Sediments record the history of contamination to estuaries. Analysis of the concentrations of toxic organic compounds, contaminant and crustal metals, organic carbon content and isotopic composition in sediment cores from two estuarine systems in Buzzards Bay allowed reconstruction of human impacts over 350 years. Vertical distributions of the contaminants correlate with changes in the nature of watershed/estuarine activities. All contaminants were highly enriched (tens to hundreds times background) in modern New Bedford Harbor sediments. Enrichment began around the turn of the 20th century for all but PCBs, which were first synthesized in the 1930s. An increase in organic carbon content and a shift of carbon isotopes toward a more terrestrial signature illustrates increasing anthropogenic impact in New Bedford as population grew along with the industrial base. Institution of environmental protection measures in the late 20th century was reflected in decreased, although still substantially elevated, concentrations of contaminants. A lack of industrial development in Apponagansett Bay resulted in much lower concentrations of the same indicators, although specific contaminants related to the early whaling industry increased significantly above background as early as the late 18th century. The similarity of indicators in older portions of cores from NBH and unimpacted Apponagansett Bay demonstrates that cores can be used to establish reference conditions as successfully as using separate sites judged a priori to represent the reference state. The historical reconstruction approach provides the basis for establishing relationships between environmental stressors and factors that drive the stressors, as well as a framework for the assessment of ecological response(s) to environmental stressors over a range of time and/or exposure scales.  相似文献

PURPOSE: The incidence rate of disease progression and stroke after the diagnosis of a moderate (50% to 79%) carotid stenosis was determined by means of color-flow duplex scanning. METHODS: During a 4-year period, 344 male veterans with moderate internal carotid artery stenoses, on one or both sides, were examined at regular intervals for a mean period of 25 months. Carotid color-flow scans were obtained semiannually. Clinical follow-up was performed to determine the incidence rate of amaurosis fugax, transient ischemic attacks, nonhemispheric symptoms, and strokes. RESULTS: New neurologic symptoms developed in 75 patients (21.8%). Fifty-one (14.8%) had ipsilateral symptoms during follow-up: 18 amaurosis fugax (5.2%), 14 transient ischemic attacks (4%), 5 nonhemispheric symptoms (1.4%), and 14 strokes (4%). Twenty-four patients (6.9%) had contralateral symptoms: 20 strokes (5.8%) and 4 transient ischemic attacks (1.2%). Life-table analysis showed that the annual rate of ipsilateral neurologic events was 8.1%, and the annual rate of stroke was 2.1%. Seventy-five patients (22%) died in the follow-up period. Disease progression to 80% to 99% stenosis or occlusion occurred in 71 of 458 vessels (15.5%). The internal carotid arteries that showed evidence of disease progression had a significantly higher initial peak systolic velocity (251 vs 190 cm/s; P <.0001) and end diastolic velocity (74 vs 52 cm/s; P < 0.0001). Black patients and patients with ischemic heart disease were at a higher risk for disease progression. We could not identify any atherosclerotic risk factors that reliably predicted patients in whom future ipsilateral neurologic symptoms were more likely to develop. However, there was an increased risk of stroke associated with progression of disease. CONCLUSION: Patients who are asymptomatic and who have moderate carotid stenoses are at significant risk for neurologic symptoms and death, but have a relatively low incidence rate of ipsilateral events. The initial flow characteristics in the stenotic vessel are predictive of future disease progression, but they are not helpful in identifying patients in whom symptoms will develop.  相似文献

An analysis is presented of early design and code change data from the software cost reduction (SCR) project, a well-reported effort conducted at the US Naval Research Laboratory from 1978 to 1988. The analyses are mostly time-based studies of the change data and relationships between the data and SCR personnel activity data. Some analyses of the change data show patterns consistent with a major goal of the SCR project: the design and development of easy-to-change software. Specifically, most changes took a day or less to uncover and resolve; the majority of changes updated at most one module. Moreover, these percentages remained fairly stable. No positive relationship appeared between error-correction effort and the number of days that an error remained in the SCR design documentation. Other analyses suggest that consistency may have been temporary. For example, the analyses suggest a stepwise growth in average change effort, and an increasing percentage of changes resulted in module interface updates. Certain specific ratios between SCR change data and personnel activity data may be possible indicators of design incompleteness  相似文献

Recombinant Escherichia coli strains were constructed which simultaneously expressed the genes encoding the (S)‐oxynitrilase from cassava (Manihot esculenta) together with the wild‐type or a mutant variant of the arylacetonitrilase from Pseudomonas fluorescens EBC191 in a single organism under the control of a rhamnose‐inducible promoter. The whole cell catalysts obtained converted benzaldehyde and potassium cyanide in aqueous media at pH 5.2 mainly to (S)‐mandelic acid and/or (S)‐mandelic amide and synthesized only low amounts of the corresponding (R)‐enantiomers. The conversion of benzaldehyde and potassium cyanide (KCN) by a whole‐cell catalyst simultaneously expressing the (S)‐oxynitrilase and the wild‐type nitrilase resulted in a ratio of (S)‐mandelic acid to (S)‐mandelic amide of about 4:3. This could be explained by the strong nitrile hydratase activity of the wild‐type nitrilase with (S)‐mandelonitrile as substrate. The relative proportion of (S)‐mandelic amide formed in this system was significantly increased by coexpressing the (S)‐oxynitrilase with a carboxy‐terminally truncated variant of the nitrilase. This whole‐cell catalyst converted benzaldehyde and KCN to mandelic amide and mandelic acid in a ratio of about 9:1. The ee of the (S)‐mandelic amide formed was calculated to be >95%.  相似文献

Excess adiposity is associated with chronic inflammation, which takes part in the development of obesity-related complications. The aim of this study was to establish whether subcutaneous (SAT) or visceral (VAT) adipose tissue plays a major role in synthesis of pro-inflammatory cytokines. Concentrations of interleukins (IL): 1β, 6, 8 and 15 were measured at the protein level by an ELISA-based method and on the mRNA level by real-time PCR in VAT and SAT samples obtained from 49 obese (BMI > 40 kg/m2) and 16 normal-weight (BMI 20–24.9 kg/m2) controls. IL-6 and IL-15 protein concentrations were higher in SAT than in VAT for both obese (p = 0.003 and p < 0.0001, respectively) and control individuals (p = 0.004 and p = 0.001, respectively), while for IL-1β this was observed only in obese subjects (p = 0.047). What characterized obese individuals was the higher expression of IL-6 and IL-15 at the protein level in VAT compared to normal-weight controls (p = 0.047 and p = 0.016, respectively). Additionally, obese individuals with metabolic syndrome had higher IL-1β levels in VAT than did obese individuals without this syndrome (p = 0.003). In conclusion, concentrations of some pro-inflammatory cytokines were higher in SAT than in VAT, but it was the increased pro-inflammatory activity of VAT that was associated with obesity and metabolic syndrome.  相似文献

For those in the prevention field who need to use risk factors to identify those at risk, mere statistical significance is not sufficient to establish clinical or policy significance. For that purpose, some measure of "potency" of risk factors is needed, that is, an effect size interpretable and meaningful in a clinical or policy context. The numerous possible candidate measures available often lead to contradictory conclusions. Which of the candidate measures is right or wrong for clinical and policy decision making depends on the fit between the trade-offs between false-positive and false-negative decisions implicit in the definition of the measure (and often unknown to the user) and those related to a specific context of application. This paper summarizes arc-flash calculation results in identifying some of the difficulties in translating arc-flash calculations into real-world improvements in safety. This article addresses the following: design considerations, operating considerations, maintenance considerations, and numerous equipment specification issues that can greatly affect the amount of incident energy a worker is exposed to. Mercury is a longstanding concern in Maritime Canada due to high levels of contamination in a number of fish and bird species. The recycled component of past releases of anthropogenic mercury may be a significant source of ongoing pollution in many areas. Historical information on mercury releases can be used to quantify past and present anthropogenic contamination. We present an inventory of historical mercury emissions from anthropogenic sources in Maritime Canada for the years 1800-1995. Long-term trends in mercury emissions and the significance of the cumulative burden of mercury released from local sources are discussed. Emissions are calculated using both historical monitoring data and the application of emission factors. The nature of current anthropogenic sources of mercury is quite different than it was several decades ago when many of the existing policies governing mercury pollution were created. Our inventory illustrates that many of the most significant sources in the past such as the chlor-alkali industry, paint containing mercury additives, and pharmaceuticals, have been largely phased out with fossil fuel combustion and waste disposal remaining as the most significant modern sources. Atmospheric emissions in Maritime Canada peaked in 1945 (> 1,750 kg year-1), and again between 1965 and 1970 (> 2,600 kg year-1). Cumulative releases of mercury from anthropogenic sources for the years 1800-1995 were between 115 and 259 t to the atmosphere alone, and 327-448 t when discharges to wastewater and effluents were included. Assuming that only 0.2% (Nriagu, 1994.) of these releases become part of the recycled fraction of current fluxes, we estimate that between 570 and 900 kg Hg year-1 is deposited in Maritime Canada from past anthropogenic sources. Modern sources within Maritime Canada contribute at least 405 kg year-1 to the total annual deposition of 1.71 t over the provinces of New Brunswick, Nova Scotia and Prince Edward Island, leaving approximately 735 kg year-1 from natural sources and long-range contamination. Further study is needed to verify these estimates and clarify the significance of natural and long-range sources of mercury in Maritime Canada.  相似文献

In a routine application of commercially available centromeric DNA probes for the prenatal screening of common trisomies involving the autosomes 13, 18, and 21, and sex chromosomes, four cases of discrepancy between fluorescence in situ hybridization (FISH) results and follow-up cytogenetic analysis were observed from a total of 516 cases of amniocentesis. In three of these cases, the results were false negative, and in one false positive. In this case, amniocentesis was performed because of a positive triple test in a 34-year-old woman with previous infertility treatment. The alpha satellite DNA probe for chromosomes 13/21 revealed five signals in 50 per cent of uncultured amniocytes, while standard cytogenetic analysis showed a normal karyotype. FISH analysis on metaphase chromosomes demonstrated the location of the additional signal in the centromeric region of chromosome 22. This additional signal was also present in the centromeric region of chromosome 22 of the mother, providing evidence for a possible inherited polymorphism in chromosome 22 responsible for unspecific hybridization with the alpha satellite probe for chromosomes 13/21 in this case. The observed polymorphism in centromeric regions may contribute to unreliability of the use of the 13/21 alpha satellite probe for prenatal screening by FISH.  相似文献
