Cytophysiology and innervation of gonadotropic cells in the pituitary of the black molly (Poecilia latipinna). An electron microscopical study

In the male black molly, poecilia latipinna, morphological and functional aspects of the gonadotropic (GTH-)cells have been studied at the ultrastructural level. The cells exclusively occupy the ventral and lateral areas of the meso-adenohypophysis. In the black molly there is evidence of the presence of only one type of gonadotropic cell. In the GTH-cells of most specimens, the rough endoplasmic reticulum is weakly developed. The secretory vesicles are characterized by cores with varying diameters; this variation was not observed in the secretory vesicles of the other types of pituitary cells, except in the TSH-cells. After applying a histochemical method for the demonstration of polysaccharides, small black deposits appear in the core of the secretory vesicles of the GTH- and TSH-cells only; this indicates the glycoproteinaceous nature of the hormones produced in these cells. Male black mollies treated with methyl-testosterone have significantly smaller GTH-cells and a lesser number of secretory vesicles and mitochondria in these cells. GTH-cell activity in Poeciliinae may be thus influenced by androgens by means of a negative feed-back mechanism. The GTH-cells are innervated by both type A and type B neurosecretory fibres. There are indications that the type A fibres may originate from the pars lateralis cells of the nucleus lateralis tuberis; the origin of the type B fibres is uncertain.     

Lamination of polyethylene composite films by ultrasonic welding: Investigation of peel behavior and identification of optimum welding parameters

The lamination of different polyethylene (PE) composite films by ultrasonic welding to fabricate peelable seals that open at defined burst pressures is investigated. The sealing time, pressure, and amplitude were varied within the range of 100–400 ms, 50–250 kPa, and 12–24 µm, respectively. T‐peel tests and electron micrographs revealed four different peel regimes, depending on the parameter combination: (I) Interlaminar peeling at low‐peel strength with uniform peeling along a weakly bonded PE lamination layer; (II) transition tearing at intermediate peel strength showing areas of interlaminar peeling and translaminar tearing; (III) translaminar tearing at high‐peel strength showing tears through the entire film; and, (IV) undefined tearing at varying tear strength occurring when vibration effects during welding lead to insufficient contact of the films or high pressures lead to a displacement of PE. Use of E mu-PIM-1 transgenic mice short-term in vivo carcinogenicity testing: lymphoma induction by benzo[a]pyrene, but not by TPA

Cyclophilins have been suggested to act as leukocyte chemotactic factors produced in the course of inflammation. Therefore we looked for the presence of cyclophilins in the synovial fluids (SF) from patients with rheumatoid arthritis (RA). Peptidyl prolyl cis-trans isomerase activity (PPIase) was measured in SF from knee punctures of 26 patients with RA and five patients with knee osteoarthritis (OA). PPIase was detected in SF from RA patients, but not in samples from OA patients. Enzyme activity was sensitive to inhibition by cyclosporin A (IC50 = 28-50 nM). Estimated concentrations of the SF-derived cyclophilin based on the enzyme activity were in the range of 11 to 705 nM. The presence of cyclophilin in the SF showed disease correlation; its concentration correlated with the number of cells in the SF (r = 0.91, P < 0.0001) and with the percentage of neutrophils in the cellular infiltrate and was higher in more acute cases of joint swelling. In immunoblots of partially purified preparations of SF from RA patients, an approximately 18-kD protein band reacted with polyclonal antibodies that recognize cyclophilin A and B, but not with antibodies specific for cyclophilin B. Sequencing of this protein revealed identity of the NH2-terminal amino acids with those of human cyclophilin A. The finding is unexpected since cyclophilin B rather than A is generally regarded as the secreted isoform, the presence of cyclophilin A being confined to the cytoplasm. Our data support the hypothesis that cyclophilins may contribute to the pathogenesis of inflammatory diseases, possibly by acting as cytokines. This may offer a possible explanation of the effectiveness of cyclosporin A in RA, in addition to the known immunosuppressive effects of the drug.     

Development of a Clinical Guide to Enhance Care for Suicidal Patients.

Suicidal thoughts and behavior are common among mental health patients and are a source of stress for clinicians, who typically receive limited formal training on suicide. The U.S. Air Force initiated a project to enhance care and increase practitioner confidence when working with suicidal patients. A clinical guide was developed containing 18 recommendations for assessing and managing suicidality, strategies for meeting the recommendations, and clinical tools to facilitate quality care. Training opportunities and marketing efforts accompanied distribution of the guide. This initial article reviews the guide's development, content, and evaluation plan as a model that other health care systems, clinics, or training programs can follow to enhance care for suicidal patients. Design and operation of a protection system for transformers with superconducting windings

Power transformers with superconducting windings need a protection system to prevent damage to the low-loss superconducting winding by an abnormally high current. The generally accepted protection technique which uses auxiliary coils has been analysed using a network representation. The current distribution between main and auxiliary coil is expressed in terms of geometrical parameters. Experimental data on current transfer and main coil recovery in a test transformer are presented and a method of obtaining a very low auxiliary coil current is suggested.
