Ubiquitin-mediated proteolysis controls the abundance of many cell cycle regulatory proteins. Recent work in Saccharomyces cerevisiae suggests that a complex consisting of Cdc53, Skp1, and a third component known as an F-box protein (termed SCF) in combination with Cdc34 specifically targets regulatory proteins for degradation, and that substrate specificity is likely to be mediated by the F-box subunit. A screen for genetic interactions with a cdc34 mutation yielded MET30, which encodes an F-box protein. MET30 is an essential gene required for cell cycle progression and met30 mutations interact genetically with mutations in SCF components. Furthermore, physical interactions between Met30, Cdc53, Cdc34, and Skp1 in vivo provide evidence for an SCFMet30 complex. We demonstrate the involvement of Met30 in the degradation of the Cdk-inhibitory kinase Swe1. Swe1 is stabilized in met30 mutants and GST-Met30 pull-down experiments reveal that Met30 specifically binds Swe1 in vivo. Furthermore, extracts prepared from cdc34 or met30 mutants are defective in polyubiquitination of Swe1. Taken together, these data suggest that SCF-mediated proteolysis may contribute to the regulation of entry into mitosis. Our data, in combination with previously published results, also provide evidence for distinct SCF complexes in vivo and support the idea that their F-box subunits mediate SCF substrate specificity.  相似文献

We have used cross-sectional scanning tunneling microscopy (STM) to study interface structure in arsenide/phosphide heterostructures grown by gas-source molecular beam epitaxy (GSMBE) and by low-pressure metalorganic vapor phase epitaxy (LP-MOVPE). High-resolution images of GSMBE samples consisting of GaAs interrupted at 200Å intervals with a 40 s P2 flux reveal substantial, growth-temperature-dependent incorporation of phosphorus with nanometer-scale lateral variations in interface structure. STM images of InGaAs/InP multiple quantum well structures grown by LP-MOVPE show evidence of interface asymmetry and extensive atomic cross-incorporation at the interfaces. Data obtained by STM have been corroborated by high-resolution x-ray diffraction and reflection high-energy electron diffraction. Together, these studies provide direct information about nanometer-scale grading and lateral nonuniformity of arsenide/phosphide interfaces that can occur under these growth conditions.  相似文献

The restriction fragment length polymorphisms (RFLPs) of 64 random and potentially related strains of Listeria monocytogenes were analysed and compared using a probe comprised of two L. monocytogenes chromosome fragments cloned into a lambda vector. Twelve RFLP types were defined using 14 isolates of clinical origin, 42 food isolates and eight food associated environmental strains. Of the RFLP types, some were common to a particular serovar and source, whereas others were widespread amongst all serovars and sources. One of the two most common RFLP patterns was associated with serovar 1/2 isolates from food or the environment, whereas another dominant pattern was associated most commonly with serovar four isolates from all sources. The potential relationships between epidemiologically related strains were examined, with the analysis of types from a suspected listeriosis outbreak, from clinical maternal-foetal cases, and from an ice-cream factory environmental study. Serotyping alone was not a sufficient marker for the comparison of these strains whereas further discrimination of strains was possible with RFLP analysis.  相似文献

Recently, graphics processing units (GPUs) have had great success in accelerating many numerical computations. We present their application to computations on unstructured meshes such as those in finite element methods. Multiple approaches in assembling and solving sparse linear systems with NVIDIA GPUs and the Compute Unified Device Architecture (CUDA) are created and analyzed. Multiple strategies for efficient use of global, shared, and local memory, methods to achieve memory coalescing, and optimal choice of parameters are introduced. We find that with appropriate preprocessing and arrangement of support data, the GPU coprocessor using single‐precision arithmetic achieves speedups of 30 or more in comparison to a well optimized double‐precision single core implementation. This paper presents a scalable parallel variational time integration algorithm for nonlinear elastodynamics with the distinguishing feature of allowing each element in the mesh to have a possibly different time step. Furthermore, the algorithm is obtained from a discrete variational principle, and hence it is termed parallel asynchronous variational integrator (PAVI). The underlying variational structure grants it outstanding conservation properties. Based on a domain decomposition strategy, PAVI combines a careful scheduling of computations with fully asynchronous communications to provide a very efficient methodology for finite element models with even mild distributions of time step sizes. Numerical tests are shown to illustrate PAVI's performance on both slow and fast networks, showing scalability properties similar to the best parallel explicit synchronous algorithms, with lower execution time. Finally, a numerical example in which PAVI needs ≈100 times less computing than an explicit synchronous algorithm is shown. Copyright © 2006 John Wiley & Sons, Ltd.  相似文献

By serving as host recipients of xenografts from both humans and animals, severe combined immunodeficient (SCID) mice have become valuable to many laboratories interested in examining the pathophysiology of different diseases. To gain insight into the usefulness of the SCID mutation in retrovirus research, rhesus monkey fetal hematolymphoid tissues (liver and thymus) were used to construct a SCID-rhesus chimeric mouse (SCID-rh) and were engrafted in the renal capsule. The size and maturation of the thymic engrafts were monitored grossly, histologically, and immunologically. SCID mice were tolerant to rhesus tissues, and thymic engrafts contained thymocytes at different stages of maturation and differentiation that had morphologic features similar to age-matched rhesus thymus. Mature single positive CD2+, CD4+, and CD8+ T lymphocytes that were phenotypically similar to rhesus T lymphocytes were present at low levels (2% to 5%) in the peripheral blood and at moderately higher levels (7% to 15%) in the spleens of SCID-rh mice obtained between 12 and 15 weeks after thymus/liver engraftment. Within 3 weeks after engraftment, > 85% of the thymocytes in the thymic engrafts were immature double positive CD4+CD8+ T cells. The highest number of positive cells were seen in thymic engrafts obtained at 12 to 18 weeks. During these weeks, > 90% of the cells were double positive (CD2+CD4+, CD2+CD8+, and CD4+CD8+). After infection of the engrafted thymus tissue with simian immonodeficiency virus (SIVmac239), PCR analysis revealed successful viral infection of engrafts at 2 and 4 weeks after infection. No significant histopathologic and flow cytometric changes were observed in the thymic engrafts at 2 and 4 weeks after infection. An unrelated lesion of thymic lymphomas involving the SCID host thymus was seen in 12% of the mice. The data presented herein suggest that the SCID-rh is a valuable model for specific studies related to thymus-retrovirus interaction and that it could be used for further studies. The results are discussed in relation to current knowledge of thymus involvement during simian and human immunodeficiency virus infection.  相似文献

The IEA published “Energy Technology Perspectives” (ETP) in June 2008. That document reports on IEA scenarios for baseline and low-CO2 alternative scenarios to 2050, across the energy economy. The study included creating scenarios for transport, using the IEA Mobility Model (MoMo). This paper reports on the transport-related ETP scenarios and describes the model used in the analysis. According to the ETP Baseline scenario, world transport energy use and CO2 emissions will more than double by 2050. In the most challenging scenario, called “BLUE”, transport emissions are reduced by 70% in 2050 compared to their baseline level in that year (and about 25% below their 2005 levels). There are several versions of the BLUE scenario, but all involve: a 50% or greater improvement in LDV efficiency, 30–50% improvement in efficiency of other modes (e.g. trucks, ships and aircraft), 25% substitution of liquid fossil fuels by biofuels, and considerable penetration of electric and/or fuel-cell vehicles. Tetralogy of Fallot was diagnosed in an acyanotic 11-month-old dog. Predicted pressure gradient across the pulmonic valve, as assessed by use of continuous wave Doppler echocardiography, was 94.5 mm Hg. Bidirectional shunting was identified by means of selective angiography. Open-heart correction was performed, using a transatrial approach with limited ventriculotomy and cardiopulmonary bypass. The hypertrophied infundibulum was resected, the ventricular septal defect was closed primarily, and a transannular pericardial patch graft was applied. Pressure gradients across the pulmonic valve were 52.9 and 22.8 mm Hg 2 weeks and 4 months after surgery, respectively. Advances in cardiopulmonary bypass, anesthetic management, and use of the transatrial approach may improve the success of open-heart correction of tetralogy of Fallot in dogs.  相似文献

Plants are naturally abundant and display high sensitivity to ecological factors to thrive in diverse environmental conditions. As sessile organisms, they have evolved complex, internal, and interplant signaling pathways with distinct structures to promptly adjust to the constantly changing environment. In the past five years, the unique ways in which they exchange information with and function in the environment have inspired an emerging field of plant nanobionics, which describes the interface between living plants and nanotechnology to impart the former with novel and useful functions. The structural merits of plant organs and organelles have also inspired the creation of plant‐derived structures through biointerfacing with nanoparticles containing electronic and optical properties. Here, the emerging applications and vision of plant nanobionics are highlighted together with related plant‐inspired materials in potentially replacing the myriad devices in the everyday lives stamped out of plastic, containing circuit boards and consuming power from the electrical grid. Applications in environmental sensing, communication devices, and energy harvesting and conversion are comprehensively discussed.  相似文献
