Here we report some recent biophysical issues on the preparation of solute-filled lipid vesicles and their relevance to the construction of “synthetic cells.” First, we introduce the “semi-synthetic minimal cells” as the liposome-based cell-like systems, which contain a minimal number of biomolecules required to display simple and complex biological functions. Next, we focus on recent aspects related to the construction of synthetic cells. Emphasis is given to the interplay between the methods of synthetic cell preparation and the physics of solute encapsulation. We briefly introduce the notion of structural and compositional “diversity” in synthetic cell populations.  相似文献

This paper presents the design and measured performance of a 1.8-GHz power amplifier featuring load mismatch protection and soft-slope power control. Load-mismatch-induced breakdown can be avoided by attenuating the RF power to the final stage during overvoltage conditions. This was accomplished by means of a feedback control system, which detects the peak voltage at the output collector node and clamps its value to a given threshold by varying the circuit gain. The issue of output power control has been addressed as well. To this end, a temperature-compensated bias network is proposed, which allows a moderate power control slope (dB/V) to be achieved by varying the circuit quiescent current according to an exponential law. The nonlinear power amplifier was fabricated using a low-cost silicon bipolar process with a 6.4-V breakdown voltage. It delivers a 33.5-dBm saturated output power with 46% maximum power-added efficiency and 36-dB gain at a nominal 3.5-V supply voltage. The device is able to tolerate a 10:1 load standing-wave ratio up to a 5.1-V supply voltage. Power control slope is lower than 80 dB/V between -15 dBm and the saturated output power level.  相似文献

To date, the majority of studies on stability of axisymmetric jets have been completed under the assumption of steady mean flow. Yet, many of the natural and man-made flows that are modeled by these jets can have an inherent unsteadiness; the effects of which on the stability and transition have not been determined. Moreover, controlled unsteadiness can be used to control stability and possibly the transition to turbulence. In this note, the effects of periodic variations of the mean flow on the stability of axisymmetric jets are examined. The problem is treated analytically. The results show that the governing equations and dispersion relation for the unsteady jet can be reduced to those governing the steady jet with a time transformation. It is shown that the periodic variations in the mean flow cause amplitude and phase modulations of the unstable modes. The implications of the modulations on the subsequent transition stages are discussed.  相似文献

The gold standard in the diagnosis of occupational asthma is the specific bronchial provocation test (sBPT), but other diagnostic criteria have been proven to have a similar sensitivity, mainly in asthma due to high molecular weight compounds. In order to assess wether some clinical findings can predict the positive response to sBPT, we studied 37 subjects (14 millers and 23 bakers) with suspected occupational asthma who underwent sBPT with wheat flour dust (dust exposure in a small cabin: geometric mean 12.1 mg/m3 for up to 30 min). A positive response to sBPT (FEV1 > 20%) was elicited in 20 subjects (11 early, 4 late, and 5 dual responses). There was no significant difference between subjects with positive or negative sBPT as regards mean age, smoking, length of employment, duration of symptoms, atopy (skin positivity to one or more common allergens) and PD20FEV1 methacholine. The percentage of subjects with work-related symptoms was significantly higher in subjects with positive sBPT with respect to subjects with negative sBPT (81% versus 41.2%, p < 0.01 by chi 2 test); furthermore, FEV1 was significantly lower in subjects with positive sBPT. The percentage of positive skin response to wheat flour extract (mean wheal diameter > or = 3 mm) was mildly but not significantly higher in subjects with positive sBPT (68.4% versus 41.2%). None of the following clinical factors (age < 35 years, asthma symptoms pre-existing occupational exposure, non smokers, atopy and bronchial hyperresponsiveness to methacholine), alone or in combination, were associated with higher prevalence of positive sBPT. We conclude that the response to sBPT in subjects with suspected occupational asthma due to flour dust can not be adequately predicted by other clinical, allergologic and functional data. Therefore, sBPT with flour dust should always be performed in subjects with suspected occupational asthma.  相似文献

A variable-gain up-conversion mixer for 5-GHz WLAN applications is presented, whose input stage is based on a novel variable gain transconductor. The proposed topology features reduced power consumption by exploiting dc current reuse for mixer biasing. Moreover, a new low-consumption control circuit is introduced, which achieves a temperature-stable and linear-in-dB characteristic, providing a 40-dB dynamic range within ±1 dB gain error.  相似文献

In this study, we report on multi-walled carbon nanotubes fabricated on silicon substrate with four different orientations via chemical vapor deposition. It is well-known that chemical treatments improve the nanotube electrochemical reactivity by creating edge-like defects on their exposed sidewalls. Before use, we performed an acid treatment on carbon nanotubes. To prove the effect of the treatment on these nanostructured electrodes, contact angles were measured. Then, sensitivities and detection limits were evaluated performing cyclic voltammetry. Two target molecules were used: potassium ferricyanide, an inorganic electroactive molecule, and hydrogen peroxide that is a product of reactions catalyzed by many enzymes, such as oxidases and peroxidases. Carbon nanotubes with tilted tips become hydrophilic after the treatment showing a contact angle of 22° ± 2°. This kind of electrode has shown also the best electrochemical performance. Sensitivity and detection limit values are 110.0 ± 0.5 μA/(mM cm2) and 8 μM for potassium ferricyanide solutions and 16.4 ± 0.1 μA/(mM cm2) and 24 μM using hydrogen peroxide as target compound. Considering the results of wettability and voltammetric measurements, nanotubes with tilted tips-based electrodes are found to be the most promising for future biosensing applications.  相似文献

In this paper, a design methodology for the optimization of transformer-loaded RF circuits is presented. The optimization procedure is based on a novel figure of merit for the integrated transformer (namely the transformer characteristic resistance), which was introduced to quantify its performance when operated as a tuned load. Using the proposed approach, a highly linear up-converter for 5-GHz wireless LAN applications was implemented in a 45-GHz-f/sub T/ SiGe HBT technology. The circuit achieved an output 1-dB compression point of 4.5 dBm and a power gain of 18 dB, while drawing only 34 mA from a 3-V power supply.  相似文献

Brazilian cacha?a can be produced in two different ways: distilled in stainless steel column or in copper alembic stills. We evaluated 36 samples of commercial non-aged cacha?as: 18 samples of sweetened cacha?as distilled in stainless steel column, and 18 samples distilled in copper alembic stills. Fingerprints were obtained through electrospray ionization mass spectrometry by recording the intensity of the 15 most abundant ions. Principal component analysis was applied to the data and separated the samples in two groups. However, after sample standardization with sugar (20?g?L?1), it was not possible to group them by type of distillation. The results showed that the technique applied did not allow differentiation of cacha?as based on the distillation system, but for the presence or absence of sugar in them.  相似文献

The effect of Maillard reaction on the mechanical properties of whey protein isolate (WPI) heat-induced gels was evaluated. WPI and dextran (15–25 kDa) conjugates were obtained by controlled dry heating during storage at 60 °C and 63% relative humidity for 2, 5 and 9 days. Changes in browning intensity and content of free amino groups were used to estimate the Maillard reaction. A decrease in free amino groups of WPI was observed when increasing polysaccharide concentration and reaction time. An increase in both a* and b* CIE Lab colour parameters indicated the development of a reddish-brown colour, typical of the Maillard reaction. Uniaxial compression and stress relaxation tests were performed to measure the mechanical properties of mixed and conjugate gels. Maillard reaction significantly modified the mechanical properties of WPI/DX gels, and even prevented fracture when conjugate gels were subjected to 80% deformation in uniaxial compression test.  相似文献
