Microcystin-affinity chromatography was used to purify 15 protein phosphatase 1 (PP1)-binding proteins from the myofibrillar fraction of rabbit skeletal muscle. To reduce the time and amount of material required to identify these proteins, proteome analysis by mixed peptide sequencing was developed. Proteins are resolved by SDS-polyacrylamide gel electrophoresis, electroblotted to polyvinylidene fluoride membrane, and stained. Bands are sliced from the membrane, cleaved briefly with CnBr, and applied without further purification to an automated Edman sequencer. The mixed peptide sequences generated are sorted and matched against the GenBank using two new programs, FASTF and TFASTF. This technology offers a simple alternative to mass spectrometry for the subpicomolar identification of proteins in polyacrylamide gels. Using this technology, all 15 proteins recovered in PP-1C affinity chromatography were sequenced. One of the proteins, PP-1bp55, was homologous to human myosin phosphatase, MYPT2. A second, PP-1bp80, identified in the EST data bases, contained a putative PP-1C binding site and a nucleotide binding motif. Further affinity purification over ATP-Sepharose isolated PP-1bp80 in a quaternary complex with PP-1C and two other proteins, PP-1bp29 and human p20. Recombinant PP-1bp80 also bound PP-1C and suppressed its activity toward a variety of substrates, suggesting that the protein is a novel regulatory subunit of PP-1.  相似文献

Continuous chemical vapour-deposited diamond-coated fibres with tungsten wire or SiC fibre cores are attractive for reinforcing metals and ceramics. The fibres have been embedded in Ti-6A1-4V alloy to produce a diamond fibre-reinforced composite. Both the fibres and the composite material are extremely difficult to cut without damage by conventional mechanical methods. The use of a Nd-YAG laser to cut these materials is described.  相似文献

Peel strengths at room temperature and under superplastic forming conditions at 530 °C were measured for diffusion-bonded joints in Al-Li 8090 alloy sheet. The bonds were made in the solid state, or via a transient liquid phase using interlayers. The effect of strain rate, sheet thickness and heat treatment were investigated. The significance of these results for the testing of DB joints and for their use in DB/SPF structures is discussed.  相似文献

Superplastic forming using conventional alloys and product forms may be a cost effective route for component manufacture. In this paper superplastic deformation of Ti-3Al-4V extruded tube is examined in terms of the strain anisotropy, microstructural changes, the texture and the post-formed tensile properties. Superplastic deformation with low flow stresses over a wide range of strain rates was observed. Strain anisotropy and surface roughening occurred during deformation and was associated with the two-phase, aligned and banded microstructure. A small reduction in the tensile properties was found and changes in the texture were noted after superplastic deformation.  相似文献

To select a source of lymphocytes for the generation of an anti-sperm-biased combinatorial phage display library, venous blood was obtained from 34 vasovasostomy (vasectomy reversal) patients approximately 3 mo after surgery. Using a variety of immunoassays, serum was analyzed for antibodies against human spermatozoa, and a patient was selected on the basis of high titer of antibodies that recognized the equatorial region of the sperm head and inhibited sperm fertilizing capacity in vitro. Total RNA isolated from the stored lymphocytes of this individual was reversed transcribed, and gamma1 (Fd) region and kappa chains were amplified by polymerase chain reaction for the successful construction of an antibody phage display library. The library was panned against human spermatozoa to isolate sperm-specific phage that recognized the equatorial region of the sperm head. Three preparations of Fab were tested via the hamster egg penetration test. Each preparation significantly (p < 0. 005) inhibited sperm-egg binding and fusion, with one preparation (designated Fab-G) causing complete inhibition. Sequence analysis of the kappa light gene encoding Fab-G revealed a 93% homology with the light chain of human anti-human immunodeficiency virus gp120 p35 variable region. This technology may have a practical application in characterization of the immune response to spermatozoa and for the design of sperm-based contraceptive vaccines.  相似文献

A fibre coating process has been used to produce continuously reinforced advanced metal-matrix composites with up to 8% volume fraction of SiC fibre. Matrix materials were an / titanium alloy (Ti-Al-V), a dispersion-strengthened titanium alloy (Ti-Al-V-Y), a rapid solidification processed aluminium alloy (Al-4.3Cr-0.3Fe), and intermetallic compounds Ti3Al and TiAl. Thick metal coatings are shown to adhere well to the fibres, no evidence is found for chemical reaction between the coating and the fibre during the coating process, and the coated fibres can be handled and bent without damage. Tensile test data for Ti-Al-V alloy reinforced with 21% SiC fibre show a modulus near to a theoretical prediction, but tensile strength significantly below prediction. Loss of strength is attributed to the formation of a brittle reaction product during hot consolidation. The advantages and potential of the coated-fibre route for MMC production are discussed.  相似文献

Peel strengths are reported for solid state or transient liquid-phase diffusion-bonded (DB) joints between aluminium-lithium 8090 alloy sheets. The joints were tested under superplastic forming (SPF) conditions at 530 °C and with a progressively increasing peel angle,, in the range 0°–60°. The sheet deformed superplastically with or without peel fracture of the joints. A deformation model is proposed which predicts a critical combination of peel strength and superplastic flow stress for DB/SPF processing of the 8090 alloy and indicates peel fracture will occur when sheet thicknesses exceed 2 and 0.8 mm in solid state and transient liquid-phase diffusion-bonded joints, respectively.  相似文献

This paper argues that the conventional methodology of software engineering is inappropriate to AI, but that the failure of many in AI to see this is producing a Kuhnian paradigm crisis. The key point is that classic software engineering methodology (which we call SPIV: Specify-Prove-Implement-Verify) requires that the problem be capable of being circumscribed or surveyed in a way that it is not, for areas of AI, like natural language processing. In addition, it also requires that a program be open to formal proof of correctness. We contrast this methodology with a weaker form complete Specification And Testability (SAT — where the last term is used in a strong sense: every execution of the program gives decidably correct/incorrect results) which captures both the essence of SPIV and the key assumptions in practical software engineering. We argue that failure to recognize the inability to apply the SAT methodology to areas of AI has prevented development of a disciplined methodology (which is unique to AI and which we call RUDE: Run-Understand-Debug-Edit) that will accommodate the peculiarities of AI and also yield robust, reliable, comprehensible, and hence maintainable AI software.  相似文献

A mathematical model has been developed to predict So2 absorption and removal during the constant rate drying period of a spray dryer. The model, based on film theory, treats the atomized slurry droplets as spheres containing discrete sorbent particles of slaked lime with the fluid uniformly distributed around the individual particles. The model includes gas and liquid phase mass transfer coefficients as well as resistance to Ca(OH)2 dissolution. A sensitivity analysis has been conducted and a comparison was made between pilot-scale experimental data and model-predicted values of S02 removal efficiency.  相似文献
