The influence of two peroxides (peroxydicarbonate/dilauroyl peroxide) with various concentrations (10–200 mmol/kg PP) and their effective opportunity to introduce long chain branched (LCB) were investigated. The dependence of a single and double extrusion step and the changes of the properties were studied. Experiments were carried out in a single screw extruder at 180°C for the first extrusion step (modification) and at 240°C for the second extrusion step (processing simulation). Melt flow rate and dynamic rheological properties were studied at a measuring temperature of 230°C. For the definitive determination of long chain branched polypropylene (LCB-PP) served the extensional rheology measurements. The mechanical properties were examined via tensile test and impact tensile test. Summarized, LCB (melt strength) could be observed via extensional rheology for all modified specimens and the mechanical properties were maintained or even improved for the modified samples. Particularly, samples containing dilauroyl peroxide display excellent mechanical properties in this study.  相似文献

The treatment of cancer with accelerator beams has a long history with betatrons, linacs, cyclotrons and now synchrotrons being exploited for this purpose. Treatment techniques can be broadly divided into the use of spread-out beams and scanned ‘pencil’ beams. The Bragg-peak behaviour of hadrons makes them ideal candidates for the latter. The combination of precisely focused ‘pencil’ beams with controllable penetration (Bragg peak) and high, radio-biological efficiency (light ions) opens the way to treating the more awkward tumours that are radio-resistant, complex in shape and lodged against critical organs. To accelerate light ions (probably carbon) with pulse-to-pulse energy variation, a synchrotron is the natural choice. The beam scanning system is controlled via an on-line measurement of the particle flux entering the patient and, for this reason, the beam spill must be extended in time (seconds) by a slow-extraction scheme. The quality of the dose intensity profile ultimately depends on the uniformity of the beam spill. This is the greatest challenge for the synchrotron, since slow-extraction schemes are notoriously sensitive. BACKGROUND: Organic milk production aims at efficient use of home‐grown feeds, especially forages, to minimise the quantity of purchased feeds. In conventional agriculture, Italian ryegrass (Lolium multiflorum Lam.) is known for its high energy content and palatability, and the aim of the present study was to examine its suitability as feed for organic dairy cows. Therefore a feeding trial was conducted comparing a diet including 50% (of silage dry matter (DM)) of Italian ryegrass silage with a control diet based on grass/clover silage (a mixture of the second cut from permanent grassland and the second cut from a perennial clovergrass ley) alone. RESULTS: Inclusion of Italian ryegrass silage in the diet increased forage intake significantly (14.5 vs 13.4 kg DM in the control group) and concentrate intake did not differ, but milk yield was slightly lower (20.3 vs 21.0 kg) owing to the low energy and protein concentration of Italian ryegrass silage. CONCLUSION: Italian ryegrass was indeed found to be highly palatable, confirming in principle its suitability as feed for organic dairy cows. However, higher energy and protein concentrations in this forage would be necessary to translate the high intakes of Italian ryegrass silage into improved milk production as well. Parathyroid hormone-related protein (PTHrP) is a causative factor of humoral hypercalcemia in breast cancer and other malignancies. We studied circulating PTHrP levels with three different immunoassays directed against different parts of the PTHrP molecule in 48 patients with breast cancer and eucalcemia. The methods used were: (a) a RIA with antibodies directed toward the midregion (63-78); (b) an immunofluorometric assay with two antibodies against 1-34 and 38-67; and (c) an immunoradiometric assay with antibodies against 1-40 and 1-72. Although most patients had PTHrP levels indistinguishable from normal when measured by all three methods, four patients had increased serum levels in the IFMA. PTHrP was detected by immunohistochemistry in tumors from nearly all patients. One patient with elevated PTHrP in plasma measured by IFMA showed intense staining of tumor by immunohistochemistry; the tumor was histologically graded as III (severe) and was the largest of all tumors in this patient group. The IFMA can identify increased serum PTHrP in some patients with breast cancer who are not hypercalcemic. This assay may be especially useful in screening patients for this tumor during a relative early phase of the disease.  相似文献

The Ca2+-sensitive K+ channel (K(Ca) channel) plays a key role in buffering pressure-induced constriction of small cerebral arteries. An amplified current through this channel has been reported in vascular smooth muscle cells obtained from hypertensive animals, implying that the expression or properties of K(Ca) channels may be regulated by in vivo blood pressure levels. In this study, we investigated this hypothesis and its functional relevance by comparing the properties, expression levels, and physiological role of K(Ca) channels in cerebral resistance arteries from normotensive and genetically hypertensive rats. Whole-cell patch-clamp experiments revealed a 4.7-fold higher density of iberiotoxin-sensitive K(Ca) channel current at physiological membrane potentials in spontaneously hypertensive rat (SHR) compared with Wistar-Kyoto (WKY) rat cerebrovascular smooth muscle cells (n = 18 and 21, respectively). However, additional single-channel analysis in detached patches showed similar levels of unitary conductance, voltage, and Ca2+ sensitivity in K(Ca) channels from WKY and from SHR membranes. In contrast, Western analysis using an antibody directed against the K(Ca) channel alpha-subunit revealed a 4.1-fold increase in the corresponding 125-kD immunoreactive signal in cerebrovascular membranes from SHR compared with WKY rats. The functional impact of this enhanced K(Ca) channel expression was assessed in SHR and WKY rat pial arterioles, which were monitored by intravital microscopy through in situ cranial windows. Progressive pharmacological block of K(Ca) channels by iberiotoxin (0.1 to 100 nmol/L) dose-dependently constricted pial arterioles from SHR and WKY rats (n = 6 to 8). The arterioles in SHR constricted 2- to 4-fold more intensely, and vasospasm occurred in some vessels. These data provide the first direct evidence that elevated levels of in situ blood pressure induce K(Ca) channel expression in cerebrovascular smooth muscle membranes. This homeostatic mechanism may critically regulate the resting tone of cerebral arterioles during chronic hypertension. Furthermore, the overexpression of distinct K+ channel types during specific cardiovascular pathologies may provide for the upregulation of novel disease-specific membrane targets for vasodilator therapies.  相似文献

Adrenal glucocorticoids exert powerful effects on cellular excitability in neuroendocrine cells and neurons, although the underlying mechanisms are poorly understood. In metabolically intact mouse anterior pituitary corticotrope (AtT20) cells glucocorticoid-induced proteins render large conductance calcium-activated potassium (BK) channels insensitive to inhibition by protein kinase A (PKA). In this study we have addressed whether this action of glucocorticoids is mediated via protein phosphatase activity at the level of single BK channels. In isolated inside-out patches from control AtT20 cells BK channels (125 pS) were inhibited by activation of closely associated PKA. Pretreatment (2 h) of cells with 1 microM dexamethasone before patch excision did not modify the intrinsic properties or expression levels of BK channel alpha-subunits in AtT20 cells. However, PKA-mediated inhibition of BK channel activity in isolated patches from steroid-treated cells was severely blunted. This effect of steroid was not observed using adenosine 5'-O-(3-thiotriphosphate) as phosphate donor or on exposure of the intracellular face of the patch with 10 nM of the protein phosphatase inhibitors okadaic acid or calyculin A but was mimicked by application of protein phosphatase 2A (PP2A) to the intracellular face of patches from control cells. Glucocorticoids did not modify total PP2A activity in AtT20 cells, suggesting that modified PP2A-like phosphatase activity closely associated with BK channels is required for glucocorticoid action.  相似文献

The tremendous increase in the lactation performance of dairy cows during the last 60 years has had dramatic consequences on fertility and vitality (fitness) of the animals. Unprecedented average lactation performance levels have resulted in an equally unprecedented reduction in longevity. Economic pressure to further reduce the age at first calving is still high, although the negative correlation between precocious maturity and longevity is well documented in the relevant literature. Ever more aggressive selection and feeding measures are being taken to shorten the rearing time of heifers in order to reduce the cost of replacement heifers. An additional decrease in the stayability of cows has to be expected when the age at first calving is lowered even further. For reasons of profitability, dairy cows should complete an average of at least four lactations. Cows from the most important dairy breeds in the United States and Bavaria, Germany, already dropped below that crucial threshold many years ago. The same phenomenon has been observed in dairy cows in Austria over the past few years. Holstein cows that were culled in Austria in 2007 had an average of only 3.3 parities. To improve this situation, dairy cattle breeding programs should put a much higher emphasis on selection for traits of longevity and lifetime performance and, in return, markedly reduce the emphasis on selection for lactation performance in general and early‐lactation performance test results in particular. Over millions of years of evolutionary adaptation, cattle have acquired the ability to utilize fibrous low‐quality plant substrate. Only a strongly forage‐based feeding regimen is, in the long run, in accordance with their digestive and metabolic configuration. The massive use of concentrates (mostly grain) in cattle rations increases the risk of digestive and metabolic disorders in cattle and consequently leads to impaired animal welfare. Finally, the careful management of scarce resources demands that a closer look be taken at the food balance in dairy rations, i.e., an awareness of how much food that is edible for humans is being fed to livestock. OBJECTIVE: To investigate the ability of the Glasgow Coma Scale score to predict hospital mortality rate for adult medical-surgical intensive care unit (ICU) patients without trauma. DESIGN: A prospective cohort analysis of adult medical-surgical patients from a nationally representative sample of 40 U.S. hospitals. PATIENTS: 15,973 consecutive, nontraumatic ICU admissions and a comparison group of 687 head trauma admissions. INTERVENTIONS: None. MEASUREMENTS AND MAIN RESULTS: Patients' gender, age, treatment location before ICU admission, comorbidities, admission diagnosis, daily physiologic measurements, Glasgow Coma Scale score, Acute Physiology and Chronic Health Evaluation (APACHE III) score, subsequent hospital mortality rate, and unit-specific sedation practices were noted. Hospital mortality rates were stratified by the first ICU day Glasgow Coma Scale score for all admissions. The relationship between the Glasgow Coma Scale score and outcome for two high mortality medical diagnoses (post-cardiac arrest and sepsis) were also examined and compared to the relationship found in patients with head trauma. The Glasgow Coma Scale score on ICU admission had a highly significant (r2 = .922, p < .0001) but nonlinear relationship with subsequent outcome in ICU patients without trauma. Discrimination of patients into high- or low-risk prognostic groups was good, but discrimination in the intermediate levels (Glasgow Coma Scale score of 7 to 11) was reduced. This relationship varied within the operative and nonoperative groups, and also within different disease categories, various age groups, and certain ranges of the Glasgow Coma Scale score. A reduced initial Glasgow Coma Scale score associated with sepsis was a combination of factors associated with a higher mortality rate than that found in patients with head trauma. The proportion of patients who could not be assigned a Glasgow Coma Scale score because of sedation/paralysis varied widely across ICUs. The overall predictive capability of the APACHE III Prognostic Scoring System was improved by incorporating the Glasgow Coma Scale score. CONCLUSIONS: We demonstrated the prognostic importance of admission levels of consciousness as measured by the Glasgow Coma Scale score on ICU and hospital mortality rates. We concluded that the Glasgow Coma Scale score may be used to stratify and predict mortality risk in general intensive care patients, but lack of sensitivity in the intermediate range of Glasgow Coma Scale Score should be noted. Ideally, the Glasgow Coma Scale score should also be applied in the context of other physiologic information and the patient's specific diagnosis. Variation in the use of sedatives in different ICUs means that imputing or substituting a value other than normal for an unobtainable Glasgow Coma Scale score may introduce a substantial treatment bias into subsequent outcome predictions.  相似文献
