The aim of this work was to identify which proteins in horse dander extracts are allergens and to characterise them. Two-dimensional PAGE showed that horse dander preparations are composed of up to 50 proteins, all having acidic isoelectric points in the pH range 3-4.5. Immunoblots of two-dimensional PAGE were used to compare the reactivity of the proteins with IgE from 23 allergic patients. Patient sera were divided into two main groups recognising either allergens of 18.5 kDa or proteins of 27-29 and 31 kDa. The proteins of 27-29 kDa and 31 kDa were all N-glycosylated and their glycan chains seem to play a role in the binding of IgE from allergic patients. The sugar composition of their carbohydrate moiety was determined and lectin-binding experiments indicated presence of terminal sialic acid linked alpha-(2-->6) to galactose, galactose linked beta-(1-->4) to N-acetylglucosamine, and possibly presence of sialic acid linked alpha-(2-->3) to galactose. The 27-29-kDa glycoproteins had heterogeneous isoelectric points, most probably due to different degrees of sialylation in their oligosaccharide chains. The two 18.5-kDa allergens exhibited slightly different isoelectric points and their N-terminal sequences were identical, showing that they most likely were isoforms of the same protein. Sequence analyses revealed that their N-terminal sequences are similar to proteins belonging to the lipocalin family. We named the two 18.5-kDa proteins Equ c 2.0101 and Equ c 2.0102, according to International Allergen Nomenclature recommendations [King, T. P., Hoffman, D., Lowenstein, H., Marsh, D. G., Platts-Mills, T. A. E. & Thomas, W. (1995) J. Allergy Clin. Immunol. 96, 5-14]. The N-terminal of the allergens of 27-29 kDa were blocked and their sequences were not determined. Their amino acid compositions were determined and comparison with acidic mammalian proteins in the Swiss-Prot database revealed high scores with lipocalin proteins. This suggests that the glycosylated horse dander allergens belong to the lipocalin family, like Equ c 2.0101 and Equ c 2.0102. 相似文献

One of the primary aims of higher education in today’s information technology enabled classroom is to make students more active in the learning process. The intended outcome of this increased IT-facilitated student engagement is to foster important skills such as critical thinking used in both academia and workplace environments. Critical thinking (CT) skills entails the ability(ies) of mental processes of discernment, analysis and evaluation to achieve a logical understanding. Critical thinking in the classroom as well as in the workplace is a central theme; however, with the dramatic increase of IT usage the mechanisms by which critical thinking is fostered and used has changed. This article presents the work and results of critical thinking in a virtual learning environment. We therefore present a web-based course and we assess in which parts of the course, and to what extent, critical thinking was perceived to occur. The course contained two categories of learning modules namely resources and interactive components. Critical thinking was measured subjectively using the ART scale. Results indicate the significance of “interactivity” in what students perceived to be critical-thinking-oriented versus online material as a resource. Results and opportunities that virtual environments present to foster critical thinking are discussed. 相似文献

The aim of this paper is to carry out extensive numerical investigations about the effect of various structural parameters on the dynamic performance of stiffened box steel bridge pier under a strong earthquake ground motion. The considered structural parameters are the local slenderness ratios of stiffener and stiffened wall as well as the global slenderness ratio of the pier. In the present study, all investigated piers are made of high tensile steel with high yield ratio. The non-linear time history analyses are carried out using in-house Finite Element Program DYNAPSS, recently developed by the first author. This program is verified by comparing its results with the results obtained by the general FE program. In this program, the geometrical nonlinearity is considered on the bases of Total Lagrangian formulation, while the non-linearity of structural steel material is considered through implementation of modified multisurface cyclic plasticity model by which real characteristics of high tensile steel material such as, Plateau, Massing type of Bauschinger??s effect, disappearing of Plateau and cyclic strain hardening, are accurately modeled. The results are closely examined in respect with the damage index criterion. From this study, it is found that the increase in slenderness ratios of stiffener does not only increase the damage index, but also may lead to the full collapse. Furthermore, the damage index increases with the increase of local slenderness ratio of stiffened wall and the global slenderness ratio. Also, it is found that when the loacal buckling is not the dominant eigen mode and the pier behave as a single degree of freedom, the natural period has destructive effects when it is close to predominant period of the earthquake. Finally, contour plots of damage index values are presented. These contour plots may be greatly useful for the design of stiffened box steel bridge piers, where, critical values of structural parameters corresponding to certain value of damage index could be determined. 相似文献

Recently, the prediction of project duration has been investigated in earned value management (EVM) using three earned value methods, planed value (PVM), earned duration (EDM) and earned schedule (ESM). The investigation has shown that ESM method outperforms on average the other two methods and fails in case of wrong warning coming from non-critical activities which suffer from delays and/or ahead of schedule. The objective of this paper is twofold: first, we study the impact of the activities' sensitivity information on the forecasting accuracy of the ESM method. Second, we test the claim that in normal conditions the project performance indicator provided by ESM at higher work breakdown structure is reliable. More precisely, activity based sensitivity measures are used as weighing parameters of the activities to improve the schedule performance of a project by removing or decreasing the negative effect of wrong warning of the non-critical activities. The computational results of a simulation study on a big benchmark projects reveal that the sensitivity information are capable of improving the forecasting accuracy of the ESM method. 相似文献

The study presented in this paper sought to explore several dimensions to online learning. Identifying the dimensions to online learning entails important basic issues which are of great relevance to educators today. The primary question is “what are the factors that contribute to the success/failure of online learning?” In order to answer this question, we need to identify the important variables that: (1) measure the learning outcome and (2) help us understand the learning experience of students using specific learning tools. In this study, the dimensions we explored are student’s attitude, affect, motivation and perception of an Online Learning Tool usage. A survey methodology was used such that validated items from previous relevant research work were adopted. 105 students completed the questionnaire. An exploratory factor analysis (EFA) was implemented on the data captured. Results of the EFA identified the items that are relevant to the present context and the items that can be used to measure the dimension to online learning. Affect and perception were found to have strong measurement capabilities with the adopted items while motivation was measured the weakest. 相似文献

Based on gelatin (Gltn) and acrylic acid (AAc), biodegradable pH‐sensitive hydrogel was prepared using gamma radiation as super clean source for polymerization and crosslinking. Incorporation of PAAc in the prepared hydrogel was confirmed by Fourier transform infrared spectroscopy (FTIR). The effect of PAAc content on the morphological structure of the prepared hydrogel swollen at pH 1, 5, and 7 was examined using scanning electron microscopy (SEM). The results showed the dependence of the porous structure of the prepared hydrogels on AAc content and the pH of the swelling medium. Swelling properties of gelatin/acrylic acid copolymer hydrogels with different AAc contents were investigated at different pH values. Swelling data showed that the prepared hydrogels possessed pronounced pH sensitivity. In vitro release studies were performed to evaluate the hydrogel potential as drug carrier using ketoprofen as a model drug. Desertification in Kuwait is a process of environmental degradation under fragile ecological conditions and intensive human
activities and the consequences of Gulf War. In Kuwait, very severe desertification prevails, due to increasing formation
of new active sandy bodies, deterioration of many areas of natural vegetation cover to less than 10%, and limited water resources
for large-scale forage production. Average annual desertified land in Kuwait is estimated to be 285 km2. In Kuwait, three indicators of land degradation are encountered. These are vegetation, soil, and surface hydrological changes.
Based on field measurements of soil compaction and vegetation changes, in the west Jahra area in the northern part of the
country, degradation levels were assessed. Results of these measurements show that the average infiltration rate in compacted
soil decreased by 53.8% in comparison with non-compacted soil, while the average soil penetration resistance in compacted
soil increased by 154.1% in comparison with non-compacted soil. The bulk density in open sites was 23.4% higher than that
in protected sites. The percentage of litter in open sites decreased by 77.3% in comparison with protected sites, while the
percentage of total vegetation in open sites decreased by 6.1% in comparison with protected sites.
