MCF7 and ZR75-1 breast cancer cells grow as adherent monolayers in tissue culture. Treatment with the serum serine protease plasmin causes them to detach and to grow as floating multicellular spheroids. Two plasmin activators, urokinase plasminogen activator and streptokinase, induce the same growth pattern changes in the presence of plasminogen. Serum contains also plasminogen activator inhibitors. Aged serum, deficient in plasminogen activator inhibitors, converts spontaneously monolayer breast cancer cells into multicellular spheroids which readily revert to monolayer growth after addition of fresh serum. Urokinase blocks the reversion. The formation of multicellular spheroids does not affect the proliferative rate of breast tumor cells but endows tumor cells with increased resistance to the chemotherapeutic drugs, doxorubicin and paclitaxel.  相似文献

BACKGROUND: With the epidemiological transition phenomenon, Tunisia is now facing the growing prevalence of chronic diseases. Environmental and behavioural changes such as the adoption of new dietary habits and a sedentary lifestyle, and the stress of urbanization and of working conditions all lead to the rise in cardiovascular disease (CVD) risk factors. METHODS: The aim of this study is to estimate the prevalence of the main CVD risk factors in an urban context. For this, we have undertaken a household epidemiological survey of a representative sample of the adult urban population of Soussa, Tunisia (n = 957). RESULTS: This study shows high prevalences of hypertension (blood pressure >160/95 mmHg) of 18.8 per cent with an adjusted rate of 15.6 per cent, history of diabetes of 10.2 per cent, and obesity (body mass index > 30) of 27.7 per cent [significantly higher in women (34.4 per cent)], android obesity 36 per cent, smoking habits 21.5 per cent [significantly higher in men (61.4 per cent)]. CONCLUSION: With this profile of CVD risk factors, Tunisia must implement a national strategy of primary prevention and heart health promotion in addition to the efforts recently made in secondary prevention of some chronic diseases such as hypertension and diabetes.  相似文献

Two distinct marine epoxy organic coatings were aged in a sodium chloride (30 g L−1) solution at different temperatures. The water absorption was evaluated using gravimetric tests onto free films and EIS experiments were realized onto attached films. A strong leaching process was evidenced by gravimetry and the FT-IR analysis of the solution after ageing revealed the presence of extracted organic components. However, EIS results were not sensitive to this phenomenon and the water uptake values, obtained with a modified Brasher–Kingsbury equation, were found to be higher than gravimetric values. The comparison of the water volume fraction obtained by EIS with the apparent water mass fraction allowed following the species desorption with immersion time. It was proposed that this leaching process was related to a loss of the polymer density and the results showed that this phenomenon was thermally activated.  相似文献

Mycotoxins are secondary metabolites produced by filamentous fungi detected in food, such are grapes. OTA was evaluated in ten handle musts from different Tunisian vineyard. This mycotoxin was found at levels 1.1 mug/L to 4.3 mug/L. A survey was conducted to assess the contamination of the Tunisian vineyard with pathogenic fungal species, in particular those responsible of the OTA production. The results were evaluated for the first time in parcels cultivated in the North, in the Centre and in the South of the country. Italia Muscate and Superior Seedless varieties were concerned at three developmental stages of the berry, setting, veraison and maturity. Carigon variety was used as positive control for musts contaminating by OTA. The main fungal species isolated were Aspergillus spp. (33.32%), Botrytis cinerea (23.32%), Alternaria spp. (12.80%), Cladosporium spp. (10.59%) and Penicillium spp. (8.3%). The isolates of the Aspergillus genus were identified as Aspergillus niger aggregate (77%), Aspergillus carbonarius (15%) and Aspergillus flavus (8%). Their presence was characterized by a significant decrease in the Centre during the veraison and a slight increase in the North and the South during the maturity stage. Furthermore, when comparing Superior Seedless and Italia Muscate cultivated in the same area, the aspergilli were particularly less abundant at the setting stage in the case of Superior Seedless. There is no correlation between the OTA amount in musts and the contamination by Aspergillus species in different vineyards and for grape varieties studied.  相似文献

Machining defects as rebuilt material and dislodgement were often induced by cutting of difficult to machining AISI 304 stainless steel. Their density increases with a decreasing of cutting speed. The effect of these defects on surface topography and residual stress was evaluated by roughness and X‐ray diffraction measurements coupled with numerical simulation. The role of the rebuilt material on the distribution of fatigue crack nucleation sites was investigated by scanning electron microscope examination of post fatigue samples loaded at different imposed strain amplitudes. The association of machining defects and fatigue crack nucleation sites was attributed to the contribution of additional tensile residual stresses induced by rebuilt material rather than local stress concentration. Moreover, the fatigue crack coalescence is promoted by increasing the rebuilt material density. When the machining defect density increases from 5 to 60 particles/mm2, the fatigue life decreases from ?22% to ?65% with respect to the electropolished surface.  相似文献

We report the results of static and dynamic mechanical measurements carried out on two epoxy-based coatings with different contents of sodium chloride solution. Both coatings presented strong lixiviation processes in saline solutions in parallel with water absorption. This was interpreted as a polymer density loss where ionic species are strongly involved and that may lead to a change of the mechanical properties of the coating. The mechanical behavior of both coatings showed clearly three domains (elastic, visco-elastic and visco-plastic), which depend on the solution ingress. In agreement with previous results obtained on epoxies, the Young modulus decreases with the solution content. Visco-elastic (VE) and visco-plastic (VP) behavior dependence with solution ingress showed two antagonist phenomena: softening (elongation increase) and hardening (ultimate stress increase). The first one is classically associated with plasticization while hardening is mainly linked with the development of internal stresses. These phenomena were correlated with the cross-linking density distribution within the coatings.  相似文献

A set of measurements have been conducted in order to determine the annual effective dose resulting from the ingestion of natural radionuclides present in eight different brands of bottled water commercialised in Tunisia. Using high-resolution gamma spectrometry technique, we have noted the presence of the following radionuclides: 214Bi, 226Ra, 228Ac, 212Pb, 235U and 40K. For all classes of age, only the radium concentration was found to be significant in the calculation of the equivalent dose. Some of the studied samples exceeded the reference level of 0.1 mSv y(-1) of effective dose.  相似文献

The surface and subsurface integrity of metallic ground components is usually characterized by an induced tensile residual stress, which has a detrimental effect on the fatigue life of these components. In particular, it tends to accelerate the initiation and growth of the fatigue cracks. In this investigation, to deliberately generate compressive residual stresses into the ground surfaces of the AISI 304 stainless steel (SS), wire brushing was applied. It was found that under the experimental conditions selected in this investigation, while the surface roughness was slightly improved by the brushing process, the surface residual stress shifted from a tensile stress (σ=+450 MPa) to a compressive stress (σ=−435 MPa). On the other hand, the work-hardened deformation layer was almost two times deeper after wire brushing. Concerning the fatigue life, an improvement of 26% in terms of endurance limit at 2×106 cycles was realized. Scanning electron microscope (SEM) observations of the fatigue fracture location and size were carried out to explain the fatigue life improvement. It was found that the enhancement of the fatigue strength could be correlated with the distribution and location of the fatigue fracture nucleation sites. Concerning the ground surfaces, it was seen that the fatigue cracks initiated at the bottom of the grinding grooves and were particularly long (150–200 μm). However, the fatigue cracks at the brushed surfaces were shorter (20–40 μm) and appeared to initiate sideways to the plowed material caused by the wire brushing. The results of the wire-brushed surface characterization have shown that significant advantages can be realized regarding surface integrity by the application of this low-cost process compared to shot peening.  相似文献
