Autoantibodies-abzymes hydrolyzing DNA, myelin basic protein, and oligosaccharides have been revealed in the sera of patients with multiple sclerosis (MS). In MS, specific microRNAs are found in blood and cerebrospinal fluid, which are characterized by increased expression. Autoantibodies, specifically hydrolyzing four different miRNAs, were first detected in the blood of schizophrenia patients. Here, we present the first evidence that 23 IgG antibodies of MS patients effectively recognize and hydrolyze four neuroregulatory miRNAs (miR-137, miR-9-5p, miR-219-2-3p, and miR-219-5p) and four immunoregulatory miRNAs (miR-21-3p, miR-146a-3p, miR-155-5p, and miR-326). Several known criteria were checked to show that the recognition and hydrolysis of miRNAs is an intrinsic property of MS IgGs. The hydrolysis of all miRNAs is mostly site-specific. The major and moderate sites of the hydrolysis of each miRNA for most of the IgG preparations coincided; however, some of them showed other specific sites of splitting. Several individual IgGs hydrolyzed some miRNAs almost nonspecifically at nearly all internucleoside bonds or demonstrated a combination of site-specific and nonspecific splitting. Maximum average relative activity (RA) was observed in the hydrolysis of miR-155-5p for IgGs of patients of two types of MS—clinically isolated syndrome and relapsing-remitting MS—but was also high for patients with primary progressive and secondary progressive MS. Differences between RAs of IgGs of four groups of MS patients and healthy donors were statistically significant (p < 0.015). There was a tendency of decreasing efficiency of hydrolysis of all eight miRNAs during remission compared with the exacerbation of the disease.  相似文献

Beyond-design-basis regimes of centrifugal contactors for U stripping and of mixer–settlers for carbonate regeneration of the solvent, 30% TBP in Isopar M, were studied from the viewpoint of the influence of incomplete uranium stripping on the phase stability of the extraction system in the step of the carbonate scrubbing of the solvent. Dynamic relationship between these two operations was determined. A mathematical model of uranium stripping at stepwise–20% deviation from the nominal value was developed and verified; it was shown that the beyond-design-basis regime develops in such apparatuses within 4 h at the total trial time longer than 8 h. The mathematical relationships obtained were used in the development of software for calculating transient processes that occur when the stream parameters in the course of carbonate regeneration of the solvent change. The beyond-design-basis regime of the solvent regeneration was found to have two regions: region of stoichiometric excess of carbonate ions over uranyl ions with the risk of precipitation and region of excess of uranyl ions over carbonate ions with acidification of the unit and uranium breakthrough with the recycled solvent. The conditions of the tests performed correspond to the second region.  相似文献

A certain pulse of electric current combined with plastic deformation is a powerful tool for improving the formability of hard-to-deform metal alloys. In recent years, much research has indicated that the current not only improves macro-mechanical properties but also influences microstructural-level phenomena such as recrystallization, local phase transformation, grain refinement, and even amorphous nanocrystallization. Despite the huge experimental dataset, virtually no focus has been placed on phenomenon??s computing. In this fashion, present paper concerned with the continuum-level numerical analysis of the pulse impact on the stress field during plastic deformation. Computation conducted herein has shown that a high density electric current pulsing weakens the stress field during plastic deformation. Ultimate results of the study should be useful in developing the novel metal processing technology.  相似文献

Character of conversion of organic matter from Domanic rocks of Pervomaiskoye field (Tatarstan) of Semiluki horizon of upper Devonian deposits in the hydrothermal-catalytic system at temperature of 300?°C in carbon dioxide medium was studied with the application of complex of oil-soluble precursors of catalysts containing Fe, Co, and Cu. In presence of catalysts complex, content of organic extract increases, in which content of hydrocarbon fractions, saturated and aromatic hydrocarbons, increases 1.5 times, while resins content decreases by two times. As result of kerogen destruction in products of experiments, the content of asphaltenes and carbonaceous substances such as carbenes and carboides increase.  相似文献

The German zoologist and geneticist Ludwig Plate was a pupil and successor of the “German Darwin” Ernst Haeckel as the director of the Institute of Zoology at Jena University. Plate campaigned for a revival of the original Darwinism. His research program, which he labelled “old-Darwinism”, proclaimed the synthesis of selectionism with “moderate Lamarckism” and orthogenesis.This article reconstructs and analyses Plate’s “old-Darwinian” synthesis and sheds light on Plate’s controversial biography, especially his conflict with Haeckel. Transplantation of various types of stem cells as a possible therapy for stroke has been tested for years, and the results are promising. Recent investigations have shown that the administration of the conditioned media obtained after stem cell cultivation can also be effective in the therapy of the central nervous system pathology (hypothesis of their paracrine action). The aim of this study was to evaluate the therapeutic effects of the conditioned medium of hiPSC-derived glial and neuronal progenitor cells in the rat middle cerebral artery occlusion model of the ischemic stroke. Secretory activity of the cultured neuronal and glial progenitor cells was evaluated by proteomic and immunosorbent-based approaches. Therapeutic effects were assessed by overall survival, neurologic deficit and infarct volume dynamics, as well as by the end-point values of the apoptosis- and inflammation-related gene expression levels, the extent of microglia/macrophage infiltration and the numbers of formed blood vessels in the affected area of the brain. As a result, 31% of the protein species discovered in glial progenitor cells-conditioned medium and 45% in neuronal progenitor cells-conditioned medium were cell type specific. The glial progenitor cell-conditioned media showed a higher content of neurotrophins (BDNF, GDNF, CNTF and NGF). We showed that intra-arterial administration of glial progenitor cells-conditioned medium promoted a faster decrease in neurological deficit compared to the control group, reduced microglia/macrophage infiltration, reduced expression of pro-apoptotic gene Bax and pro-inflammatory cytokine gene Tnf, increased expression of anti-inflammatory cytokine genes (Il4, Il10, Il13) and promoted the formation of blood vessels within the damaged area. None of these effects were exerted by the neuronal progenitor cell-conditioned media. The results indicate pronounced cytoprotective, anti-inflammatory and angiogenic properties of soluble factors secreted by glial progenitor cells.  相似文献

Multicomponent, multiphase heat-protecting multicoats formed by anodic microarc oxidation (AMO) can optimally solve the problem of the heat protection of pistons of internal combustion engines. Based on the revealed correlations, the chemical composition of the bath and the range of the AMO conditions are optimized; some engineering ways of obtaining heat-resistant, up to 200 μm thick ceramic protective coatings on Silumin surfaces of complicated profile are proposed.  相似文献

Efficient yellow–green electroluminescence emission at λmax = 530 nm with CIE coordinates x = 0.3913, y = 0.4947 was obtained with organic light-emitting devices based on tris(2-methyl-8-quinolinolate) scandium (1). The device with the configuration of indium tin oxide/N,N′-bis(3-methylphenyl)-N,N′-diphenylbenzidine/1/Yb exhibits current efficiency of 3.1 cd/A and power efficiency of 1.8 lm/W at a luminance of 100 cd/m2. The DFT calculations demonstrate that structural changes of the scandium complex 1 influence the electroluminescence spectrum, the better agreement with experimental data being achieved when monodentate ligands are taken into consideration.  相似文献
