Antitubercular activity of lomefloxacin in an experiment

The effect of lomefloxacin was studied on mice with experimental infection due to Mycobacterium tuberculosis. The antibiotic was administered in doses of 100 and 200 mg/kg. It was shown that the use of lomefloxacin for a month provided a lower death rate of the animals with progressing acute generalized tuberculosis, a lower level of the lesions in the internal organs and a lower number of the Mycobacterium isolates from them. The efficacy of the treatment depended on the drug dose. When lomefloxacin was used in a dose of 200 mg/kg, the survival rate was much higher and the number of the epithelial unicellular granulomas in the tissue of the lung and spleen was markedly decreased while with the lower dose the indices did not differ from those in the control.     

Simulation of contact stresses and forces during hot rolling of thin wide strips with allowance for a stick zone and elastic regions in the deformation zone

A new procedure for the calculation of contact stresses and hot-rolling forces for wide strips 0.8–1.5 mm thick has been developed and tested. This procedure takes into account the presence of a stick zone in the deformation zone and stress distributions in both elastic and plastic regions in the deformation zone. The average error in the force calculation according to the new procedure is 5%, which is more than two times smaller than the calculation error of well-known force calculation procedures. The developed procedure is used to simulate the contact stresses in the deformation zones of working stands in a six-stand 1700 mill during rolling of strips thinner than 1.0 mm. A number of new relations for the state of stress in a strip have been revealed upon simulation. Some of these relations are as follows: in the last stands, the length of elastic regions accounts for 10–17% of the total deformation-zone length; the maximum normal contact stresses are 1300–1400 MPa, which corresponds to the stresses in the deformation zones of cold-rolling mills; the stick-zone length accounts for 85–99% of the deformation-zone length; and the contact stresses in the stick zone are virtually independent of the friction coefficient. The developed calculation procedure can be used to optimize the technological regimes of wide-strip mills.     

Human gamma-interferon expression in the mammary gland of transgenic mice

Transgenic mice carrying a hybrid gene consisting of ovine beta-lactoglobulin gene sequences and human gamma-interferon (hIFN-g) cDNA were produced. hIFN-g expression in the mammary gland of two lactating transgenic founder females was found. The concentration of active hIFN-g in the milk was estimated as being ca. 1800 IU/ml. The hIFN-g ability to express in the mammary gland was found in the progeny of transgenic founder male.     

State of stress in the deformation zone during rolling of high-strength plate steel

A new mathematical model is developed for the state of stress in the deformation zone for plate rolling, including rolling under controlled conditions at a low temperature in the last passes. The stick zone during plate rolling is shown to account for 88–99% of the deformation zone, and the fraction of elastic regions in the deformation zone in the last passes at low temperatures can reach 12%. When the stick zone and elastic regions of the deformation zone are taken into account, the roll-force calculation error is less than 5–6.5%. The model developed can be used to optimize plate-rolling conditions, including the conditions of integrated deformation-thermal production.     

Reproductive health of the family as a goal of industrial medicine

Modern demographic situation stressed the problems of human reproductive health associated with industrial medical topics. The article suggests to regard a family (child and two parents) as a unit under study in industrial medicine. This methodic approach should work in scientific programs reviewing influence of industrial ecology and environmental factors on reproductivity. The authors suggest elaboration of universal strategy for ecologic and epidemiologic studies to form a scientific branch "Epidemiology of reproductive health" and to elaborate methods for better health management during the present demographic crisis.     

Effects of antioxidant state on individual susceptibility to silicosis (an experimental study)

The authors proved that animals having higher natural antioxidant activity and lower intensity of lipid peroxidation are more resistant to fibrogenic effects of quartz. The peritoneal macrophages obtained from those animals show higher resistance to quartz exposure "in vitro". Parameters characterizing antioxidant system should be included into a group of indexes describing propensity to silicosis.     

Heat denaturation of hemocytocardiotoxin from the venom of the Central Asian cobra

It has been stated that boiled for three hours haemocytocardiotoxin (HT) from cobra poison loses "direct" hemolytic activity and is unable to potentiate a haemolytic effect of phospholipase A. Surface activity of HT does not change. It is shown that in the course of heat denaturation the aggregation of toxin molecules to dimers and trimers takes place and electrophoretic mobility is decreased. The fluorescence of HT tyroxin residues supported the fact of its irreversible heat denaturation.     

Quantitative Metabolomic Analysis of Changes in the Rat Blood Serum during Autophagy Modulation: A Focus on Accelerated Senescence

Autophagy is involved in the maintenance of cellular homeostasis and the removal of damaged proteins and organelles and is necessary to maintain cell metabolism in conditions of energy and nutrient deficiency. A decrease in autophagic activity plays an important role in age-related diseases. However, the metabolic response to autophagy modulation remains poorly understood. Here, we for the first time explored the effects of (1) autophagy activation by 48 h fasting, (2) inhibition by chloroquine (CQ) treatment, and (3) combined effects of fasting and CQ on the quantitative composition of metabolites in the blood serum of senescent-accelerated OXYS and control Wistar rats at the age of 4 months. By means of high-resolution ¹H NMR spectroscopy, we identified the quantitative content of 55 serum metabolites, including amino acids, organic acids, antioxidants, osmolytes, glycosides, purine, and pyrimidine derivatives. Groups of 48 h fasting (induction of autophagy), CQ treatment (inhibition of autophagy), and combined effects (CQ + fasting) are clearly separated from control groups by principal component analysis. Fasting for 48 h led to significant changes in the serum metabolomic profile, primarily affecting metabolic pathways related to fatty acid metabolism, and led to metabolism of several amino acids. Under CQ treatment, the most affected metabolites were citrate, betaine, cytidine, proline, tryptophan, glutamate, and mannose. As shown by two-way ANOVA, for many metabolites the effects of autophagy modulation depend on the animal genotype, indicating a dysregulation of metabolome reactivity in OXYS rats. Thus, the metabolic responses to modulation of autophagy in OXYS rats and Wistar rats are different. Altered metabolites in OXYS rats may serve as potential biomarkers of the manifestation of the signs of accelerated aging. Metabolic signatures characteristic to fasting and CQ treatment revealed in this work might provide a better understanding of the connections between metabolism and autophagy.
