The noninvasive sampling of dermal interstitial fluid (ISF) for the monitoring of clinical biomarkers is a greatly appealing area of research. The identification of molecular biomarkers in biological fluids has been accelerated with -omics analyses but remains limited in ISF because of its time-consuming and complex extraction process. Here, the generation of microneedle (MN) patches made of superabsorbent acrylate-based hydrogels for the rapid sampling of dermal ISF is described to explore its proteome. In depth, iterative optimization allows the identification of novel acrylate-based compositions with the required chemical, mechanical, and biocompatibility properties allowing proteomic analysis of the extracted ISF for the first time after sampling with swelling MNs. The generated MN arrays show no cytotoxic effect, successfully cross the stratum corneum, and can collect up to 6 µL of dermal ISF in 10 min in vivo. Proteomics lead to the detection of 176 clinically relevant biomarkers in the collected samples validating the use of ISF as a relevant bodily fluid for disease monitoring and diagnostic. Importantly, it is discovered that extraction fingerprint is strongly dependent on the MNs chemistry, and thus specific biomarkers could be selectively extracted by tuning the composition of the patch, making the system versatile and specific.  相似文献

Sarcopenia is defined as the age-related loss of skeletal muscle mass, quality, and strength. The pathophysiological mechanisms underlying sarcopenia are still not completely understood. The aim of this work was to evaluate, for the first time, the expression of NLRP3 inflammasome in bovine skeletal muscle in order to investigate the hypothesis that inflammasome activation may trigger and sustain a pro-inflammatory environment leading to sarcopenia. Samples of skeletal muscle were collected from 60 cattle belonging to three age-based groups. Morphologic, immunohistochemical and molecular analysis were performed to assess the presence of age-related pathologic changes and chronic inflammation, the expression of NLRP3 inflammasome and to determine the levels of interleukin-1β, interleukin-18 and tumor necrosis factor alpha in muscle tissue. Our results revealed the presence of morphologic sarcopenia hallmark, chronic lymphocytic inflammation and a type II fibers-selective NLRP3 expression associated to a significant decreased number of immunolabeled-fibers in aged animals. Moreover, we found a statistically significant age-related increase of pro-inflammatory cytokines such as interleukin-1β and interleukin-18 suggesting the activation of NLRP3 inflammasome. Taken together, our data suggest that NLRP3 inflammasome components may be normally expressed in skeletal muscle, but its priming and activation during aging may contribute to enhance a pro-inflammatory environment altering normal muscular anabolism and metabolism.  相似文献

Extracellular vesicles (EVs) are a heterogeneous group of cell-derived submicron vesicles released under physiological or pathological conditions. EVs mediate the cellular crosstalk, thus contributing to defining the tumor microenvironment, including in epithelial ovarian cancer (EOC). The available literature investigating the role of EVs in EOC has been reviewed following PRISMA guidelines, focusing on the role of EVs in early disease diagnosis, metastatic spread, and the development of chemoresistance in EOC. Data were identified from searches of Medline, Current Contents, PubMed, and from references in relevant articles from 2010 to 1 April 2020. The research yielded 194 results. Of these, a total of 36 papers, 9 reviews, and 27 original types of research were retained and analyzed. The literature findings demonstrate that a panel of EV-derived circulating miRNAs may be useful for early diagnosis of EOC. Furthermore, it appears clear that EVs are involved in mediating two crucial processes for metastatic and chemoresistance development: the epithelial–mesenchymal transition, and tumor escape from the immune system response. Further studies, more focused on in vivo evidence, are urgently needed to clarify the role of EV assessment in the clinical management of EOC patients.  相似文献

In this paper, new homogeneous hot-electron injection data at 300 K and 77 K is provided covering applied voltages from well below to well above the Si-SiO2 barrier height, and a wide range of oxide fields. We found that, in contrast to the MOSFET case, homogeneous injection shows two different regimes for accelerating voltages below and above the barrier height. A simple interpretation of the data is proposed, and supported by Monte Carlo (MC) simulations of the injection experiment. Essentially, the two regimes are the signature of a marked transition between an electron population mostly heated by the electric field, and a tail population created by additional but less efficient energy gain mechanisms, leading to a sharp transition in the carrier distribution function. The details of the bias and temperature dependence of injection are then interpreted as the combined effect of tunneling and carrier distribution. Furthermore, possible implications on MOSFET gate currents are briefly discussed  相似文献

Carrier transport across the semiconductor space-charge region of a silicon triangular barrier diode was investigated by a Monte Carlo simulation. Oscillations of the electron mean kinetic energy are observed as a function of position along the uphill slope of the barrier under bias. At a given point on the uphill slope, the energy distribution function shows an oscillatory behavior, with a periodicity corresponding to the optical phonon energy. These oscillations are shown to be due to the nonequilibrium dynamics of the electron interaction with optical phonons in the situation when other inelastic electron scattering processes are negligible. The energy oscillations are superimposed on a smooth cooling of the distribution in the transport toward the top of the barrier, as current flows through the system. A comparison with the thermionic theory quantifies the importance of nonequilibrium effects in short-range electronic transport  相似文献

State-of-the-art device simulation is applied to the analysis of possible scaling strategies for the future CMOS technology, adopting the ultrathin silicon body (UTB) double-gate (DG) MOSFET and considering the main figures of merit (FOM) for the high-performance N-MOS transistor. The results of our analysis confirm the potentials of UTB-DG MOSFETs. In particular, the possibility to control the short-channel effects by thinning the silicon layer is fully exploited allowing to adopt almost undoped silicon channel, leading to reduced transversal field. As a consequence, the impact of surface roughness at the Si-oxide interface and the gate tunneling leakage current are substantially reduced compared to the case of highly doped bulk MOSFETs. According to our results, thanks to the suppression of gate leakage current, scaling of the UTB-DG MOSFET down to the 32 nm technology node appears possible adopting -based gate dielectrics. In spite of the improved mobility at given inversion charge density, the simulated on-currents are substantially lower than those required by the 2005 ITRS for the 45 and 32 nm nodes . Nonetheless, thanks to relaxed scaling of the oxide thickness, hence to reduced gate capacitance, the requirements in terms of intrinsic delay and power-delay product can be satisfied. The issue of variability is analyzed by evaluating the dependence of the key FOM on the variation of critical dimensions such as the thickness of the gate oxide and of the silicon layer.  相似文献
