There is emerging evidence of the efficiency of probiotic, prebiotic and synbiotic treatments in inflammatory bowel diseases (IBDs) and one of their long-term complications, colorectal cancer (CRC). In this study, various strains of probiotic lactic acid bacteria, prebiotic glucooligosaccharides (GOS) or a synbiotic combination of the two were screened for anti-inflammatory and anti-proliferative effects in different in vitro models in the context of such diseases. To mimic IBD response to Gram negative bacteria, HT-29 cells were sensitised to inflammatory response to lipopolysaccharide (LPS) by IFNγ which increased expression of TLR4, the LPS biosensor, and were then treated by probiotics, prebiotics and synbiotics. Secreted IL-8 and activated NF-κB were monitored as inflammation biomarkers. A selection of active strains were then subjected to a second inflammatory cell culture model consisting of inflammatory activated transgenic Caco-2 cells transfected by a reporter gene under the control of NF-κB inducible promoter. Quantification of reporter gene expression allowed us to demonstrate some probiotic inhibitory properties or to confirm such characteristics in two different models. Proliferation of cancerous HT-29 cells was monitored by XTT assay. Only three probiotic strains induced a proliferation decrease, but with a lack of reproducibility. Binary or ternary probiotic associations, complemented or not by prebiotic GOS, significantly decreased proliferation, especially with a synbiotic association of Bifidobacterium breve, Lactococcus lactis and oligoalternan, a GOS. This combination was selected for the following experiments. We showed the involvement of both bacterial and carbohydrate compounds of this synbiotic in the observed effect by dose range tests. We demonstrated that this decrease in proliferation may be due to an induction of a differentiated phenotype, as shown by the up-regulation of intestinal alkaline phosphatase, a biomarker of differentiation, monitored by real-time RT-PCR in HT-29 cells treated by the selected synbiotics. Thus, this study demonstrates the ability of probiotics to exert anti-inflammatory effects and shows some anti-proliferative characteristics for a specific synbiotics. These products should be further evaluated in animal models to confirm the in vitro results.  相似文献

The primary goal of this study was to determine the residual nitrite-reducing effects of glucono-δ-lactone (GDL) and ascorbic acid (AA) in Turkish-type fermented sausage. Response surface methodology was used to investigate the simultaneous effects of processing variables, e.g., nitrite (0–200 mg/kg sausage), GDL (0–0.5% w/w) and AA (0–500 mg/kg sausage) on pH, moisture, residual nitrite, proteolysis, lipolysis, peroxide, yield and sensory score of the sausage. AA significantly ( P <  0.01) decreased the residual nitrite level; however, quadratic effect of AA showed an inverse effect. GDL did not produce the same effect on the residual nitrite level. The processing variables had no prominent effect on the proteolysis, lipolysis and peroxide. GDL and AA addition showed a negative effect on the yield and sensory scores of the sausages. The regression model predicted the optimum residual nitrite content to be 0.75 mg/kg. The critical values for nitrite, GDL and AA to obtain this optimum level were 147.56 mg/kg, 0.17% and 565 mg/kg, respectively.

PRACTICAL APPLICATIONS


Residual nitrite level is a major problem in the Turkish-type fermented sausage product with respect to some health concerns, residual nitrite causes a possible formation of the carcinogenic N -nitrosamines. The results of this study can be applied to reduce the residual nitrite level in sausage and to find the optimum levels of nitrite, glucono-δ-lactone and ascorbic acid levels to maintain the characteristic sausage quality properties. Based on the results, ascorbic acid can be used to reduce the residual nitrite levels in the sausage.  相似文献

Various aspects of enzyme-catalysed racemate resolution arediscussed by using examples from hydrolysis of butanoates ofglycerol derivatives. Primary esters of various 1,2-ketals gavelow enantioselectivity (E). The results with secondary estersvaried. The highest E value (> 100) was obtained when theprimary ether groups were methyl and phenylethyl. For the correspondingmethyl, phenyl diether E was observed to increase as the hydrolysisproceeded. Tungsten (W) is an important material in tokamak walls and divertors.The W ion charge state distribution and the dynamic behavior of ions play important roles in the investigation of plasma-wall interactions using laser-ablation-based diagnostics such as laser-induced breakdown spectroscopy and laser-induced ablation spectroscopy.In this work,we investigate the temporal and spatial evolutions of differently charged ions in a nanosecond-laser-produced W plasma in vacuum using time-of-flight mass spectroscopy.Ions with different charge states from 1 to 7 (W+ to W7+) are all observed.The temporal evolutions of the differently charged ions show that ions with higher charge states have higher velocities,indicating that space separation occurs between the differently charged ion groups.Spatially-resolved mass spectroscopy measurements further demonstrate the separation phenomenon.The temporal profile can be accurately fitted by a shifted Maxwell-Boltzmann distribution,and the velocities of the differently charged ions are also obtained from the fittings.It is found that the ion velocities increase continuously from the measured position of 0.75 cm to 2.25 cm away from the target surface,which indicates that the acceleration process lasts through the period of plasma expansion.The acceleration and space separation of the differently charged ions confirm that there is a dynamic plasma sheath in the laser-produced plasma,which provides essential information for the theoretical laser-ablation model with plasma formation and expansion.  相似文献

This research focuses on the investigation of remote-sensing techniques for the detection of coastal sub-aerial springs and submarine groundwater discharges using airborne thermal and hyperspectral imagery. Very high spatial resolution thermal and hyperspectral images were acquired using Thermal Airborne Broadband Imager 320 (TABI-320) and Compact Airborne Spectrographic Imager 550 (CASI-550) sensors. Extensive in situ spectroradiometer and oceanographic measurements were carried out in parallel with thermal and hyperspectral image acquisitions. Experiments and analysis of the data show that the combined use of very high spatial resolution airborne thermal and hyperspectral sensors for the detection of relatively small sub-aerial coastal springs and submarine groundwater discharges proves to be a very efficient and operational method. Very high spatial resolution thermal data were able to detect even very small coastal sub-aerial springs. On the other hand, the hyperspectral data were the most appropriate for detecting relatively small submarine groundwater discharges, which were not detected on thermal imagery, due to the increase in turbidity that these discharges cause. This is confirmed by the strong correlations between the hyperspectral data and the in situ measured turbidity-related water inherent optical properties.  相似文献

Cast shadows caused by sparse clouds usually degrade spaceborne and airborne imagery. They result from the decrease of the direct solar beam due to the presence of a non-transparent cloud. The reduction of the downwelling solar flux density can be quantified during an air campaign, if the aircraft flies beneath the cloud and is equipped with an add-on instrument that measures the total downwelling solar irradiance. The objective of this work is to exploit such data for the de-shadowing of airborne hyperspectral imagery. Initially, the specific illumination and viewing conditions during the image acquisition, which allow the use of at-sensor downwelling irradiance data for the de-shadowing of airborne hyperspectral imagery, are outlined. Then a methodology is proposed that estimates the radiometric enhancement coefficients from the at-sensor irradiance data and correlates them with the image data using a shadow map. Improvements of the quality of the shadow maps are suggested. Performance assessment showed that at-sensor irradiance data could be satisfactorily utilized for compensating the cast shadow effects on remotely sensed imagery. It also highlighted the importance of generating and using an accurate shadow map and the particular difficulties for the air campaign planning raised by the requirement of exploitable at-sensor irradiance data.  相似文献

Al-substituted Li7La3Zr2O12 samples processed under argon show enhanced Li-ion transport and interfacial properties in symmetrical cells with lithium electrodes, compared to those prepared in air. In particular, the samples prepared under argon have higher ionic conductivities and lower interfacial impedances in symmetrical lithium cells, and show better DC cycling characteristics. The electronic conductivities are also somewhat higher. Pellets subjected to thermal treatment under the two types of atmospheres have different colors but exhibit similar microstructures. X-ray diffraction experiments suggest that there are slight structural differences between the two types of samples, but few dissimilarities were observed in elemental composition, distribution of ions, oxidation states, or bond lengths using laser-induced breakdown spectroscopy (LIBS), x-ray photoelectron spectroscopy (XPS), and extended x-ray absorption fine structure spectroscopy (EXAFS) to analyze the materials. Additionally, there was no evidence that La or Zr were reduced during the processing under Ar. Possible explanations for the improved electrochemical properties of the sample prepared under Ar compared to the one prepared in air include differences in grain boundary chemistries and conductivities and/or a small concentration of oxygen vacancies in the former.  相似文献

Frozen molluscs (squid, octopuses, and cuttlefish) and crustaceans (shrimp) were irradiated using a cobalt-60 gamma source, at different doses, in order to investigate the effects of gamma radiation on their microbial population, organoleptic characteristics, lipid profile, and tropomyosin content. Irradiation of shrimp and squid with either 2.5 or 4.7 kGy reduced mesophilic bacteria contamination to low or nondetectable levels, respectively, whereas irradiation of octopus and cuttlefish with the same doses reduced the bacterial population. Irradiation treatment had no significant (P > 0.05) effect on the total lipid content and the major detected classes of polar and neutral lipids, whereas it significantly (P < 0.05) increased the contents of neutral lipids in octopus mantle and in shrimp muscle and cephalothorax samples. The total fatty acid content and the omega-3: omega-6 fatty acid ration was not affected. A dose-dependent significant (P < 0.05) decrease in the ratio of polyunsaturated fatty acids:saturated fatty acids was observed. With the increase in radiation dose, redness (a) and yellowness (b) values showed a variation, whereas the lightness (L) value was significantly (P < 0.05) decreased in mollusc mantles and shrimp muscle and increased in shrimp cephalothorax. The total of color changes ( delta E) increased (P < 0.05) as the dose increased. Significant (P < 0.05) changes in textural properties were observed with radiation treatment in octopus tentacles and in squid and cuttlefish mantle. The amount of tropomyosin, which is the major mollusc and crustacean allergen in the irradiated organisms, was reduced by gamma radiation, depending on the dose.  相似文献

A comparative study has been conducted of the major lipid classes composition, as well as the fatty acid and carotenoid content in the yolk of conventional eggs from five avian species (ostrich, turkey, quail, duck and goose); the nutritional indices were calculated. The neutral lipids were the major yolk fractions but their proportions varied among species. All yolks and especially ostrich’s yolk were found to be an excellent source of dietary lecithin. Quail yolk displayed the lowest fat and cholesterol content and the lowest values for the cholesterol index (CI) and cholesterol-saturated fat index (CSI). It is therefore more appropriate for a healthier diet. Turkey and goose yolks contained significantly (P < 0.05) higher ω-3 fatty acid proportion and ω-6/ω-3 ratio. The turkey yolk was characterised by the lowest AI and TI values, which are recommended for a healthy diet. Quail yolk lipids contained a favourable PUFA/SFA ratio. All the examined yolks contained highly bioavailable functional nutrients, such as lutein and zeaxanthin.  相似文献
