The authors studied the role of gamma-aminobutyric acid (GABA) in 2 types of forgetting of fear in the developing rat. One type of forgetting studied was that observed after an intermediate retention interval (the "Kamin effect"); the other type studied was that observed after a longer interval (infantile amnesia). Rats were given pairings of an auditory conditioned stimulus with shock, and learned fear was assessed by freezing. Forgetting at an intermediate retention interval (1 hr) was not alleviated by the GABAA receptor partial inverse agonist FG7142 (0, 1, 5, or 10 mg/kg), whereas forgetting at a longer retention interval (48 hr) was alleviated. These results suggest that in the developing rat, forgetting observed at different retention intervals is mediated by different physiological mechanisms. Rats received a single pairing of an auditory conditioned stimulus (CS) with a footshock unconditioned stimulus (US). The fear (freezing) that had accrued to the CS was then extinguished. Injection of naloxone prior to this extinction significantly impaired the development of extinction. This impairment was mediated by opioid receptors in the brain and was not observed when naloxone was injected after extinction training. Finally, an injection of naloxone on test failed to reinstate extinguished responding that had already accrued to the CS. These experiments show that opioid receptors regulate the development, but not the expression, of fear extinction and are discussed with reference to the roles of opioid receptors in US processing, memory, and appetitive motivation. The use of two generalised carrier transport models to account for the ND−1 dependence of the specific contact resistance (ρc) of metal-semiconductor Ohmic contacts to n-type GaAs is proposed. Both models include the effects of thermionic emission and diffusion across the high-low barrier junction a priori. Calculations of ρc, and comparison with experimental data, show conclusively that thermionic emission is the dominant transport mechanism across the barrier. It is stressed that these models do not rely on prior choices of either of the transport processes. These conclusions are arrived at a posteriori.  相似文献

Autonomic dysfunction in insulin-dependent diabetic (IDDM) patients has been associated with abnormalities of left ventricular function and an increased risk of sudden death. A group of 30 patients with IDDM and 30 age, sex and blood pressure matched control subjects underwent traditional tests of autonomic function. In addition, baroreceptor-cardiac reflex sensitivity (BRS) was assessed using time domain (sequence) analysis of systolic blood pressure and pulse interval data recorded non-invasively using the Finapres beat-to-beat blood pressure recording system. 'Up BRS' sequences-increases in systolic blood pressure associated with lengthening of R-R interval, and 'down BRS' sequences-decreases in systolic blood pressure associated with shortening of R-R interval were identified and BRS calculated from the regression of systolic blood pressure on R-R interval for all sequences. We also assessed heart rate variability using power spectral analysis and, after expressing components of the spectrum in normalised units, assessed sympathovagal balance from the ratio of low to high frequency powers. IDDM subjects underwent 2-D echocardiography to assess left ventricular mass index. Standard tests of autonomic function revealed no differences between IDDM patients and control subjects, but dramatic reductions in baroreceptor-cardiac reflex sensitivity were detected in IDDM patients. 'Up BRS' when supine was 11.2 +/- 1.5 ms/mmHg (mean +/- SEM) compared with 20.4 +/- 1.95 in control subjects (p < 0.003) and when standing was 4.1 +/- 1.9 vs 7.6 +/- 2.7 ms/mmHg (p < 0.001). Down BRS when supine was 11.5 +/- 1.2 vs 22 +/- 2.6 (p < 0.001) and standing was 4.4 +/- 1.9 vs 7.3 +/- 2.5 ms/mmHg (p < 0.003). There were significant relations between impairment of the baroreflex and duration of diabetes (p < 0.001) and poor glycaemic control (p < 0.001). From a fast Fourier transformation of supine heart rate data and using a band width of 0.05-0.15 Hz as low-frequency and 0.2-0.35 Hz as high frequency total spectral power of R-R interval variability was significantly reduced in the IDDM group for both low-frequency (473 +/- 62.8 vs 746.6 +/- 77.6 ms2 p = 0.002) and high frequency bands 125.2 +/- 12.9 vs 459.3 +/- 89.8 ms2 p < 0.0001. When the absolute powers were expressed in normalised units the ratio of low frequency to high frequency power (a measure of sympathovagal balance) was significantly increased in the IDDM group (2.9 +/- 0.53 vs 4.6 +/- 0.55, p < 0.002 supine: 3.8 +/- 0.49 vs 6.6 +/- 0.55, p < 0.001 standing). Thus, time domain analysis of baroreceptor-cardiac reflex sensitivity detects autonomic dysfunction more frequently in IDDM patients than conventional tests. Impaired BRS is associated with an increased left ventricular mass index and this abnormality may have a role in the increased incidence of sudden death seen in young IDDM patients.  相似文献

The relative risk of death by calendar date of diagnosis was investigated in a population-based incident cohort of 845 (463 males:382 females) IDDM diagnosed in Leicestershire before the age of 17 years between 1940 and 1989. The mortality status of 844 (99.9%) patients was determined as of the 31 December 1991, representing 14,346 person-years of risk. Trends in relative risk of death were investigated using Cox proportional hazards modelling for within cohort comparisons and age/sex and calendar time adjusted standardized mortality ratios (SMR) using generalized linear modelling for external comparisons. Median age at diagnosis was 10 years (range 3 months to 16 years); median duration of diabetes 15 years (range 1-51 years). Forty-four patients had died (5.2%; median age at death 31 years, range 11-51 years). A further four patients died at presentation (within 24 h) from ketoacidosis and are excluded from all analyses. Calendar date of diagnosis was found to be an important predictor of mortality. Adjusting for attained age there was evidence of a decline in relative risk of death with calendar date of diagnosis of 3.4% (95% CI, 0.005-6.9%) per annum, equivalent to a 32% fall per decade (95% CI, 5-51%), or 84% (95% CI, 21-97) from 1940 to 1989. The data are consistent with a large fall in mortality between the 1940s and 1950s representing over 50% of the total reduction in mortality between 1940 and 1991. Neither sex nor age at diagnosis were significant predictors of mortality. Over the study period 1940-89 the SMR (male and female combined) fell from 981 (541-1556) to 238 (60-953) relative to the general population. This population-based study shows that the prognosis for Type 1 (insulin-dependent) diabetes mellitus has improved markedly over the period 1940-1991.  相似文献

A range of powdered ethyl‐vinyl acetate (EVA) copolymers and poly(vinyl chloride) (PVC) formulations were compounded at PVC:EVA ratios of 100:0, 60:40, 50:50, 40:60, and 0:100. Two grades of EVA with 20% and 27% of vinyl acetate (VAc) (EVA I and EVA II) and two grades of PVC with K values of 56 and 71 (PVC I and PVC II) were used in the investigation. Mechanical analysis was performed on injection molded samples of these blends, and the results showed that the tensile and flexural moduli decreased significantly with increasing EVA concentration. Rheological analysis was performed by using dual capillary rheometry, and the results showed only slight changes in shear viscosity with increasing EVA content, even at lower shear rates. Dynamic mechanical thermal analysis showed partial miscib lity of the PVC and EVA over the range of concentrations studied.  相似文献

Immiscible blends of ethylene-propylene-diene-monomer (EPDM) and polyoxymethylene (POM), when EPDM is the major phase were compatibilized on the addition of an ionomer, poly(ethylene-co-methacrylic acid). The inclusion of the ionomer reduced the interfacial tension between the two phases, such that the diameter of the POM domains were significantly reduced to between 0.5 and 2 μm, typical of that required to toughen ductile polymers. The mechanical properties of the resultant compatibilized blends were significantly enhanced with increases in Young's modulus (↑54%), tensile strength (σ, ↑139%), elongation at break (ε, ↑97%), and tensile toughness (↑500%) with increasing ionomer content, relative to EPDM rubber alone. The ShoreA hardness of the compatibilized blend was 70.1 compared with 56.8 for the immiscible binary blend and, 50.2 for neat EPDM rubber.  相似文献

In this study a MADYMO (mathematical dynamic modelling) model has been used to identify the influence of leg fracture on the injuries sustained by the pedestrian during front end impact with a vehicle. A factorial study of a MADYMO pedestrian and vehicle model are used to investigate the effect of different leg fracture tolerances, geometry, and vehicle compliance on the criteria measured in the European Enhanced Vehicle-safety Committee (EEVC) pedestrian safety tests. These criteria include knee bending, knee shear response, and lower leg bone (tibia) acceleration. The main study examines the spread of typical values of lower limb tolerance based on reported literature and contrasts the response of weaker, low-strength bones, normal tolerance, and limbs which do not fracture. Results show that knee bending angles and therefore ligament strains are significantly increased when fracture does not occur, and are decreased in bones exhibiting a low-strength response. Bone fracture tolerance is shown to be a significant parameter influencing knee bending. The parameters are compared to show that knee shear is significantly influenced by vehicle bumper compliance and that both criteria are heavily influenced by bumper height. Vehicles with more aggressive geometry, higher bumpers, and larger bumper lead were considered for comparative purposes.  相似文献

The relative incidence of accidental injury for Navy enlisted men during 1974-1977 was analyzed for 68 occupations, three pay grade or job responsibility levels, and duty status (on or off duty) at the time of the injury. Thirteen "blue-collar", predominantly sea-going or construction occupations, were significantly higher than the Navy injury rate norm and 26 "white-collar" occupations, typically in clerical, supply, administrative or technical specialties, were significantly lower than the Navy norm. An inverse linear relationship was found between injury rate and job responsibility for all occupations combined. This relationship varied by occupation, however, suggesting that experience and familiarity with the work environment may not moderate risks to the same degree in all occupational fields. Occupations with high on-duty injury rates tended to have high off-duty rates as well, suggesting at least some common causal factors.  相似文献

The authors used rats to study the impact of a history of opiate exposures on behavioral and autonomic responses to restraint stress. Brief restraint (30 min) provoked tachycardia and a pressor response, anxiety (as indexed by social interaction), grooming, and reduced exploration. The pressor response was reduced at 1 day, but not 7 days, after last opiate exposure; tachycardia was unaffected (Experiment 1). Stress-induced anxiety was potentiated 1 and 7 days after last opiate exposure (Experiment 2), and this potentiation was a function of dose (Experiment 3) and duration (Experiment 4) of opiate exposure. The results show that a history of opiate exposures alters vulnerability to stress and has implications for understanding coping, anxiety, and emotionality in former opiate users.
