Agrobacterium tumefaciens transfers part of its Ti plasmid, the T-DNA, to plant cells during tumorigenesis. It is routinely used for the genetic modification of a wide range of plant species. We report that A. tumefaciens can also transfer its T-DNA efficiently to the filamentous fungus Aspergillus awamori, demonstrating DNA transfer between a prokaryote and a filamentous fungus. We transformed both protoplasts and conidia with frequencies that were improved up to 600-fold as compared with conventional techniques for transformation of A. awamori protoplasts. The majority of the A. awamori transformants contained a single T-DNA copy randomly integrated at a chromosomal locus. The T-DNA integrated into the A. awamori genome in a manner similar to that described for plants. We also transformed a variety of other filamentous fungi, including Aspergillus niger, Fusarium venenatum, Trichoderma reesei, Colletotrichum gloeosporioides, Neurospora crassa, and the mushroom Agaricus bisporus, demonstrating that transformation using A. tumefaciens is generally applicable to filamentous fungi.  相似文献

OBJECTIVES: The three purposes of this study are to: (1) describe the relationship between the prevalence of coronal caries and root caries; (2) describe the relationship between the three-year incidence of coronal caries and root caries; and (3) if the two conditions are associated, develop a multiple regression model that identifies characteristics distinguishing people who had increments of both root caries and coronal caries from people who had increments of either coronal caries or root caries, or who had no new caries. METHODS: Dental examinations and interviews were conducted in the homes of a randomly selected, stratified sample of people over the age of 65 years in five North Carolina counties. The relationships between coronal and root D and DF were analyzed through contingency table analyses, and ordinal logistic regression was used to identify characteristics that differentiated people who had both coronal and root D over the three years from people who had either coronal or root D and people who had no new disease. RESULTS: Evidence of root and coronal caries in whites was much more likely to be in the form of fillings, while for blacks, it was more likely to be in the form of untreated decay. Prevalence rates of coronal and root D and DF were significantly associated for both blacks and whites. Incidence rates based on DF indicated that root and coronal caries were not associated in whites, but were associated in blacks. People more likely to experience both types of caries had more gingival recession at baseline, greater average attachment loss over the three years, and lactobacilli at baseline. In addition, the presence of Porphymonas gingivalis at three years was important for whites. CONCLUSIONS: It appears that coronal and root caries do tend to appear together in the same individuals, but fillings attenuate that relationship. The impact of dental treatment on the epidemiology of dental caries appears to be considerable and calls into question whether the F component of the caries index is related to disease as defined by epidemiologic criteria.  相似文献

We cloned the murine full-length cDNA encoding Ahch, the mouse homologue of DAX1 (DSS-AHC Region on Human X Chromosome, Gene1) which is the gene responsible for human X-linked adrenal hypoplasia congenita (AHC) and hypogonadotropic hypogonadism (HH). Sequence analysis revealed that the murine and human cDNAs have 65% aa identity and 75% aa similarity overall. The cysteine residues in the putative DNA binding domain, which may interact with Zn2+ ions to form zinc fingers, are 100% conserved between the two species, indicating that the novel zinc-finger structures in DAX1 may be functional. In addition, mouse interspecific backcrosses show that the Ahch gene is closely linked to the glycerol kinase locus, GyK, on the mouse X chromosome, indicating that the order of the loci is conserved in this syntenic region between mouse and human.  相似文献

A study of the impact of machine-paced (M/P) and self-paced (S/P) work on job satisfaction of 28 female industrial assembly workers was evaluated in which M/P work was confounded with simplified work and the S/P job was confounded with enriched tasks. Results indicated the following:

1. 1. Over three-quarters of workers were more satisfied in S/P jobs, while only less than one-quarter were more satisfied in M/P jobs.

2. 2. The 16PF personality test effectively predicts (0·88 multiple correlation) the satisfaction ratios of M/P to S/P jobs.

The authors tested the acculturation gap-distress hypothesis by examining whether parent-adolescent acculturation gaps were associated with greater conflict and youth conduct problems among 260 high-risk Mexican American families. The authors operationalized acculturation gaps in 2 ways: parent-youth mismatches in acculturation style, and parent-youth discrepancies in acculturation toward both mainstream and heritage cultures. Acculturation gaps were common, but results of hierarchical regression analyses indicated that parent-youth discrepancies in acculturation toward mainstream and heritage cultures were not related to increased conflict or youth conduct problems. Conduct problems were no higher in families in which the adolescent was more aligned with mainstream culture than the parent. Unexpectedly, the authors found more youth conduct problems in families in which the youth was more aligned with traditional culture than the parent. The results call into question the assumption that the more rapid acculturation of adolescents to American culture inevitably leads to distress in minority families. This review provides an evaluation of the correlates and/or risk factors associated with disordered eating and the pursuit of muscularity among adolescent boys. One of the main conclusions is that similar factors and processes are associated with both behavioral problems. Several factors found to be consistently associated with disordered eating among boys are also similar to those found with girls. These include body mass index, negative affect, self-esteem, perfectionism, drug use, perceived pressure to lose weight from parents and peers, and participation in sports that focus on leanness. A chromatographic adsorption unit was designed and built to study the adsorption of alkanes in zeolites for the hydrocarbon trap systems in three‐way catalysts, to assist in the selection of optimal materials for this application. The experimental apparatus used a zeolite bed in place of the column in an ordinary gas chromatograph and could be accurately modeled to determine the adsorption equilibrium constants for simple alkanes in MFI zeolites. The adsorption of iso‐pentane was studied in BEA zeolites with varying Si/Al2 ratios, before and after ex situ zeolite aging simulating engine exhaust, and in the presence of water vapor. The elution times were shown to depend directly on the zeolite adsorption capacity. The primary effect of water was to decrease the iso‐pentane adsorption capacity by partial filling of the zeolite pores through adsorption of water at acid sites. Laser sources offer a possible solution to the problem of low light throughput in direct-view microscopes (DVMs). However, coherent source DVMs have been shown to suffer from problems such as increased sidelobes in the depth response because of coherent cross talk between neighboring apertures. We explore theoretically how source coherence affects the depth responses of DVMs by employing various aperture spacings and number of apertures. We show that, contrary to expectation, closely spaced apertures can result in decreased full width at half-maximum of the depth response curve. We explain this as an effect of destructive interference when cross talk between neighboring apertures occurs. Using apertures arranged in a square grid as an example, we move on to show that the use of aperture arrays that consist of regularly arranged apertures can accentuate the problematic sidelobes of the depth response. We show that arranging pinholes in a rectangular grid rather than a square grid can improve the optical sectioning strength significantly. Finally, by examination of the depth responses corresponding to the infinite-pinhole-array limit, we make some general statements about source coherence and the characteristics of arrays that are likely to perform well.  相似文献

We consider how to interpret, filter, and cross-correlate complex-value color (hue and saturation) images by using a single discrete Fourier transform: the spatiochromatic discrete Fourier transform. The model defines new types of spatiochromatic oriented sinusoidal gratings, termed rainbow gratings, which encode the variation of color over space. We demonstrate how color-opponent detectors observed within the vertebrate visual system can be easily defined by linear filters within this representation. This model also allows us to filter and detect both spatial and chromatic patterns in images by using a single cross-correlation procedure. In doing so, we explore a new form of the Cauchy-Schwartz inequality applied to complex-valued scalar products. Results demonstrate the power of this form of spatiochromatic matched filtering in detecting signals embedded in such a significant amount of noise that they are not visible to the unaided human eye.  相似文献
