Lipoxidation in almond-derived products was investigated using the chemiluminescence (CL) and thiobarbituric acid-reactive substances (TBARS) methods to detect the first and later reaction products, respectively. The effects of light during storage at 5 °C, 22 °C and 40 °C were studied, as well as the effects of combined heat/water activity treatments in the 60–120 °C and 0.38–0.72 range. During storage, light was found to enhance the CL and TBARS values, and specific responses were observed in almond paste and the final Calisson product. During the heating of almond paste, as the initial water activity (aw) increased, the CL rate constants increased during heating to 60 °C and 80 °C, but interestingly, these values decreased during further heating to 120 °C, whereas the maximum TBARS rate constants occurred at aw 0.57 at all the heating temperatures tested. The activation energies, based on the CL and TBARS values, decreased specifically when the aw increased from 0.38 to 0.72, giving overall values ranging from110 kJ mol−1 to 60 kJ mol−1. Likewise, in the same water activity range, the temperature-dependent rate constant enhancing factor (Q10) decreased from 3.3 to 1.6. 相似文献

Acute liver injury (ALI) is a severe disorder resulting from excessive hepatocyte cell death, and frequently caused by acetaminophen intoxication. Clinical management of ALI progression is hampered by the dearth of blood biomarkers available. In this study, a bioinformatics workflow was developed to screen omics databases and identify potential biomarkers for hepatocyte cell death. Then, discovery proteomics was harnessed to select from among these candidates those that were specifically detected in the blood of acetaminophen-induced ALI patients. Among these candidates, the isoenzyme alcohol dehydrogenase 1B (ADH1B) was massively leaked into the blood. To evaluate ADH1B, we developed a targeted proteomics assay and quantified ADH1B in serum samples collected at different times from 17 patients admitted for acetaminophen-induced ALI. Serum ADH1B concentrations increased markedly during the acute phase of the disease, and dropped to undetectable levels during recovery. In contrast to alanine aminotransferase activity, the rapid drop in circulating ADH1B concentrations was followed by an improvement in the international normalized ratio (INR) within 10–48 h, and was associated with favorable outcomes. In conclusion, the combination of omics data exploration and proteomics revealed ADH1B as a new blood biomarker candidate that could be useful for the monitoring of acetaminophen-induced ALI. 相似文献

Starting from our previously reported hit, a series of 1,5-diaryl-1,2,3-triazole-4-carbohydrazones were synthesized and evaluated as inhibitors of the YAP/TAZ-TEAD complex. Their binding to hTEAD2 was confirmed by nanodifferential scanning fluorimetry, and some of the compounds were also found to moderately disrupt the YAP-TEAD interaction, as assessed by a fluorescence polarization assay. A TEAD luciferase gene reporter assay performed in HEK293T cells and RTqPCR measurements in MDA-MB231 cells showed that these compounds inhibit YAP/TAZ-TEAD activity to cells in the micromolar range. In spite of the cytotoxic effects displayed by some of the compounds of this series, they are still good starting points and can be suitably modified into an effective and viable YAP-TEAD disruptor in the future. 相似文献

Integrated water resources management (IWRM) has been prescribed in the global water policy literature for decades. This article looks at how the concept has been applied in Nepal. It highlights the normative approach in IWRM policy formulation, the existing institutional barriers to apply it and how these resulted in the framing of IWRM ‘implementation’ as merely a compilation of donor-funded projects. Current discourse on IWRM highlights the need to shift the emphasis from national policy formulation to local adaptive, pragmatic approaches to IWRM. This article brings to light the need to identify potential entry points to scale up locally rooted water management approaches towards the development of nested institutional set-ups in water resources management. 相似文献

Pharmacokinetics (PK) refers to the time course of drug concentrations in the body and since knowledge of PK aids understanding of drug efficacy and safety, numerous PK studies are performed in animals and humans during the drug development process. In vitro to in vivo extrapolation and physiologically based pharmacokinetic (PBPK) modeling are tools that integrate data from various in silico, in vitro, and in vivo sources to deliver mechanistic quantitative simulations of in vivo PK. PBPK models are used to predict human PK and to evaluate the effects of intrinsic factors such as organ dysfunction, age, and genetics as well as extrinsic factors such as co-administered drugs. In recent years, the use of PBPK within the industry has greatly increased. However, insufficient data on how the abundance of metabolic enzymes and membrane transporters vary in different human patient populations and in different species has been a limitation. A major advance is therefore expected through reliable quantification of the abundance of these proteins in tissues. This review describes the role of PBPK modeling in drug discovery and development, outlines the assumptions involved in integrating protein abundance data, and describes the advances made and expected in determining abundance of relevant proteins through mass spectrometric techniques. 相似文献

In order to investigate the transient behaviour of a solid oxide electrolysis cell, tests were carried out on a single SOEC both under steady-state and transient conditions for 600 h. Steady-state operation was performed at the reference current of −20 A, corresponding to the thermoneutral voltage (1.28 V), whereas transient tests consisted in 1800 square waves applied over 140 cumulated hours, with a current of −1 A and −20 A as well as 0 A and −20 A. Independently from the operating conditions, the measured voltage degradation rate of 5% (70 mV)/1000 h was stable for the entire experiment. Consequently, it was concluded that an SOEC can be operated under on–off conditions with no increase of the degradation rate. This result paves the way for modular operation of a high temperature electrolyser fed with intermittent electrical energy. 相似文献

High Temperature ELectrolysers (HTEL) operate around 1073 K with steam at 1073 K. Water is previously heated up in the Balance of Plant by the hot outlet gases and optionally by electrical heaters. If liquid water is fed to the system, vaporisation needs are covered by electrical heating, which leads to a low system efficiency of 89% vs. HHV. Using steam instead of liquid water would suppress vaporisation needs, thus increase the efficiency. This work aims to analyse the potential benefits of a steam supply. Calculation is performed without considering the energy required to preheat water. Results show that feeding the system with low-temperature steam instead of liquid water enables a system efficiency jump of 18%. Further increasing the steam temperature to 933 K negligibly impacts the system efficiency and is therefore unnecessary. It is concluded that a low-temperature steam source is sufficient to increase significantly the HTEL system efficiency. 相似文献
