Li(+) influx by bovine chromaffin cells, obtained from bovine adrenal medulla, was studied in intact cell suspensions using (7)Li NMR spectroscopy with the shift reagent [Tm(HDOTP)](4-). The influx rate constants, k(i), were determined in the absence and in the presence of two Na(+) membrane transport inhibitors. The values obtained indicate that both voltage sensitive Na(+) channels and (Na(+)/K(+))-ATPase play an important role in Li(+) uptake by these cells. (7)Li NMR T(1) and T(2) relaxation times for intracellular Li(+) in bovine chromaffin cells provided a T(1)/T(2) ratio of 305, showing that Li(+) is highly, immobilized due to strong binding to intracellular structures. Using fluorescence spectroscopy and the Mg(2+) fluorescent probe, furaptra, the free intracellular Mg(2+) concentration in the bovine chromaffin cells incubated with 15 mM LiCl was found to increase by about mM after the intracellular Li(+) concentration reached a steady state. Therefore, once inside the cell, Li(+) is able to displace Mg(2+) from its binding sites.  相似文献

A method using normal phase high performance liquid chromatography for the analysis of olefins and diolefins in cracked petroleum samples is proposed and validated in this work. A silver-impregnated silica column is used to separate olefin and diolefin species, resulting in accurate quantification of both group-types. Separations are completed using an isocratic hexane mobile phase, with no sample clean up or backflushing required. Results from the analysis of 11 model compounds are shown, as well as quantification data for four representative samples; figures of merit are also described.  相似文献

In sequel to our recent report,[1] we demonstrate in this article the effect of chemistry on nonisothermal tempering and softening behavior of dual-phase steels. The martensite morphology and tempered structure were analyzed using electron microscopy. Electron energy loss spectrometry and energy-dispersive X-ray spectroscopy were used for a compositional analysis. It was observed that characteristic of the tempered structure in dual-phase (DP) steels was a function of prior martensite structure and chemistry, which in turn influence the extent of softening.  相似文献

Multi-mission altimeter measurements from TOPEX, Jason-1, and Jason-2 satellite altimetry over the 1993–2009 time span are used to characterize the local linear sea level trend (LSLT) around Taiwan. The results show that the long-term changes of default geophysical and range corrections, i.e. the inverted barometer correction, wet tropospheric correction, and sea state bias correction, have significant impacts on the determination of local LSLT. The trend of default corrections contribute more than 1.4 mm year?1 along the coastline of China mainland and 2.1 mm year?1 to local LSLT in the Taiwan Strait. The default-corrected altimetric data exhibit highest and lowest local LSLTs in the southeast and northwest of Taiwan, respectively. The regional LSLTs of 3.8 mm year?1 and 4.6 mm year?1 are estimated from the default-corrected and uncorrected altimetric data in the study area, respectively.  相似文献

The current study compares the antibacterial activity of zinc oxide nanostructures (neZnO). For this purpose, two bacterial strains, Escherichia coli (ATCC 4157) and Staphylococcus aureus (ATCC 29213) were challenged in room light conditions with the aforementioned materials. Colloidal and hydrothermal methods were used to obtain the quasi-round and quasi-diamond platelet-shape nanostructures. Thus, the oxygen vacancy (VO) effects on the surface of neZnO are also considered to assess its effects on antibacterial activity. The neZnO characterization was achieved by X-ray diffraction (XRD), a selected area electron diffraction (SAED) and Raman spectroscopy. The microstructural effects were monitored by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Furthermore, optical absorption ultraviolet visible spectrophotometry (UV-Vis) and X-ray photoelectron spectroscopy (XPS) analyses complement the physical characterization of these nanostructures; neZnO caused 50 % inhibition (IC50) at concentrations from 0.064 to 0.072 mg/mL for S. aureus and from 0.083 to 0.104 mg/mL for E. coli, indicating an increase in activity against S. aureus compared to E. coli. Consequently, quasi-diamond platelet-shaped nanostructures (average particle size of 377.6±10 nm) showed enhanced antibacterial activity compared to quasi-round agglomerated particles (average size of 442.8±12 nm), regardless of Vo presence or absence.  相似文献

Colorectal cancer (CRC) has been ranked as one of the cancer types with a higher incidence and one of the most mortal. There are limited therapies available for CRC, which urges the finding of intracellular targets and the discovery of new drugs for innovative therapeutic approaches. In addition to the limited number of effective anticancer agents approved for use in humans, CRC resistance and secondary effects stemming from classical chemotherapy remain a major clinical problem, reinforcing the need for the development of novel drugs. In the recent years, the phenoxazines derivatives, Nile Blue analogues, have been shown to possess anticancer activity, which has created interest in exploring the potential of these compounds as anticancer drugs. In this context, we have synthetized and evaluated the anticancer activity of different benzo[a]phenoxazine derivatives for CRC therapy. Our results revealed that one particular compound, BaP1, displayed promising anticancer activity against CRC cells. We found that BaP1 is selective for CRC cells and reduces cell proliferation, cell survival, and cell migration. We observed that the compound is associated with reactive oxygen species (ROS) generation, accumulates in the lysosomes, and leads to lysosomal membrane permeabilization, cytosolic acidification, and apoptotic cell death. In vivo results using a chicken embryo choriollantoic membrane (CAM) assay showed that BaP1 inhibits tumor growth, angiogenesis, and tumor proliferation. These observations highlight that BaP1 as a very interesting agent to disturb and counteract the important roles of lysosomes in cancer and suggests BaP1 as a promising candidate to be exploited as new anticancer lysosomal-targeted agent, which uses lysosome membrane permeabilization (LMP) as a therapeutic approach in CRC.  相似文献

Morphological parameters and renal flow were assessed and measured in six clinically healthy dogs with negative serological and parasitological examinations for canine visceral leishmaniasis (CVL) and six dogs with CVL‐positive serological and parasitological tests and asymptomatic condition of the disease. Kidney length and diameter were measured and compared with the aorta/kidney ratio. Alterations in flow were measured using the flow velocity (cm/s) and resistance index (RI). Renal function was assessed by serum biochemical examination (urea, creatine, total proteins, albumin, and globulin) and urinalysis. CVL‐positive animals presented heterogeneous kidney echotexture, reduced vascular flow, hypoperfusion, lesser visualization of the arcuate arteries, high RI values (1.06 ± 0.28 for the right kidney and 1.10 ± 0.30 for the left kidney), reduced diastolic flow, and high systolic peak. They also presented positive creatine/RI correlation and between the renal artery vascular flow and kidney diameter. Histopathological analysis of CVL‐positive animals was compatible and confirmed hemodynamic changes observed by Doppler ultrasound. The findings in the present study showed that Doppler ultrasonography is a valid method to assess and detect alterations in flow in renal pathological processes with kidney vascular damage, as in the case of leishmaniasis, and can help in the diagnosis of animals with this infection. A titanium-polyoxometalate with a µ-hydroxo dimeric structure [(PW11O39Ti)2OH]7? ((PW11Ti)2OH) was used efficiently for the desulfurization of a model diesel containing a mixture of various refractory sulfur compounds present in real fuels. The catalytic performance of the µ-hydroxo dimeric compound was compared in its homogeneous form and also when immobilized in the trimethylammonium-functionalized SBA-15 ((PW11Ti)2OH@TM-SBA-15). An optimization study was performed using the homogeneous and heterogeneous catalysts to obtain high catalytic efficiency, sustainability and cost-effectiveness of the system. Different optimized conditions were found using the homogeneous and heterogeneous catalysts. Lower amounts of solvent extraction (MeCN, 175 µL), catalyst (0.5 µmol of active center) and oxidant (50 µL) were used to produce sulfur-free model diesel after 2 h at 70?°C, using the heterogeneous (PW11Ti)2OH@TM-SBA-15 catalyst. On the other hand, complete desulfurization was achieved with homogeneous (PW11Ti)2OH catalyst after only 40 min, although higher amounts of MeCN (750 µL), catalyst (1.5 µmol) and oxidant (75 µL) were used. Both systems combined liquid–liquid extraction and catalytic oxidation. Using the heterogeneous catalyst, adsorption of oxidized-sulfur compound was also observed. Both homogeneous and heterogeneous desulfurization systems presented a high capacity to be reused/recycled for consecutive desulfurization cycles.  相似文献

Acquired immunodeficiency syndrome is a disease with profound effects on the global society, as it affects individual lives, communities, societies, and even nations. As governments try to gear up on the war against this pandemic, an issue of importance pertains to the use of information systems. The systems are used to collect data on the prevalence of the disease, and analyze and transmit the data from the lower levels of the health administration where the testing is done to the higher levels of national policy making where different kinds of interventions are designed. In this article, we argue that the manner in which the representation of the disease is constructed is an important area of concern as it shapes the picture of the disease prevalence and influences how these statistics then get used for the planning of interventions such as supplying drugs and pro-health campaigns. Drawing from the domain of Science and Technology Studies (STS), with a focus on Bruno Latour's (1999) ideas of “circulating reference” we first develop the theoretical notion of representation, and then apply it to an empirical analysis of the reporting systems, both paper- and computer-based, of HIV/AIDS in Mozambique. The successive movements of information about HIV/AIDS prevalence across the various administrative levels are seen as translations that are being constructed through the communication and work practices of the health care workers at different levels, and are shaped by the political interests of the different stakeholders involved, including international agencies and national health authorities. Latour's notion of circulating reference is drawn upon to analyze the question of what is lost, what is gained, and what remains invariant through the successive stages of translation in the construction and use of the representation. © 2005 Wiley Periodicals, Inc.  相似文献
