Sudden heating of a fluid in a tube or narrow channel induces localized pressure gradients and transient flow which can significantly affect the heat transfer rate. Experiments with a long, pulse-heated tube containing refrigerant 113 produced induced flow as well as pressure oscillations. The characterization of many cytokines involved in the control of hematopoiesis has led to intense investigation into their potential use in ex vivo culture to expand progenitor numbers. We have established the optimum ex vivo culture conditions that allow substantial amplification of transient engrafting murine stem cells and which, simultaneously, augment the ability to sustain serial bone marrow transplantation (BMT). Short-term incubation of unfractionated BM cells in liquid culture with stem cell factor (SCF) and interleukin-11 (IL-11) produced a 50-fold amplification of clonogenic multipotential progenitors (CFU-A). Following such ex vivo expansion, substantially fewer cells were required to rescue lethally irradiated mice. When transplanted in cell doses above threshold for engraftment, BM cells expanded ex vivo resulted in significantly more rapid hematopoietic recovery. In a serial transplantation model, unmanipulated BM was only able to consistently sustain secondary BMT recipients, but BM expanded ex vivo has sustained quaternary BMT recipients that remain alive and well more than 140 days after 4th degree BMT. These results show augmentation of both short-term recovery posttransplant and the ability to serially transplant marrow by preincubation in culture with SCF and IL-11.  相似文献

TRPM7 plays an important role in cellular Ca2+, Zn2+ and Mg2+ homeostasis. TRPM7 channels are abundantly expressed in ameloblasts and, in the absence of TRPM7, dental enamel is hypomineralized. The potential role of TRPM7 channels in Ca2+ transport during amelogenesis was investigated in the HAT-7 rat ameloblast cell line. The cells showed strong TRPM7 mRNA and protein expression. Characteristic TRPM7 transmembrane currents were observed, which increased in the absence of intracellular Mg2+ ([Mg2+]i), were reduced by elevated [Mg2+]i, and were inhibited by the TRPM7 inhibitors NS8593 and FTY720. Mibefradil evoked similar currents, which were suppressed by elevated [Mg2+]i, reducing extracellular pH stimulated transmembrane currents, which were inhibited by FTY720. Naltriben and mibefradil both evoked Ca2+ influx, which was further enhanced by the acidic intracellular conditions. The SOCE inhibitor BTP2 blocked Ca2+ entry induced by naltriben but not by mibefradil. Thus, in HAT-7 cells, TRPM7 may serves both as a potential modulator of Orai-dependent Ca2+ uptake and as an independent Ca2+ entry pathway sensitive to pH. Therefore, TRPM7 may contribute directly to transepithelial Ca2+ transport in amelogenesis.  相似文献

Severe vibrations, known as chatter, occur often in both hot and cold rolling of steel. A recent chatter outbreak at the second stand of hot strip mill in Hastings, Australia, prompted an investigation into the causes of chatter by a literature review, the development and deployment of mathematical models, and a rigorous analysis of plant observations. The investigation suggests that the frictional conditions in the roll gap are the principal cause of chatter in this mill, though residual chatter marks on work rolls can occasionally cause it. The frictional conditions appear to be associated with the thickness and properties of oxide formed on rolls.  相似文献

One of the challenges of tissue engineering, a promising cell-based treatment for damaged or diseased cartilage, is designing the scaffold that provides structure while the tissue regenerates. In addition to the scaffold material’s biocompatibility, mechanical properties, and ease of manufacturing, scaffold interactions with the cells must also be considered. In cartilage tissue engineering, a range of scaffolds with various degrees of cell attachment have been proposed, but the attachment density and type have yet to be optimized. Several techniques have been developed to modulate cell adhesion to the scaffold. These studies suggest that the need for cell attachment in cartilage tissue engineering may vary with cell type, stage of differentiation, culture condition, and scaffold material. Further studies will elucidate the role of cell attachment in cartilage regeneration and enhance efforts to engineer cell-based cartilage therapies.  相似文献

This paper looks ahead to the mid-term and longer-term effects of the eCommerce revolution. We are in the last few years of the domination of the industrial economy — soon the information economy, or network economy, will take its place as the main engine of wealth. The underlying trends that are driving this revolution are discussed and guidelines offered for success in the new commercial world. Often these guidelines seem counter-intuitive and against conventional commercial wisdom.The paper describes how customer and business relationships are being re-formed by Internet technologies, why the revolution is an all-pervasive irresistible force, and how it threatens previously profitable industries, while creating a multitude of new ones.Perhaps the most fundamental change for companies will be the advent of supply networks, an alternative to traditional, linear supply chains. For consumers the most fundamental change will be the increase in their buying power. The new role of advertisers, the importance of trust, the threat from 'Big Brother', and the changes in the global economy are all examined.There are four main sections that look at issues for companies, customers, network-economy trends, and finally, a section that describes why some companies are at risk.  相似文献

In contrast to secondary pattern superposition, where the fields reflected from the main reflector arising from each element are superimposed in the far field of the reflector, the approach presented here sums the primary fields at the reflector surface before the physical optics radiation integral is performed. The method allows each feed array element to have arbitrary position, orientation, pattern, and excitation (magnitude and phase). In addition, it is inherently efficient because evaluation of only one time-consuming radiation integral is required, rather than one per feed element as in secondary superposition. The method allows for accurate calculation of the power radiated from the feed, permitting the reflector gain and spillover efficiency to be determined within the context of a single computer program. The accuracies and characteristics of this method are demonstrated with several examples  相似文献
