The construction of rhizobial strains which increase plant biomass under controlled conditions has been previously reported. However, there is no evidence that these newly constructed strains increase legume yield under agricultural conditions. This work tested the hypothesis that carefully manipulating expression of additional copies of nifA and dctABD in strains of Rhizobium meliloti would increase alfalfa yield in the field. The rationale for this hypothesis is based on the positive regulatory role that nifA plays in the expression of the nif regulon and the fact that a supply of dicarboxylic acids from the plant is required as a carbon and energy source for nitrogen fixation by the Rhizobium bacteroids in the nodule. These recombinant strains, as well as the wild-type strains from which they were derived, are ideal tools to examine the effects of modifying or increasing the expression of these genes on alfalfa biomass. The experimental design comprised seven recombinant strains, two wild-type strains, and an uninoculated control. Each treatment was replicated eight times and was conducted at four field sites in Wisconsin. Recombinant strain RMBPC-2, which has an additional copy of both nifA and dctABD, increased alfalfa biomass by 12.9% compared with the yield with the wild-type strain RMBPC and 17.9% over that in the uninoculated control plot at the site where soil nitrogen and organic matter content was lowest. These increases were statistically significant at the 5% confidence interval for each of the three harvests made during the growing season. Strain RMBPC-2 did increase alfalfa biomass at the Hancock site; however, no other significant increases or decreases in alfalfa biomass were observed with the seven other recombinant strains at that site. At three sites where this experiment was conducted, either native rhizobial populations or soil nitrogen concentrations were high. At these sites, none of the recombinant strains affected yield. We conclude that RMBPC -2 can increase alfalfa yields under field conditions of nitrogen limitation, low endogenous rhizobial competitors, and sufficient moisture.  相似文献

The gate bias polarity dependence of charge trapping and time-dependent dielectric breakdown (TDDB) in nitrided and reoxidized nitrided silicon dioxides prepared by rapid thermal processing (RTP) is reported. Charge trapping during high-field injection can be reduced by rapid thermal nitridation for both substrate and gate injection. While reoxidation of nitrided oxides shows further reduction in charge trapping for substrate injection, degradation is observed for gate injection. Similar effects are observed for TDDB: reoxidized nitrided oxides show charge-to-breakdown in excess of 300 C/cm2 for substrate injection, but less than 30 C/cm2 for gate injection. These effects are related to the nitrogen and hydrogen profiles in the oxides. By tailoring the process conditions, a symmetric behavior of NO and RONO films with low charge trappings and Q BD in excess of 50 C/cm2 is possible, making them attractive as long-lifetime dielectrics from EEPROM (electrically erasable programmable ROM) and flash EEPROM technologies  相似文献

Flexible electronics utilizing single crystalline semiconductors typically require post-growth processes to assemble and incorporate the crystalline materials onto flexible substrates. Here we present a high-precision transfer-printing method for vertical arrays of single crystalline semiconductor materials with widely varying aspect ratios and densities enabling the assembly of arrays on flexible substrates in a vertical fashion. Complementary fabrication processes for integrating transferred arrays into flexible devices are also presented and characterized. Robust contacts to transferred silicon wire arrays are demonstrated and shown to be stable under flexing stress down to bending radii of 20 mm. The fabricated devices exhibit a reversible tactile response enabling silicon based, nonpiezoelectric, and flexible tactile sensors. The presented system leads the way towards high-throughput, manufacturable, and scalable fabrication of flexible devices.  相似文献

People report excessive, unwanted social interactions, insufficient privacy, and are observed to be more socially withdrawn under crowded living conditions. If people socially withdraw from one another as a way to cope with crowding, this could lead to an unintended side effect of breakdowns in socially supportive relationships. A breakdown in socially supportive relationships could, in turn, account for some of the pathological consequences of chronic, high density living conditions. An empirical study of 175 male heads of household among inner city residents in India suggests that the adverse effects of residential crowding on psychological health are mediated by a breakdown of social support systems. Amphibian embryo trypsin inhibitor (ATI) is shown to be a temporary inhibitor in which full trypsin activity can eventually be restored. Lineweaver-Burk plots and other binding data indicate a reversible change from competitive to non-competitive inhibition during incubation of enzyme with inhibitor in the absence of substrate. The inhibitor is active while sequestered in the yolk, and the sequestered inhibitor co-purifies with lipovitellin when the yolk is fractionated. It is further demonstrated that the inhibitor slowly leaches from the yolk in a soluble form, and it is suggested that this leakage is the result of a negative feedback controlled proteolysis within the yolk. Possible interactions between ATI and endogenous trypsin-like proteases are discussed with particular regard to the potential involvement of those materials in the control of certain developmental events; notably melanogenesis.  相似文献

Seventeen weight-trained males were divided into an overtraining group [OT; n = 11; age = 22.0 +/- 0.9 (SE) yr] that weight trained their legs daily for 2 wk with 100% 1 repetition maximum relative intensity on a squat machine and a control group (n = 6; age = 23.7 +/- 2.4 yr) that exercised 1 day/wk with low relative intensity (50% 1 repetition maximum). Test batteries including strength assessments and resting and exercise-induced concentrations of epinephrine and norepinephrine were conducted at the beginning, middle, and end (tests 1-3, respectively) of the study. Strength capabilities decreased by test 3 for the OT group (P < 0.05). Resting catecholamine concentrations did not change for either group during the study, whereas exercise-induced concentrations of both epinephrine (test 1 = 3,407.9 +/- 666.6 pmol/l, test 2 = 7,563.7 +/- 1,210.6 pmol/l, test 3 = 6,931.6 +/- 919.3 pmol/l) and norepinephrine (test 1 = 42.9 +/- 7.4 nmol/l, test 2 = 70.0 +/- 8.8 nmol/l, test 3 = 85.2 +/- 14.5 nmol/l) significantly increased by tests 2 and 3 for only the OT group. Correlation coefficients suggested decreased responsitivity of skeletal muscle to sympathetic nervous system activity. It appears that altered exercise-induced sympathetic nervous system activity accompanies high relative intensity resistance exercise overtraining and may be among the initial responses to the onset of the previously theoretical sympathetic overtraining syndrome.  相似文献

The subseabed is currently being considered as a disposal site for nuclear waste, and if it does become an option, monitoring to detect escape of the disposed material will be essential. In this paper, we define types of nuclear waste and present the dimensions of the monitoring problems that would be encountered in ocean disposal. We then summarize the characteristics of a number of physical, chemical, biological, and ecological monitoring methods. We also describe the advances and developments that will be necessary before the monitoring functions and support systems can be employed.  相似文献

A study of interface shear strengths between smooth and textured high density polyethylene (HDPE) geomembranes (GMs) and a woven∕nonwoven needle-punched geosynthetic clay liner (GCL) is presented. Tests were performed using a large direct shear machine capable of measuring peak and large displacement (200 mm) shear strengths. The failure surface was located at the GM∕GCL interface for all tests conducted, corresponding to a normal stress range of 1–486 kPa. Small positive pore pressures were measured for all interfaces at peak shear strength. Thus, the practice of preparing failure envelopes using total normal stress, instead of effective normal stress, appears to be conservative. Interface shear strengths for textured GMs placed against the nonwoven side of the GCL were higher than those corresponding to the woven side. By comparison, differences in peak shear strength for laminated and coextruded GM interfaces were relatively less. Limited tests showed that peak and large displacement shear strengths were independent of displacement rate and dependent on the shear direction of the GM. The quantity of extruded bentonite at the interfaces generally increased with normal stress and was less for nonwoven geotextile interfaces than for woven geotextile interfaces. Implications of the findings to the testing of GM∕GCL interfaces and the characterization of GM∕GCL interface shear strength are discussed.  相似文献

Improved nitrogen-fixing inoculum strains for leguminous crops must be able to effectively compete with indigenous strains for nodulation, enhance legume productivity compared to the productivity obtained with indigenous strains, and maintain stable expression of any added genes in the absence of selection pressure. We constructed a transposable element containing the tfx region for expression of increased nodulation competitiveness and the par locus for plasmid stability. The transposon was inserted into tetA of pHU52, a broad-host-range plasmid conferring the H2 uptake phenotype. The resulting plasmid, pHUTFXPAR, conferred the plasmid stability, trifolitoxin production, and H2 uptake phenotypes in the broad-host-range organism Sinorhizobium sp. strain ANU280. The broad applications of a transposon conferring plasmid stability are discussed.  相似文献
