We recently reported the successful use of the loop-mediated isothermal amplification (LAMP) reaction for hepatitis B virus (HBV) DNA amplification and its optimal primer design method. In this study, we report the development of an integrated isothermal device for both amplification and detection of targeted HBV DNA. It has two major components, a disposable polymethyl methacrylate (PMMA) micro-reactor and a temperature-regulated optical detection unit (base apparatus) for real-time monitoring of the turbidity changes due to the precipitation of DNA amplification by-product, magnesium pyrophosphate. We have established a correlation curve (R2 = 0.99) between the concentration of pyrophosphate ions and the level of turbidity by using a simulated chemical reaction to evaluate the characteristics of our device. For the applications of rapid pathogens detection, we also have established a standard curve (R2 = 0.96) by using LAMP reaction with a standard template in our device. Moreover, we also have successfully used the device on seven clinical serum specimens where HBV DNA levels have been confirmed by real-time PCR. The result indicates that different amounts of HBV DNA can be successfully detected by using this device within 1 h. 相似文献

SOH(SQL over HDFS)系统通常将数据存储于分布式文件系统HDFS中,采用Map/Reduce或分布式查询引擎来处理查询任务。得益于HDFS以及Map/Reduce的容错能力和可扩展性,SOH系统可以很好地应对数据规模的飞速增长,完成分析型查询处理。然而,在处理选择型查询或交互式查询时,这类系统暴露出性能上的缺陷。本文提出一个通用的索引技术,可以应用于SOH系统中,以提高其查询处理的效率。分析了SOH系统访问HDFS文件的过程,指出了其中影响数据加载时间的关键因素;提出了split层和split内部双层索引机制;设计并实现了聚集索引和非聚集索引。最后,在标准数据集上进行了大量实验,并与现有基于HDFS的索引技术进行了比较。实验结果表明,所提出的索引技术可以有效地提高查询处理的效率。 相似文献

After spinal cord injury (SCI) in mammals, neuronal regeneration is limited; in contrast, such regeneration occurs quickly in zebrafish. Member A of the acidic nuclear phosphoprotein 32 (ANP32a) family is involved in neuronal development, but its function is controversial, and its involvement in zebrafish SCI remains unknown. To determine the role of zebrafish ANP32a in the neuronal regeneration of SCI embryos, we microinjected ANP32a mRNA into embryos from zebrafish transgenic line Tg(mnx1:GFP) prior to SCI. Compared to control SCI embryos, the results showed that the regeneration of spinal cord and resumption of swimming capability were promoted by the overexpression of ANP32a mRNA but reduced by its knockdown. We next combined fluorescence-activated cell sorting with immunochemical staining of anti-GFAP and immunofluorescence staining against anti-PH3 on Tg(gfap:GFP) SCI embryos. The results showed that ANP32a promoted the proliferation and cell number of radial glial cells at the injury epicenter at 24 h post-injury (hpi). Moreover, when we applied BrdU labeling to SCI embryos derived from crossing the Tg(gfap:GFP) and Tg(mnx1:TagRFP) lines, we found that both radial glial cells and motor neurons had proliferated, along with their increased cell numbers in Anp32a-overexpression SCI-embryos. In this study, a new framework of vision-based estimation is developed using some data fusion schemes to obtain previewed
road curvatures and vehicular motion states based on the scene viewed from an in-vehicle camera. The previewed curvatures
are necessary for the guidance of an automatically steering vehicle, and the desired vehicular motion variables, including
lateral deviation, heading angle, yaw rate, and sideslip angle, are also required for proper control of the vehicular lateral
motion via steering. In this framework, physical relationships of previewed curvatures among consecutive images, motion variables
in terms of image features searched at various levels in the image plane, and dynamic correlation among vehicular motion variables
are derived as bases of data fusion to enhance the accuracy of estimation. The vision-based measurement errors are analyzed
to determine the fusion gains based on the technique of a Kalman filter such that the measurements from the image plane and
predictions of physical models can be properly integrated to obtain reliable estimations. Off-line experimental works using

Industrial Internet of Things (IIoT) is an emerging technology that relies on the use of massively connected sensor nodes to gather industrial-related data. The collected data are used for postanalysis to generate insights for reducing production down time, cost optimization, and predictive maintenance. One of the key requirements for sensor node in such application is the data confidentiality; as such sensor data may potentially leak the manufacturing and industrial secret to their competitors. In this paper, a field programmable gate array (FPGA)-based sensor node with Advanced Encryption Standard (AES) crypto-processor is proposed to safeguard the sensor data. A novel queue system is proposed to further reduce the data processing time and energy consumption. The proposed queue system is able to achieve 1.48× speed up and ∼16% energy reduction, which makes it a competitive candidate for Industrial IoT applications. The technique developed in this paper can also be extended to implement FPGA-based gateway with encryption feature, which is very useful for edge computing in IoT applications. 相似文献
