PURPOSE: To examine in the human trabecular meshwork lysosomal enzymes and one inhibitor of serine proteases that actively participate in the degradation of macromolecules into low molecular weight constituents. METHODS: Using an avidin-biotin-peroxidase technique, lysosomal proteases and alpha 1-proteinase inhibitor were examined in the trabecular meshwork of 23 human eyes with donor ages ranging from 2 to 90 years. These eyes were categorized into three age groups (< or = 20, 21 to 49, and > or = 50 years). Histochemical staining for lysosomal hydrolases was also performed on frozen sections of 20 human eyes. The staining was analyzed by an image analyzer and the levels of lysosomal proteases were further measured by biochemical assays. RESULTS: The trabecular meshwork from all the eyes stained intensely against antibodies to cathepsins B and G and alpha 1-proteinase inhibitor. The staining for elastase was weaker but evident. Image analyses revealed that the staining intensity for each protease or inhibitor was similar in all age groups. The staining in the uveal meshwork appeared to be the strongest among all the trabecular meshwork regions. Biochemical assays of tissue extracts confirmed that the enzyme and inhibitor levels were comparable among the three donor age groups. Activities of two lysosomal hydrolases, acid phosphatase and acid esterase, were also found in trabecular meshwork cells of 20 eyes. No apparent difference in enzyme activities was found with increasing age, and variation related to region was not observed. CONCLUSIONS: This study demonstrated the age-independent distribution of a variety of lysosomal enzymes and a protease inhibitor in the human trabecular meshwork. The presence of these proteins suggests a possible role in the metabolic operation of the trabecular meshwork.  相似文献

In Experiment 1, water-deprived Sprague-Dawley rats were given 5 min access to saccharin. This tube retracted, an empty tube advanced, and the rats were given 1 hr to lick the empty tube on a fixed-ratio 10 lick contingency to self-administer saline or cocaine (0.33 mg/infusion) via an intravenous catheter. The results showed that rats avoided intake of saccharin after saccharin-cocaine pairings and that greater avoidance of the gustatory cue was associated with greater cocaine self-administration. In Experiment 2, a similar dose-response function was obtained with either the empty tube or a lever as the operant. In Experiment 3, avoidance of the saccharin cue and the propensity to self-administer cocaine were maintained after at least 1 month of abstinence. Many grid connected power electronic systems, such as STATCOMs, UPFCs, and distributed generation system interfaces, use a voltage source inverter (VSI) connected to the supply network through a filter. This filter, typically a series inductance, acts to reduce the switching harmonics entering the distribution network. An alternative filter is a LCL network, which can achieve reduced levels of harmonic distortion at lower switching frequencies and with less inductance, and therefore has potential benefits for higher power applications. However, systems incorporating LCL filters require more complex control strategies and are not commonly presented in literature. This paper proposes a robust strategy for regulating the grid current entering a distribution network from a three-phase VSI system connected via a LCL filter. The strategy integrates an outer loop grid current regulator with inner capacitor current regulation to stabilize the system. A synchronous frame PI current regulation strategy is used for the outer grid current control loop. Linear analysis, simulation, and experimental results are used to verify the stability of the control algorithm across a range of operating conditions. Finally, expressions for ""harmonic impedance" of the system are derived to study the effects of supply voltage distortion on the harmonic performance of the system.  相似文献

The study of trace metal cycling by aquatic protists is limited by current analytical techniques. Standard "bulk" element analysis techniques that rely on physical separations to concentrate cells for analysis cannot separate cells from co-occurring detrital material or other cells of differing taxonomy or trophic function. Here we demonstrate the ability of a synchrotron-based X-ray fluorescence (SXRF) microprobe to quantify the elements Si, Mn, Fe, Ni, and Zn in individual aquatic protist cells. This technique distinguishes between different types of cells in an assemblage and between cells and other particulate matter. Under typical operating conditions, the minimum detection limits are 7.0 x 10(-16) mol microm(-2) for Si and between 5.0 x 10(-20) and 3.9 x 10(-19) mol microm(-2) for Mn, Fe, Ni, and Zn; this sensitivity is sufficient to detect these elements in cells from even the most pristine waters as demonstrated in phytoplankton cells collected from remote areas of the Southern Ocean. Replicate analyses of single cells produced variations of <5% for Si, Mn, Fe, and Zn and <10% for Ni. Comparative analyses of cultured phytoplankton cells generally show no significant differences in cellular metal concentrations measured with SXRF and standard bulk techniques (spectrophotometry and graphite furnace atomic absorption spectrometry). SXRF also produces two-dimensional maps of element distributions in cells, thereby providing information not available with other analytical approaches. This technique enables the accurate and precise measurement of trace metals in individual aquatic protists collected from natural environments.  相似文献

The Finniss River in the wet-dry tropics of northern Australia has received acid rock drainage (ARD) contaminants from the Rum Jungle uranium/copper mine site over more than four decades. Annual-cycle loads of Cu, Zn, Mn, and sulfate, calculated from daily water and flow measurements, have been determined both prior to and following mine-site remediation, that began in the early 1980s. The effects of varying contaminant loads on the relative abundances of seven fish species, sampled by enmeshing nets during dry seasons, were determined by nonmetric multidimensional scaling (nMDS), in combination with cluster-analysis and other nonparametric statistical techniques. These analyses showed that (i) prior to remediation, the impacted region of the Finniss River in 1974 had significantly dissimilar (P < 0.001) and more heterogeneous fish communities, generally characterized by reduced diversity and abundance, compared to sites unexposed to elevated contaminant water concentrations and (ii) postremediation, recovery in fish communities from the impacted region was indicated because they were not significantly dissimilar from those sampled at contemporary (P = 0.16) unimpacted sites, that were also similar to preremedial unimpacted sites. Even though considerable contaminant loads are still being delivered to the impacted region of the Finniss River over the annual cycle, the recovery in fish diversity and abundances is consistent with (a) reductions of in situ contaminant water concentrations at the time of fish sampling, (b) reductions in annual-cycle contaminant loads of sulfate, Cu, Zn, and Mn by factors of 3-7, (c) greatly reduced frequencies of occurrence and magnitude of elevated contaminant water concentrations over the annual cycle, that was most pronounced for Cu, and (d) the absence of extensive fish-kills during the first-flushes of contaminants into the Finniss river proper at the beginning of the wet season, that were observed prior to remediation. As such, the results indicate that there has been ecological benefit to the Finniss River attributable to remedial works undertaken at the Rum Jungle mine site. Recovery in abundances of these fishes may also be due to their time-dependent evolution of tolerance to mine-waste contaminants over their long period of exposure.  相似文献

In Experiment 1, water-deprived rats had 5-min access to saccharin followed by active or yoked intravenous delivery of saline or cocaine (0.33 mg/infusion). Both cocaine groups avoided intake of the saccharin cue following saccharin–cocaine pairings; however, the rats in the yoked condition exhibited greater avoidance of the taste cue than did the actively administering rats. Experiment 2 evaluated subsequent self-administration behavior on fixed- and progressive-ratio schedules of reinforcement. The results showed that prior yoked exposure to cocaine reduced subsequent drug-taking behavior on a progressive-ratio but not on a fixed-ratio schedule. Finally, Experiment 3 used a choice test to determine the impact of yoked drug delivery on the relative preference for cocaine versus water. The results showed that rats with a history of self-administering cocaine preferred to perform operant behaviors on the side of the chamber previously paired with cocaine, whereas the rats with a history of yoked delivery of cocaine avoided this side. These data show that, in most rats, the unpredictable, uncontrollable delivery of cocaine protects against the subsequent motivation for cocaine through an aversive mechanism. PURPOSE: Keratoconus is characterized by thinning and scarring of the central portion of the cornea. This study was performed on keratoconus corneas to examine the expression of proteins related to wound healing including vimentin, an intermediate filament protein, and tenascin, and extracellular matrix protein. The expression of stress-related cytokines, heat shock proteins and ubiquitin was also investigated. METHODS: Corneal buttons were collected from patients with keratoconus, normal subjects and patients with other corneal diseases such as pseudophakic bullous keratopathy. Immunofluorescence staining was performed on frozen sections for vimentin and tenascin, and immunoperoxidase staining was carried out on paraffin sections for cytokines, heat shock proteins and ubiquitin. RESULTS: To varying degrees, all proteins examined, except tenascin and heat shock protein 90, were found to be expressed in normal human corneas. The expression of vimentin, tenascin, transforming growth factor-beta, interleukin-1, heat shock protein 27, and ubiquitin was enhanced in keratoconus corneas. A similar enhancement however was also observed in other diseased corneas. CONCLUSIONS: Altered expression of several wound healing or stress-related proteins was noted in keratoconus corneas. The alterations appear to be nonspecific injury or wound responses in association with corneal diseases.  相似文献

Validated suboutcome measures that represent intermediate links between more molecular in-session changes and ultimate outcome. The present study involved the collection of pre- and postsession ratings from 53 patients in a 20-session protocol of cognitive therapy, which yielded anxiety shift, depression shift, cognitive shift, optimism shift, and therapeutic alliance. From a series of regression analyses of repeated measures with a generalized estimating equations approach, results regarding the predictive relationship of these variables to a number of patient and therapist-rated outcome criterion variables indicated that change in cognition and quality of the therapeutic alliance were the strongest predictors.
