrent fixed-interval variable-interval schedule (Experiment 3) or with distinct discriminative stimuli (Experiments 4 and 5), negatively accelerated response patterns were obtained. Even in these latter cases, however, the response patterns were a joint function of the physical separation of the two schedules and the ratio of fixed-time or fixed-interval to variable-interval schedule food presentations. The results of the five experiments are discussed in terms of the contributions of both reinforcement variables and discriminative stimuli in determining the effects of intruding response-independent food into a stream of operant behavior.  相似文献

Four pigeons responded on a concurrent-chains schedule in four experiments that examined whether the effectiveness of a stimulus as a conditioned reinforcer is best described by a global approach, as measured by the average interreinforcement interval, or by a local contextual approach, as measured by the onset of the stimulus preceding the conditioned reinforcer. The interreinforcement interval was manipulated by the inclusion of an intertrial interval, which increased the overall time to reinforcement but did not change the local contingencies on a given trial A global analysis predicted choice for the richer alternative to decrease with the inclusion of an intertrial interval, whereas a local analysis predicted no change in preference. Experiment 1 examined sensitivity to intertrial intervals when each was signaled by the same houselight that operated throughout the session. In Experiment 2, the intertrial interval always was signaled by the stimulus correlated with the richer terminal link. In Experiment 3, the intertrial interval was signaled by the keylights correlated with the initial links and two novel houselights. Experiment 4 provided free food pseudorandomly during the intertrial interval. In all experiments, subjects' preferences were consistent with a local analysis of choice in concurrent chains. These results are discussed in terms of delay-reduction theory, which traditionally has failed to distinguish global and local contexts.  相似文献

Four experiments assessed the effects of trial (T) and intertrial (I) durations on magazine approach behavior in rats. In Experiments 1 and 2, different groups of animals were conditioned with various combinations of I and T durations. The rate of acquisition, in terms of the number of trials required to reach various acquisition criteria, generally was faster in groups trained with large I:T ratios. There also were differences in rate of acquisition and terminal response rates between groups trained with identical I:T ratios but with different absolute I and T durations. Differences evident at the end of conditioning persisted during a common test with various combinations of I and T durations. Experiments 3 and 4 provided a more specific test of the predictions of 2 general classes of theories and found results that were consistent with those theories that characterize group differences as indicative of differences in learning, rather than in performance. Several recent studies have shown that chromatin, the DNA-protein complex that packages genomic DNA, has an important function in learning and memory. Dynamic chromatin modification via histone deacetylase (HDAC) inhibitors and histone acetyltransferases may enhance hippocampal synaptic plasticity and hippocampus-dependent memory. Little is known about the effects of HDAC inhibitors on extinction, a learning process through which the ability of a previously conditioned stimulus, such as a conditioning context, to evoke a conditioned response is diminished. The authors demonstrate that administration of the HDAC inhibitors sodium butyrate (NaB) systemically or trichostatin A (TSA) intrahippocampally prior to a brief (3-min) contextual extinction session causes context-evoked fear to decrease to levels observed with a long (24-min) extinction session. These results suggest that HDAC inhibitors may enhance learning during extinction and are consistent with other studies demonstrating a role for the hippocampus in contextual extinction.
