Chromatographic Studies on the Polydispersity of Hydroxyethyl Starch. A representative sample of clinically used hydroxyethyl starch was separated by semipreparative HPSEC into narrow fractions in the range of approx. 3.000 to 800.000 D. The original sample and selected fractions were characterised by gaschromatographic methylation analysis according to their substitution degrees MS and DS, which were differentiated by the substitution positions at C2, C3 and C6 of the anhydroglucoses and their kind of glycosidic bonding α-1, α-1,4 or α-1,4,6, respectively. Furthermore, polydispersity in relations to the degree of branching was determined. Mark-Houwink and molecular weight distribution parameters determined by multi-detection HPSEC are reported. The presented data demonstrate a extensive homogeneity of the original sample. The clinical relevance is discussed.  相似文献

Examined the feasibility of applying the static strength and endurance relationships suggested in 1960 by W. Rohmert to a simple manual task. Results indicate that there was a relationship between grip strength expenditure and impairments in task performance similar in respect to the static strength and endurance relationships suggested. Rare pediatric non-compaction and restrictive cardiomyopathy are usually associated with a rapid and severe disease progression. While the non-compaction phenotype is characterized by structural defects and is correlated with systolic dysfunction, the restrictive phenotype exhibits diastolic dysfunction. The molecular mechanisms are poorly understood. Target genes encode among others, the cardiac troponin subunits forming the main regulatory protein complex of the thin filament for muscle contraction. Here, we compare the molecular effects of two infantile de novo point mutations in TNNC1 (p.cTnC-G34S) and TNNI3 (p.cTnI-D127Y) leading to severe non-compaction and restrictive phenotypes, respectively. We used skinned cardiomyocytes, skinned fibers, and reconstituted thin filaments to measure the impact of the mutations on contractile function. We investigated the interaction of these troponin variants with actin and their inter-subunit interactions, as well as the structural integrity of reconstituted thin filaments. Both mutations exhibited similar functional and structural impairments, though the patients developed different phenotypes. Furthermore, the protein quality control system was affected, as shown for TnC-G34S using patient’s myocardial tissue samples. The two troponin targeting agents levosimendan and green tea extract (-)-epigallocatechin-3-gallate (EGCg) stabilized the structural integrity of reconstituted thin filaments and ameliorated contractile function in vitro in some, but not all, aspects to a similar degree for both mutations.  相似文献

The greatest risk factor for developing Alzheimer’s disease (AD) is increasing age. Understanding the changes that occur in aging that make an aged brain more susceptible to developing AD could result in novel therapeutic targets. In order to better understand these changes, the current study utilized mice harboring a regulatable mutant P301L human tau transgene (rTg(TauP301L)4510), in which P301L tau expression can be turned off or on by the addition or removal of doxycycline in the drinking water. This regulatable expression allowed for assessment of aging independent of prolonged mutant tau expression. Our results suggest that P301L expression in aged mice enhances memory deficits in the Morris water maze task. These behavioral changes may be due to enhanced late-stage tau pathology, as evidenced by immunoblotting and exacerbated hippocampal dysregulation of glutamate release and uptake measured by the microelectrode array technique. We additionally observed changes in proteins important for the regulation of glutamate and tau phosphorylation that may mediate these age-related changes. Thus, age and P301L tau interact to exacerbate tau-induced detrimental alterations in aged animals.  相似文献

Bronchoalveolar lavage (BAL) specimens (n = 213) from AIDS and non-HIV immunosuppressed patients were investigated for the presence of Pneumocystis carinii infection by fluorescence microscopy of Papanicolaou-stained slides. Alveolar casts, extracellular pneumocysts and phagocytosed cysts and their degradation products in pulmonary alveolar macrophages were identified. The number of phagocytosed pneumocysts within human pulmonary alveolar macrophages was recorded and correlated with the number of extracellular cysts and alveolar casts, in both groups of patients. Both phagocytic and degradation capacity were depressed in AIDS patients. This observation may explain the large number of extracellular organisms found in BAL specimens of AIDS patients compared with non-HIV-positive immunocompromised individuals.  相似文献

Docker provides a good basis to run composite applications in the cloud, especially if those are not cloud-aware, or cloud-native. However, Docker concentrates on managing containers on one host, but SaaS providers need a container management solution for multiple hosts. Therefore, a number of tools emerged that claim to solve the problem. This paper classifies the solutions, maps them to requirements from a case study and identifies gaps and integration requirements. We close some of these gaps with our own integration components and tool enhancements, resulting in the currently most complete management suite.  相似文献

The majority of the world's large river systems is affected by dams. The influences of unnatural regimes induced by flow management are wide‐ranging from both biotic and abiotic standpoints. However, many of these effects are not evident over short (1–2 years) periods (e.g. impacts of annual flow variation). This study examines the long‐term effects of annual flow variation on the macroinvertebrate community in the Chattahoochee River (GA) in the reaches below Buford Dam, the major water control structure on the river. Quarterly, macroinvertebrate samples were taken from 2001 to 2011 using Surber and Hester–Dendy plate samplers at six locations spread across 65 km below the dam. Data were analysed via analysis of similarities to determine differences in community composition between high‐flow (mean discharge = 58.27 m3/s) and low‐flow (mean discharge = 26.53 m3/s) years. Taxa that contributed most to community differences were determined via similarity percentages analyses and subsequent t‐tests. Several insect taxa (e.g. Cheumatopsyche and Ceratopsyche caddisfly larvae, Maccaffertium mayfly nymphs and Taeniopteryx stonefly nymphs) were more prevalent under the high‐flow regime. Non‐insect macroinvertebrates (e.g. Crangonyx amphipods, Tricladida flatworms and Caecidotea isopods) were more abundant under low‐flow conditions. In terms of taxon richness, no significant effects of flow regime were detected. Implications of macroinvertebrate patterns for the fishery and ecological health of the river are discussed. Supplementation of calcium and phosphate is recommended in nutrition of low birth weight infants to ensure a physiological development of postnatal bone mineralisation. To investigate whether high dose calcium supplementation increases the risk of renal calcification in preterm infants, serial ultrasound examinations were performed in 30 preterm infants (gestational age 29.5 (26-35) weeks; birth weight 1382 (610-2010) g) before, during and after oral calcium and phosphate supplementation. Total calcium input was on average 3.1 mmol/kg/d, total phosphate input on average 2.1 mmol/kg/d). All children showed normal kidney ultrasound before entering the study. During mineral supplementation three children developed hyperechoic renal medullary pyramids, the typical ultrasound pattern of nephrocalcinosis. Supplementation was stopped immediately and pathologic ultrasound patterns disappeared in all 3 children.  相似文献

Material and methods: The growth and the regression of the experimental tumor S 180 was investiaged by volume measurements and by cytophotometric studies on 50 mice in each of a pilot and in the present main experiment. In addition, histological, cytological and cytokinetic examinations were performed. Results and discussion: Beginning on day 18, a spontaneous regression of the tumor was found in 20% and 38% of the animals, respectively. DNA-measurements were performed with a scanning-microspectrophotometer on tumor tissue imprints and tumor cell suspensions and were also carried out with a flow-through cytophotometer ICP on suspensions. DNA-histograms were plotted on days 4, 7, 12, 18, 20, 22, 26 and 29 after the transplantation. These revealed a constant position of the maxima at 2C for normal cells and at 4C and 8C for the tumor cells. In particular, by measuring a large total number of 4,968,000 nuclei with the ICP, distinct changes were found in the proportion of the single nuclei classes during growth and spontaneous regression. This technique also enables a quantitative measurement to be made of the DNA of cell debris from necrosis. With growing tumos, the proportion of the tumor cells increased to a maximum of 65% on day 18 and decreased to 37% on day 29. The DNA of cell debris grew from 8% to 47%. The proportion of normal cells was only 10% to 15% in the final phases. In tumors with a spontaneous regression the proportion of the tumor cell nuclei was 20% on day 18 and 11% on day 29. The proportion of the DNA of cell debris was again about 45%. The proportion of normal nuclei was greatly increased to 45%. Histological and cytological evidence together with measurements of the areas of nuclei and incorporation of 3H-TdR confirmed that the normal cells, the number of which increased during spontaneous regression were cells of a fibrovascular granulation tissue. While the rate of the DNA-synthesis of growing tumors continously decreased with age there was also a rapid decrease of the labelling index of spontaneously regressing tumors between days 12 and 18. In the final phase only the nuclei of the granulation tissue were labelled.  相似文献
