The gangliosides of melanoma and other tumours of neuroectodermal origin are suitable targets for immune intervention with tumour vaccines. The optimal vaccines in current use contain ganglioside plus bacillus Calmette-Guérin and induce considerable morbidity. We have screened a variety of new adjuvants in the mouse, and describe one antigen-delivery system, proteosomes, which is especially effective. Highly hydrophobic Neisserial outer membrane proteins (OMP) form multimolecular liposome-like vesicular structures termed proteosomes which can readily incorporate amphiphilic molecules such as GD3 ganglioside. The optimal GD3/proteosome vaccine formulation for induction of GD3 antibodies in the mouse is determined. Interestingly, the use of potent immunological adjuvants in addition to proteosomes augments the IgM and IgG antibody titres against OMP in these vaccines but GD3 antibody titres are unaffected. The application of proteosomes to enhance the immune response to GD3 extends the concept of the proteosome immunopotentiating system from lipopeptides to amphipathic carbohydrate epitopes such as cell-surface gangliosides. The demonstrated safety of meningococcal OMP in humans and the data in mice presented here suggest that proteosome vaccines have potential for augmenting the immunogenicity of amphipathic tumour antigens in humans.  相似文献

One hundred and twelve bilateral thyroidectomies for solitary thyroid nodules with suspected malignancy were performed. The incidence of malignancy in the 112 primary nodules was 42%. Twenty-nine of the 112 contralateral lobes (26%) contained malignancy, which was unsuspected in 80%. Of these 29, 20 were foci of papillary cancer < or = 5 mm and 9 were larger papillary tumors or follicular carcinoma. We analyzed these 112 patients to determine whether there was a simple method to identify those patients at risk for contralateral, unsuspected malignancy. With use of the AMES clinical staging retrospectively, 70 of the 112 patients were classified as having low-stage disease. Fifty percent (35) had cancers on the primary side and 27% (19) on the contralateral side. Of these 19 contralateral cancers, 14 were papillary cancers < or = 5 mm, 4 were papillary cancer > 5 mm and one was a 1.5 cm follicular carcinoma, a similar distribution as in the whole group of 112. When the AMES analysis then excluded those thought to be at risk for multicentricity or papillary carcinoma and examined female patients only with nonpapillary frozen sections, nonpapillary aspiration cytological results, and no history of radiation exposure, no further reduction in the proportion of contralateral cancers (7 of 26, 27%) was found. Fifty-five of the 112 patients underwent preoperative ultrasound scans. In those cases in whom the contralateral lobe had no intraoperative palpable or preoperative sonographic mass, 5 of 20 still had contralateral cancers, but all were papillary < or = 5 mm.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The culture movement challenged the universality of the self-enhancement motive by proposing that the motive is pervasive in individualistic cultures (the West) but absent in collectivistic cultures (the East). The present research posited that Westerners and Easterners use different tactics to achieve the same goal: positive self-regard. Study 1 tested participants from differing cultural backgrounds (the United States vs. Japan), and Study 2 tested participants of differing self-construals (independent vs. interdependent). Americans and independents self-enhanced on individualistic attributes, whereas Japanese and interdependents self-enhanced on collectivistic attributes. Independents regarded individualistic attributes, whereas interdependents regarded collectivistic attributes, as personally important. Attribute importance mediated self-enhancement. Regardless of cultural background or self-construal, people self-enhance on personally important dimensions. Self-enhancement is a universal human motive. Previous studies of Pacific salmonid passage over Snake River dams indicated slowed passage at transition pools, the transition area between the fishway entrance and the fish ladder. In 2001 and 2002, we conducted an experiment to determine if modified weirs affected adult salmon and steelhead passage times and route selection through the Lower Granite Dam transition pool. Fish attraction flows through the lower ladder weirs were experimentally increased using removable panels. During the experiment we monitored radio‐tagged adult Chinook salmon and steelhead to determine passage routes and times through the transition pool. The weir treatment increased the number of spring–summer Chinook salmon passing straight through the transition pool compared to those exiting the transition pool to the collection channel or tailrace. Mean passage times through the transition pool differed among routes and were significantly lower during treatment periods for the exit‐to‐collection channel route in spring‐summer Chinook salmon, but not for other routes. Passage times among routes differed in steelhead, but there was no evidence of treatment effects on route use or passage time. Fall Chinook exhibited similar trends in route use and passage time to spring–summer Chinook, but differences were not significant, perhaps because of relatively small sample size. Total dam passage times did not differ by treatment or route for any run. Fish depth during passage of the transition pool suggested that most fish passed through submerged orifices and supported the hypothesis that increased water velocity through these orifices caused the increase in straight‐through passage in spring–summer Chinook. Collectively, the results suggested the weir modifications provided improvement to passage through the transition pool for spring–summer Chinook and no evidence of negative effects on other runs. The effect of thermal shock on the spalling of oxides from the surfaces of several commercial alloys was determined. The average cooling rate was varied from approximately 240°C/sec to less than 1.0°C/sec during cyclic oxidation tests in air. The tests consisted of 100 cycles of 1 hr at the maximimum temperature (1100 or 1200°C). The alloys were HOS-875, TD-Ni, TD-NiCrAl, In-601, In-702, and B-1900 plus Hf. All of these alloys exhibited partial spalling within the oxide rather than total oxide loss down to bare metal. Thermal shock resulted in deformation of the metal, which in turn resulted, in most cases, in change of the oxide failure mode from compressive to tensile. Tensile failures were characterized by cracking of the oxide and little loss, while compressive failures were characterized by explosive loss of platelets of oxide. This behavior was confirmed by examination of mechanically stressed oxide scales. The thermally shocked oxides spalled less than the slow-cooled samples with the exception of TD-NiCrAl. This material failed in a brittle manner rather than by plastic deformation. The HOS-875 and the TD-Ni did not spall during either type of cooling. Thus, the effect of thermal shock on spalling is determined, in large part, by the mechanical properties of the metal.  相似文献

The property changes of Epon 828 crosslinked by either diethylenetriamine or m-phenylenediamine at several temperatures have been studied by two in situ techniques. The mechanical property changes were followed by continuously monitoring the changes in the fundamental frequency of vibration during cure of a resin-coated substrate acting as a cantilever reed. The disappearance of the reactive epoxy group was followed in situ by continuously monitoring the absorbance of the infrared epoxide band at 915 cm.?1. Correlation of the two techniques was accomplished by normalizing both the mechanical property and infrared data. The apparent energy of activation was calculated to be 10.4 and 11.4 kcal. /mole, respectively, for the m-phenylenediamine and diethylenetriamine systems by the mechanical technique, and 11.0 kcal./mole for the m-phenylenediamine crosslinked system by the infrared technique.  相似文献

Cyclic and isothermal oxidation behavior on some Ni-Cr alloys   总被引:1,自引:0,他引:1  
Additions of 3 wt.% Mn and 3 wt.% Si were made to Ni-20Cr. These alloys, along with Ni-20Cr and Ni-40Cr were oxidized for 100 1-hr cycles at 1100°C and 50 1-hr cycles at 1200° C. Oxidation behavior was judged by sample weight and thickness change, metallography, x-ray diffraction, and electron microprobe analysis. These tests showed that Ni-40Cr and Ni-20Cr-3Si were about the same and were the most oxidation-resistant alloys. Ni-20Cr-3Mn was not as oxidation resistant, especially at 1200° C. Ni-20Cr was far less oxidation resistant than any of the other alloys. The Ni-40Cr and Ni-20Cr-3Si relied on a protective layer of Cr2O3 for their oxidation resistance. A SiO2 layer was noted beneath the Cr2O3 layer on the Ni-20Cr-3Si, but had apparently only a second-order effect. The source of improved protection of the Ni-20Cr-3Mn was apparently the formation of a relatively adherent MnCr2O4 layer at the metal-oxide interface.  相似文献

We report a first combination of diamond anvil cell radial x-ray diffraction with in situ laser heating. The laser-heating setup of ALS beamline 12.2.2 was modified to allow one-sided heating of a sample in a diamond anvil cell with an 80 W yttrium lithium fluoride laser while probing the sample with radial x-ray diffraction. The diamond anvil cell is placed with its compressional axis vertical, and perpendicular to the beam. The laser beam is focused onto the sample from the top while the sample is probed with hard x-rays through an x-ray transparent boron-epoxy gasket. The temperature response of preferred orientation of (Fe,Mg)O is probed as a test experiment. Recrystallization was observed above 1500 K, accompanied by a decrease in stress.  相似文献

In this paper, a hybrid algorithm consisting of particle swarm optimization and pattern search algorithm is proposed to evaluate and optimize the design and operation of microgrids (MGs) in combined gas and power networks. Key performance indicators (KPIs) are modeled and proposed to evaluate and assess MGs. The paper begins by proposing a comprehensive study to define KPIs, which are used to evaluate the following MG parameters: economical efficiency, reliability, environmental conservation, and power quality. Multi‐objective evaluation functions are then developed by building a relationship matrix of MG and KPI components. The results are then displayed as optimized power generation percentages for each technology with values for four KPI categories: cost, quality, reliability and environmental friendliness. Two case studies are examined in this paper; both the province of Ontario and Toronto regional zone under all system parameters with varying percentage of generation via gas technology. Results indicated that the optimal scenario for both Ontario and Toronto was achieved at hybrid PSO–patern search percentage generation via gas technology with improved cost KPI and other KPIs remaining approximately constant.
