Ultrashort laser pulses have enabled highly precise and delicate processing of biological specimens. We present the results of using femtosecond laser pulses for dissection of zona pellucida (ZP) in mouse embryos during assisted hatching procedure. We studied the effects of application of femtosecond laser radiation in the infrared (1028 nm) and visible (514 nm) wavelength ranges. Laser irradiation parameters were optimized so as not to compromise the viability of the treated embryos. We have demonstrated that application of femtosecond laser pulses with the energies in the range of 250–320 nJ (for the wavelength of 1028 nm) and 47–112 nJ (for 514 nm) resulted in efficient ZP dissection. Femtosecond laser-assisted ZP drilling does not slow down the development of pre-implantation embryos and leads to 90–95% frequency of complete hatching. The thermal effects can be significantly lower when femtosecond lasers are used as compared to continuous wave or long-pulse lasers. It is crucial when dealing with living cells or organisms. By optimizing femtosecond laser radiation parameters assisted hatching as well as a wide range of embryo-surgical procedures can be efficiently performed, thus creating a great potential of using femtosecond lasers as a multi-purpose “tool of choice” for specialists in the fields of embryology and developmental biology.  相似文献

The results are presented of the works in the field of development of equipment, investigation techniques, and technologies for biology and medicine performed in the Joint Institute for High Temperatures of the Russian Academy of Scienses (JIHT RAS). On the base of the new generation infrared femtosecond lasers, the experimental models are developed and manufactured of laser tweezers, scalpel, and the “tweezers-scalpel” combined system. The results are presented of the experimental studies on the noncontact mammalian cell fusion (blastomeres of mouse embryos on day 1.5 of development) by means of the femtosecond laser pulses.  相似文献

Epigenetic regulation of gene expression is necessary for maintaining higher-order cognitive functions (learning and memory). The current understanding of the role of epigenetics in the mechanism of Alzheimer’s disease (AD) is focused on DNA methylation, chromatin remodeling, histone modifications, and regulation of non-coding RNAs. The pathogenetic links of this disease are the misfolding and aggregation of tau protein and amyloid peptides, mitochondrial dysfunction, oxidative stress, impaired energy metabolism, destruction of the blood–brain barrier, and neuroinflammation, all of which lead to impaired synaptic plasticity and memory loss. Ultrashort peptides are promising neuroprotective compounds with a broad spectrum of activity and without reported side effects. The main aim of this review is to analyze the possible epigenetic mechanisms of the neuroprotective action of ultrashort peptides in AD. The review highlights the role of short peptides in the AD pathophysiology. We formulate the hypothesis that peptide regulation of gene expression can be mediated by the interaction of short peptides with histone proteins, cis- and transregulatory DNA elements and effector molecules (DNA/RNA-binding proteins and non-coding RNA). The development of therapeutic agents based on ultrashort peptides may offer a promising addition to the multifunctional treatment of AD.  相似文献

The goals of the present study were to characterize the phages isolated by magnetic nanocomplex, to determine the effect of natural polyphenols on phage activity against E. coli and the stability of free and encapsulated bacteriophages under UV irradiation, and their efficacy against E. coli on romaine lettuce (Lactuca sativa L. var. longifolia) as a fresh food model. The morphology of the two magnetically isolated phages were characterized using the transmission electron microscope (TEM). All isolated phages were classified in a morphological family of tailed phages of the order Caudovirales, Myoviridae family. The PCR results showed that the stx1 and cI genes were not on any phage. Assays were carried out to determine the effect of four polyphenolic natural extracts on phages under UV radiation. The highest activity and stability of phages expressed in phage titer (PFU/ml) were observed in the presence of polyphenols from tarbush leaves (Flourensia cernua DC). Coliphages were encapsulated in alginate/chitosan and alginate/PVA carriers with and without polyphenols from tarbush leaves. Encapsulation led to a higher phage stability under UV radiation. All encapsulated formulations and free coliphages were applied on the surfaces of romaine lettuce containing E. coli cells. An effective reduction of 2 log of E. coli titers from lettuce was observed. Encapsulated phage formulations with and without polyphenols from tarbush leaves could be considered for future application in fresh food preservation.  相似文献

The synthesis of crystalline, nonagglomerated, and perfectly dispersible Co3O4 nanoparticles with an average size of 3–7 nm using a solvothermal reaction in tert‐butanol is reported. The very small size and high dispersibility of the Co3O4 nanoparticles allow for their homogeneous deposition on mesoporous hematite layers serving as the photoactive absorber in the light‐driven water splitting reaction. This surface treatment leads to a striking photocurrent increase. While the enhancement of hematite photoanode performance by cobalt oxides is known, the preformation and subsequent application of well‐defined cobalt oxide nanoparticles are novel and allow for the treatment of arbitrarily complex hematite morphologies. Photoelectrochemical and transient absorption spectroscopy studies show that this enhanced performance is due to the suppression of surface electron–hole recombination on time scales of milliseconds to seconds.  相似文献

Dielectric response of vinylidene fluoride-hexafluoropropylene copolymer, crystallized in different polymorph modifications, to a high-voltage electric field was studied. Polarization switching at high electric fields below coercive was detected. It was found that crystallization of the copolymer predominantly in nonpolar α-phase is accompanied by more intensive growth of electric displacement at polarization. For the sample with higher content of polar γ-phase, the value of high-voltage conductivity “anomalously” decreased with increasing field at fields above coercive. The data on the field dependences of the remnant polarization showed that this should be attributed to an increase of the effective capture cross section of deep traps of γ-phase polar planes for impurity and injected carriers. X-ray diffraction has revealed the field-induced transition of a part of the chains of the amorphous phase to the crystal.  相似文献

Tyrosyl-DNA phosphodiesterase 1 (TDP1) catalyzes the cleavage of the phosphodiester bond between the tyrosine residue of topoisomerase 1 (TOP1) and the 3′ phosphate of DNA in the single-strand break generated by TOP1. TDP1 promotes the cleavage of the stable DNA–TOP1 complexes with the TOP1 inhibitor topotecan, which is a clinically used anticancer drug. This article reports the synthesis and study of usnic acid thioether and sulfoxide derivatives that efficiently suppress TDP1 activity, with IC50 values in the 1.4–25.2 μM range. The structure of the heterocyclic substituent introduced into the dibenzofuran core affects the TDP1 inhibitory efficiency of the compounds. A five-membered heterocyclic fragment was shown to be most pharmacophoric among the others. Sulfoxide derivatives were less cytotoxic than their thioester analogs. We observed an uncompetitive type of inhibition for the four most effective inhibitors of TDP1. The anticancer effect of TOP1 inhibitors can be enhanced by the simultaneous inhibition of PARP1, TDP1, and TDP2. Some of the compounds inhibited not only TDP1 but also TDP2 and/or PARP1, but at significantly higher concentration ranges than TDP1. Leader compound 10a showed promising synergy on HeLa cells in conjunction with the TOP1 inhibitor topotecan.  相似文献
