The damaging effects of ionizing radiation (IR) on bone mass are well-documented in mice and humans and are most likely due to increased osteoclast number and function. However, the mechanisms leading to inappropriate increases in osteoclastic bone resorption are only partially understood. Here, we show that exposure to multiple fractions of low-doses (10 fractions of 0.4 Gy total body irradiation [TBI]/week, i.e., fractionated exposure) and/or a single exposure to the same total dose of 4 Gy TBI causes a decrease in trabecular, but not cortical, bone mass in young adult male mice. This damaging effect was associated with highly activated bone resorption. Both osteoclast differentiation and maturation increased in cultures of bone marrow-derived macrophages from mice exposed to either fractionated or singular TBI. IR also increased the expression and enzymatic activity of mitochondrial deacetylase Sirtuin-3 (Sirt3)—an essential protein for osteoclast mitochondrial activity and bone resorption in the development of osteoporosis. Osteoclast progenitors lacking Sirt3 exposed to IR exhibited impaired resorptive activity. Taken together, targeting impairment of osteoclast mitochondrial activity could be a novel therapeutic strategy for IR-induced bone loss, and Sirt3 is likely a major mediator of this effect. 相似文献

Internet video streaming is a widely popular application however, in many cases, congestion control facilities are not well
integrated into such applications. In order to be fair to other users that do not stream video, rate adaptation should be
performed to respond to congestion. On the other hand, the effect of rate adaptation on the viewer should be minimized and
this extra mechanism should not overload the client and the server. In this paper, we develop a heuristic approach for unicast
congestion control. The primary feature of our approach is the two level adaptation algorithm that utilizes packet loss rate
as well as receiver buffer data to maintain satisfactory buffer levels at the receiver. This is particularly important if
receiver has limited buffer such as in mobile devices. When there is no congestion, to maintain best buffer levels, fine grain
adjustments are carried out at the packet level. Depending on the level of congestion and receiver buffer level, rate shaping
that involves frame discard and finally rate adaptation by switching to a different pre-encoded video stream are carried out.
Additive increase multiplicative decrease policy is maintained to respond to congestion in a TCP- friendly manner. The algorithm
is implemented and performance results show that it has adaptation ability that is suitable for both local area and wide area
networks.

in St. Louis, U.S.A. in 1985. After his doctorate study, he joined Computer Engineering Department of Ege University as an
assistant professor where he became an associate professor in 1988. During the period of 1992–1994, he worked in Department
of Computer Technology of Nanyang Technological University of Singapore as a Visiting Senior Fellow. He then joined International
Computer Institute of Ege University as a Professor where he is currently the director. In the period of 2000–2001 he worked
in Department of Computer Science of Loyola University of Chicago as a Visiting Professor. His current research interests
include adaptive video streaming and Internet performance measurements. Dr. Tunali is married with an eighteen year old son.
Aylin Kantarci received B.Sc., M.Sc. and Ph.D. degrees all from Computer Engineering Department of Ege University, Izmir, Turkey, in 1992,
1994 and 2000, respectively. She then joined the same department as an assistant professor. Her current research interests
include adaptive video streaming, video coding, operating systems, multimedia systems and distributed systems.
Nukhet Ozbek received B.Sc. degree in Electrical and Electronics Engineering from School of Engineering and M.Sc. degree in Computer Science
from International Computer Institute both in Ege University, Izmir, Turkey. From 1998 to 2003 she worked in the DVB team
of Digital R&D at Vestel Corporation, Izmir-Turkey that produces telecommunication and consumer electronics devices. She is
currently a Ph.D. student and a research assistant at International Computer Institute of Ege University. Her research areas

One fundamental observation in cancer etiology is that the rate of malignancies in any mammalian population increases exponentially as a function of age, suggesting a mechanistic link between the cellular processes governing longevity and carcinogenesis. In addition, it is well established that aberrations in mitochondrial metabolism, as measured by increased reactive oxygen species (ROS), are observed in both aging and cancer. In this regard, genes that impact upon longevity have recently been characterized in S. cerevisiae and C. elegans, and the human homologs include the Sirtuin family of protein deacetylases. Interestingly, three of the seven sirtuin proteins are localized into the mitochondria suggesting a connection between the mitochondrial sirtuins, the free radical theory of aging, and carcinogenesis. Based on these results it has been hypothesized that Sirt3 functions as a mitochondrial fidelity protein whose function governs both aging and carcinogenesis by modulating ROS metabolism. Sirt3 has also now been identified as a genomically expressed, mitochondrial localized tumor suppressor and this review will outline potential relationships between mitochondrial ROS/superoxide levels, aging, and cell phenotypes permissive for estrogen and progesterone receptor positive breast carcinogenesis. 相似文献

In 2 experiments, 40 graduate and 60 undergraduate students wrote preliminary and final drafts of a persuasive document. In Exp I, the need for Ss to comply with mechanics (i.e., punctuation and spelling), to form complete sentences, and to sequence their ideas, respectively, were eliminated one at a time from preliminary-draft formats. The elimination of sentence-formation and sequence operations produced corresponding increments in persuasive argument production. In Exp II, Ss with average verbal ability (as assessed by the Scholastic Aptitude Test) benefited more from the elimination of these structure operations than did Ss with low verbal ability. When allowed to freely choose their typical preliminary-draft format, Ss with average verbal ability usually selected proposition-based formats (e.g., lists, outlines, and diagrams) that dispensed with the need for structure operations. Ss with average verbal ability also produced more sentences, more arguments per sentence, and fewer mechanical errors than did those with low verbal ability. Space is a high-stress environment. One major risk factor for the astronauts when they leave the Earth’s magnetic field is exposure to ionizing radiation from galactic cosmic rays (GCR). Several adverse changes occur in mammalian anatomy and physiology in space, including bone loss. In this study, we assessed the effects of simplified GCR exposure on skeletal health in vivo. Three months following exposure to 0.5 Gy total body simulated GCR, blood, bone marrow and tissue were collected from 9 months old male mice. The key findings from our cell and tissue analysis are (1) GCR induced femoral trabecular bone loss in adult mice but had no effect on spinal trabecular bone. (2) GCR increased circulating osteoclast differentiation markers and osteoclast formation but did not alter new bone formation or osteoblast differentiation. (3) Steady-state levels of mitochondrial reactive oxygen species, mitochondrial and non-mitochondrial respiration were increased without any changes in mitochondrial mass in pre-osteoclasts after GCR exposure. (4) Alterations in substrate utilization following GCR exposure in pre-osteoclasts suggested a metabolic rewiring of mitochondria. Taken together, targeting radiation-mediated mitochondrial metabolic reprogramming of osteoclasts could be speculated as a viable therapeutic strategy for space travel induced bone loss. 相似文献
