Alginate as a versatile naturally occurring biomaterial has found widespread use in the biomedical field due to its unique features such as biocompatibility and biodegradability. The ability of its semipermeable hydrogels to provide a favourable microenvironment for clinically relevant cells made alginate encapsulation a leading technology for immunoisolation, 3D culture, cryopreservation as well as cell and drug delivery. The aim of this work is the evaluation of structural properties and swelling behaviour of the core-shell capsules for the encapsulation of multipotent stromal cells (MSCs), their 3D culture and cryopreservation using slow freezing. The cells were encapsulated in core-shell capsules using coaxial electrospraying, cultured for 35 days and cryopreserved. Cell viability, metabolic activity and cell–cell interactions were analysed. Cryopreservation of MSCs-laden core-shell capsules was performed according to parameters pre-selected on cell-free capsules. The results suggest that core-shell capsules produced from the low viscosity high-G alginate are superior to high-M ones in terms of stability during in vitro culture, as well as to solid beads in terms of promoting formation of viable self-assembled cellular structures and maintenance of MSCs functionality on a long-term basis. The application of 0.3 M sucrose demonstrated a beneficial effect on the integrity of capsules and viability of formed 3D cell assemblies, as compared to 10% dimethyl sulfoxide (DMSO) alone. The proposed workflow from the preparation of core-shell capsules with self-assembled cellular structures to the cryopreservation appears to be a promising strategy for their off-the-shelf availability.  相似文献

Innovative amperometric biosensors for monitoring the level of wastewater pollution have been constructed on the surface of the gold planar electrodes C220AT “DropSens” by using the organic–inorganic ureasil‐based composites as host matrixes and immobilized commercial laccase from Trametes versicolor . It was found that the biosensor based on the ureasil–chalcogenide glass composite is characterized by a very high sensitivity (67,540 А M?1 m?2) that is 38.3 times higher than for pure ureasil (the sensitivity of the bioelectrode was calculated as 1762 А M?1 m?2). On the other hand, application of the ureasil–chalcogenide glass composite with incorporated silver nanoparticles (NPs) synthesized by high‐dose (1.0 × 1017 Ag+/cm2) 30 keV Ag+ ion implantation results in decreasing the biosensor sensitivity up to 2390 times (the sensitivity of the bioelectrode was 28.3 А M?1 m?2). The role of additives (chalcogenide glass and silver NPs) in the ureasil matrix on the biofunctionality of the biosensors produced is considered. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. The effect of the chemical composition and heat treatment on the microstructure and abrasive wear resistance of V-Mn, V-Ni-Cr, and V-Mo spheroidal vanadium carbide cast irons (18–23 vol %) has been studied. The wear resistance has been determined under conditions of wear by abrasives with various hardnesses, i.e., corundum and quartz and compared to that of high-chromium cast iron with 13% Cr. It has been found that the advisability of using high-vanadium cast irons is governed by the hardness of the abrasive. When a hard abrasive, i.e., corundum was used, V-Mo cast iron with the maximum concentration of spheroidal VC carbides, which were uniformly distributed in the martensitic matrix, had the highest wear resistance. When a soft abrasive, i.e., quartz, was applied, high-chromium cast iron with a hardness of 68 HRC, which contained the largest amount of M7C3 carbides, was more wear-resistant. In the course of isothermal exposure at 300–1000°C, V-Ni-Cr and V-Mo cast irons with an austenitic structure had high resistance to phase and structural transformations. However, the properties and microstructure of V-Mo cast irons with a martensitic matrix depended strongly on the temperature of exposure during heat treatment.  相似文献

The results of an investigation of the effect of bulk quenching from temperature in the range of 760–1050°C, cryogenic treatment (–196°C) and surface plasma hardening on the abrasive-erosion wear of frugally alloyed V–Cr–Mn–Ni cast irons with spheroidal vanadium carbides have been presented in this article. It has been found that cast irons containing 5.0–7.5% V, 4.5–9.0% Cr, and 5.5–5.7% (total) of Mn and Ni after heat treatment have a 2–3-fold advantage in wear resistance compared to the prototype high-vanadium cast iron (11.9% V, 12.9% Mn). The maximum wear resistance of cast irons studied is achieved by quenching at 760°C followed by plasma surface hardening, as well as quenching at 840°C, followed by cryogenic treatment. These treatments result in the formation of an optimum microstructure that consists of spheroidal vanadium carbides, eutectic carbides M7C3, and a martensite-austenite matrix reinforced by secondary carbides. The increase in quenching temperature leads to an increase in the amount of residual austenite and decrease in the erosive wear resistance of cast irons.  相似文献

The safety of food additives E407 and E407a has raised concerns in the scientific community. Thus, this study aims to assess the local and systemic toxic effects of the common food additive E407a in rats orally exposed to it for two weeks. Complex evaluations of the effects of semi-refined carrageenan (E407a) on rats upon oral exposure were performed. Local effects of E407a on the intestine were analyzed using routine histological stains and CD68 immunostaining. Furthermore, circulating levels of inflammatory markers were assessed. A fluorescent probe O1O (2- (2′-OH-phenyl)-5-phenyl-1,3-oxazole) was used for evaluating the state of leukocyte cell membranes. Cell death modes of leukocytes were analyzed by flow cytometry using Annexin V and 7-aminoactinomycin D staining. Oral administration of the common food additive E407a was found to be associated with altered small and large intestinal morphology, infiltration of the lamina propria in the small intestine with macrophages (CD68+ cells), high systemic levels of inflammation markers, and changes in the lipid order of the phospholipid bilayer in the cell membranes of leukocytes, alongside the activation of their apoptosis. Our findings suggest that oral exposure to E407a through rats results in the development of intestinal inflammation.  相似文献
