Cell therapy of the post-infarcted myocardium is still far from clinical use. Poor survival of transplanted cells, insufficient regeneration, and replacement of the damaged tissue limit the potential of currently available cell-based techniques. In this study, we generated a multilayered construct from adipose-derived mesenchymal stromal cells (MSCs) modified to secrete stem cell factor, SCF. In a rat model of myocardium infarction, we show that transplantation of SCF producing cell sheet induced activation of the epicardium and promoted the accumulation of c-kit positive cells in ischemic muscle. Morphometry showed the reduction of infarct size (16%) and a left ventricle expansion index (0.12) in the treatment group compared to controls (24–28%; 0.17–0.32). The ratio of viable myocardium was more than 1.5-fold higher, reaching 49% compared to the control (28%) or unmodified cell sheet group (30%). Finally, by day 30 after myocardium infarction, SCF-producing cell sheet transplantation increased left ventricle ejection fraction from 37% in the control sham-operated group to 53%. Our results suggest that, combining the genetic modification of MSCs and their assembly into a multilayered construct, we can provide prolonged pleiotropic effects to the damaged heart, induce endogenous regenerative processes, and improve cardiac function.  相似文献

The authors presented the cases of two children with inflammatory myofibroblastic (IMF) tumor and reviewed the literature to facilitate the preoperative recognition, delineate the clinical features, and describe the natural history of this entity. The first child had IMF tumor arising from the mesentery of the small intestine. He presented with an abdominal mass associated with severe inflammatory response manifested by fever, impaired growth, thrombocytosis, and microcytic, hypochromic anemia. After surgical resection, his fever resolved and his growth rate and the laboratory abnormalities normalized. Five months after initial diagnosis, the fever, anemia, and thrombocytosis recurred along with two tumors arising from the omentum and the abdominal soft tissue. After the second surgery, he remains free of recurrent disease for 30 months. The second child presented with a lung mass that was radiologically indistinguishable from pulmonary sequestration. After surgical resection, she remains free of recurrent disease for 18 months. IMF tumor should be considered in any solid tumor that occurs in association with a chronic inflammatory response. IMF tumor should also be considered in the differential diagnosis of pulmonary sequestration.  相似文献

Random lasing originates from pumping disordered materials by high-intensity and high-frequency light. The performance of random lasers is restricted due to the absorption of pumping light within the interfacial layer. The lifetime of optical modes there is short due to the emission of photons to the outside, and it is hard to pump them sufficiently for lasing. The opportunity to excite the modes far from the material surface using two-photon absorption is investigated within the diffusion model. The authors show that lasing requires lower pump power for the two-photon pumping mechanism under conditions of negligible absorption of the emitted light. Experimental implications and restrictions are discussed.  相似文献

Conclusions Experiments at the Magnezit Plant confirmed the feasibility of drying magnesia refractories with the flue gas in the first positions of a tunnel kiln. Drying the refractories in the section where the flue gas is tapped off does not result in more rejects or in degradation of the properties of the fired products.Compounding the processes of drying and firing magnesia refractories in a single unit, viz., the tunnel kiln, makes it possible to dispense with the driers the freed floorspace of which can be used for installing equipment for mechanizing the loading of the products on the kiln trucks.Translated from Ogneupory, No. 4, pp. 15–21, April, 1976.  相似文献

Transposons of gypsy group are assigned to LTR-containing retrotransposons present in the genomes of invertebrates, fungi, and plants. In this work, a theoretical analysis of the potential products of ORFs of these retrotranposons was conducted. Alignments were obtained and trees of similarity were constructed for domains of the POL region. On the basis of the obtained data, two hypothetically monophyletic subgroups of transposons were distinguished within the framework of the gypsy group, settling the genomes of taxonomically related organisms (the subgroup of "true" gypsy of insects and the subgroup of gypsy-like transposons of plants and fungi). A number of peculiarities of the topology of these trees hypothetically indicate cases of genetic conversion and recombination of domains accompanying the evolution of this group. The amino acid substitution fixation rate was evaluated on the basis of comparison of sequences of the protein products of ORFs. Estimates of the time of divergence of subgroups of gypsy-group transposons are significantly less than estimates of the times of divergence of their host species. One explanation for this discrepancy might be the hypothesis of settlement by transposons of the genomes of isolated host species.  相似文献

The current study examined the self-excited thermoacoustic instability of hydrogen/methane premixed flames using a variable-length combustor (300–1100 mm). The global dynamic pressure, heat release rate oscillation, together with the flame dynamics were studied. Results showed that both the hydrogen concentration and the chamber length were critical in determining the acoustic oscillation mode and instability trend. Low-frequency primary acoustic modes (<200 Hz) were mainly excited when the hydrogen concentration was low, whereas primary acoustic modes with relatively higher frequencies (~400 Hz) tended to occur in cases with a high hydrogen proportion (>40%). For primary acoustic modes lower than 200 Hz, the primary oscillation frequency tended to increase linearly with a rising hydrogen proportion. Heat release oscillation and flame dynamics analyses demonstrated that for the flame with large-scale shape deformation, the initial addition of hydrogen would intensify the heat release oscillation. Nevertheless, a further increase in the hydrogen level tended to inhibit the heat release oscillation by weakening the flame shape deformation. Eventually, a sufficient high-level of hydrogen addition would weaken the primary acoustic modes that have similar frequencies.  相似文献

This work describes new type of combustion instability for which the 3-way coupling between mixing, flame heat release, and acoustics is modified by local buoyancy effects. Measurements of fuel mixture fraction are made for a non-premixed jet flame in a combustion chamber to assess the dynamics of mixing under imposed acoustic oscillations (22-55 Hz). Infrared laser absorption and phase resolved acetone-planar laser induced fluorescence are used to measure the fuel mixture fraction and then the degree of fuel/air mixing is calculated by determining the unmixedness. Results show acoustic excitation causes oscillations in the degree of fuel/air mixing at the driving frequency, which results in oscillatory flame behavior. This oscillatory flame behavior couples to the buoyancy and this in turn affects the mixing. Results also show that the mixing becomes less effective when the excitation frequency is increased or when the flame is present, compared to the non-reacting case. This work describes a key coupling mechanism that occurs when buoyancy is a significant factor in the flow field.  相似文献

Crosslinked poly(ester urethane)s and their acrylate derivatives based on trifunctional polycaprolactone and trifunctional aliphatic isocyanates were synthesized. Biodegradable scaffolds with uniform, controlled micron-scale porosity were fabricated with these materials. Mechanical and swelling properties of monolithic and microporous materials were studied. Cytotoxicity, hydrolytic, and enzymatic degradation and their effects on mechanical properties of the biodegradable scaffolds were investigated. The polymer degradation products were found not to be cytotoxic at moderate concentrations and to permit cell attachment and spreading. Degradation rates and mechanical properties could be tuned to desired performance criteria for a given application by adjusting crosslink density and the ratio of hard segment to soft segment. © 2019 Wiley Periodicals, Inc.
