Drowned river mouth lakes are major features of coastal Great Lakes habitats and may influence nutrient and organic matter contributions from watersheds to near shore coastal zones. In May through October 2003, we measured loads of nutrients, surficial sediment, and seston to track the delivery of riverine-derived materials from the lower Muskegon River Watershed (MRW) into the near shore area of southeast Lake Michigan. Nutrient flux data indicated that seasonal loads of 1800 metric tons (MT) of particulate organic carbon, 3400 MT of dissolved organic carbon, and 24 MT of total phosphorus were discharged from the lower Muskegon River, with approximately 33% of TP load and 53% of the POC load intercepted within the drowned river mouth terminus, Muskegon Lake. Carbon: phosphorus molar ratios of seston in Muskegon River (C:P = 187) and Muskegon Lake (C:P = 176) were lower than in Lake Michigan (C:P = 334), indicating phosphorus limitation of phytoplankton in near shore Lake Michigan. Isotopic signatures of seston collected in Muskegon Lake were depleted in δ13C (− 30.8 ± 1.6‰) relative to the isotope signatures of seston from Lake Michigan (− 26.2 ± 1.3‰) or the mouth of the Muskegon River (− 28.1 ± 0.5‰), likely due to the presence of biogenic methane in Muskegon Lake. Seston δ15N increased on a strong east-to-west gradient within Muskegon Lake, indicating significant microbial processing of nutrients. The extent of nutrient uptake in Muskegon Lake altered the chemical and isotopic characterization of seston flowing into Lake Michigan from Muskegon River. 相似文献

The high temperature corrosion resistance of Ni-25.9Cr-13.5Al-1.2Y-0.6Si and Ni-10.2Co-12.4Cr-16.0Al-0.5Y-0.2Hf alloys was assessed in sulfidation/oxidation environments.In the environment with a sulfur partial pressure of 1Pa. and an oxygen partial pressure of 10^-19Pα,both these alloys exhibited three distinct stages in the weight gain-time curve when tested at 700℃.In the initial stage, selective sulfidation of Cr suppressed the formation of the other metal sulfides,resulting in lower weight gains.In the transient stage, breakdown and cracking of Cr sulfides and insufficient concentration of Cr at the outer zone led to the rapid formation of Ni sulfides and a rapid increase in weight.In the steady-state stage, corrosion was controlled by the diffusion of anions and/or cations, which led to a parabolic rate law. 相似文献

A carbon budget was produced for each month of the International Field Year for the Great Lakes (IFYGL) year (April 1972 to March 1973) to determine the importance of the various sources and sinks of carbon. Major sources were found to be CO2 which was fixed in organic matter during primary production and inorganic carbon in tributary streams, especially the Niagara River. The major sinks were found to be inorganic carbon outflow at the St. Lawrence River and net CO2 gas exchange between the inorganic carbon pool and the atmosphere. Inflow and outflow of organic matter in rivers, sedimentation of organic and inorganic matter, ground water transport, and municipal and industrial perturbations accounted in total for less than 10% of the annual budget.The lake had an inventory of approximately 4.0 × 1010 kg of inorganic carbon and approximately an order of magnitude less organic carbon. The riverborne flux of inorganic carbon of 0.5 × 1010 was 13% of the lake's inventory, assuming complete mixing; a minimum mean residence time of 8 years can be calculated from that inventory.The seasonal cycle inherent in the fixation of carbon in primary production was primarily balanced by a complementary seasonal cycle in the air-lake CO2 gas exchange system. The lake acts as a sink for CO2 gas in the warm months when primary productivity is highest and as a source of CO2 in the colder part of the year.The IFYGL year had higher than normal rates of water flow, but this does not appear to have perturbed the inorganic carbon system. Interaction with the exsorptive transporter P-glycoprotein (P-gp) is a possible source of peculiarities in drug pharmacokinetics, including dose-dependent absorption, drug-drug interactions, intestinal secretion, and limited permeability of the blood-brain barrier. Among the established in vitro methods of the analysis of drug interactions with P-gp, none directly quantifies the affinity of ligands with P-gp. Instead, they measure the result of a membrane permeation and a receptor-binding process; this may lead to difficulties in the interpretation of results. An assay for quantification of drug affinity to the transporter is presented on the basis of the radioligand-binding assay principle. This has the advantage of directly quantifying the interaction between drugs and P-gp. Because of the reversible and competitive interaction of numerous substrates with P-gp, a radioligand-binding assay was developed by taking [3H]verapamil and [3H]vinblastine as radioligands and the human intestinal Caco-2 cells, overexpressed with P-gp by culturing in the presence of vinblastine or transfecting with multidrug resistance gene MDR-1 as receptor preparation. The assay was performed in 96-well plates and has the potential to be used as a high-throughput method. A clear induction of the expression of P-gp was demonstrated in the Caco-2 cells grown in the presence of vinblastine, as well as in the transfected cells, although to a lesser extent. Both radioligands were shown to bind to P-gp. Verapamil was the radioligand of choice for further investigations due to its lower nonspecific binding to the transporter preparation. Kinetics as well as specificity of the binding of verapamil to the P-gp preparation were demonstrated. A two-affinity model was found to adequately describe the data derived from saturation as well as from competition experiments, in accordance with previous findings on two exsorption sites for P-gp. The binding properties of [3H]verapamil and [3H]vinblastine to a P-gp preparation derived from induced Caco-2 cells are described. The concentration-dependent displacement of the radioligand by nonlabeled substrates for P-gp should be a suitable principle for the determination of drug affinity to the respective binding sites at the human intestinal multidrug transporter P-gp. 相似文献
