Shared metabolomic patterns at delivery have been suggested to underlie the mother-to-child transmission of adverse metabolic health. This study aimed to investigate whether mothers with gestational diabetes mellitus (GDM) and their offspring show similar metabolomic patterns several years postpartum. Targeted metabolomics (including 137 metabolites) was performed in plasma samples obtained during an oral glucose tolerance test from 48 mothers with GDM and their offspring at a cross-sectional study visit 8 years after delivery. Partial Pearson’s correlations between the area under the curve (AUC) of maternal and offspring metabolites were calculated, yielding so-called Gaussian graphical models. Spearman’s correlations were applied to investigate correlations of body mass index (BMI), Matsuda insulin sensitivity index (ISI-M), dietary intake, and physical activity between generations, and correlations of metabolite AUCs with lifestyle variables. This study revealed that BMI, ISI-M, and the AUC of six metabolites (carnitine, taurine, proline, SM(-OH) C14:1, creatinine, and PC ae C34:3) were significantly correlated between mothers and offspring several years postpartum. Intergenerational metabolite correlations were independent of shared BMI, ISI-M, age, sex, and all other metabolites. Furthermore, creatinine was correlated with physical activity in mothers. This study suggests that there is long-term metabolic programming in the offspring of mothers with GDM and informs us about targets that could be addressed by future intervention studies.  相似文献

OBJECTIVE: The structure of the collagen scar during healing of a myocardial infarction is a determinant of the function of the remodeled tissue. We hypothesize that the passive deformations of both scar and normal tissue are related to the underlying collagen uncoiling as the tissue stretches, and that the unloaded tortuosity of the collagen may be a determinant of tissue stiffness at low ventricular pressure. Hence collagen uncoiling and tissue strain were measured during passive loading in normal tissue, and in healing infarct tissue. METHODS: Left ventricles of rats were infarcted by ligation of the left anterior descending artery for 2 weeks. Surface strains were measured during passive inflation in the scar region in one set of excised hearts, and other arrested hearts were fixed at different ventricular pressures, after which collagen tortuosity was measured in the infarcted and normal tissue. RESULTS: Passive loading strains were smaller in the scar in both the fiber and cross-fiber directions. Tortuosity decreased with load in normal and infarcted tissue, with fibrils tending to straighten more in the scar tissue at higher pressures (1.056 +/- 0.009 vs. 1.024 +/- 0.009 at P = 20 mmHg) with similar tortuosities at zero pressure (1.110 +/- 0.012 vs. 1.098 +/- 0.019). The decrease in tortuosity with strain was greater for the infarcted tissue. CONCLUSIONS: The greater stiffness of infarcted tissue at low pressure is not due to 'straightened' collagen fibers, and there may be a different three-dimensional structure of infarct vs. normal coiled collagen fibers which can affect the material properties of these tissues.  相似文献

The rapid advances in molecular biology have begun to shift many of the bottlenecks in genome research from the laboratory to the data analysis facility. The pace at which this has occurred creates a situation in which software development always has to catch up with the flow of data. Since such large-scale processes were not anticipated, the analysis infrastructure has not been fully established. Furthermore, most systems that have been built were designed by the biologists who collected the data. More recently, computer scientists, mathematicians, and engineers have taken an interest in this problem. This has had a positive effect, since it has created a tight synergy between the informatics and the biology. Several principles affected the design of the system developed at TIGR. Each of the sample preparation, sequencing, and analysis steps had to be managed, scheduled, and tracked. This information had to be made readily available to those who needed it for carrying out their tasks. Different skill levels of the users had to be taken into account. The degree of human intervention at each step had to be evaluated and built into the design. A mixed processing environment of Macintosh and Unix platforms had to be integrated. Most importantly, the system had to save time, reduce error, and ensure uniformity of the analysis and quality of the results. In the authors' experience, the tools they have built work well because of their early decisions as to which systems to use for development. IGF-1 and its receptors have been identified in many tissues including the central nervous system (CNS). We have previously demonstrated that injection of insulin directly into the cerebral ventricles (ICV) is followed by a drop in mean arterial pressure (MAP) associated with an increase in skeletal muscle blood flow. Given the similarities between the IGF-1 and insulin molecules and their respective receptors, we have investigated the effect of ICV administration of IGF-1 on systemic blood pressure and blood flow in selected vascular beds. ICV cannulas were implanted into normal rats and the animals were allowed to recover for 3 to 4 days. The femoral artery and vein were cannulated for blood pressure monitoring and blood sampling and blood flow probes placed around the iliac, the renal and the superior mesenteric artery were used to assess regional blood flow. ICV injection of IGF-1 resulted in a significant decrease in MAP with a nadir at 15 minutes and a gradual return to baseline by 60 minutes; heart rate increased 40 minutes after the injection. IGF-1 significantly enhanced vascular flow and conductance in the iliac, but not in the renal and superior mesenteric arteries. The effects of IGF-1 were much smaller than those observed previously with equimolar amounts of insulin. We conclude that IGF-1 can decrease MAP by selectively increasing blood flow to skeletal muscle through a direct action on the central nervous system.  相似文献

This article examines the psychological impact of participating in sigmoidoscopy screening for colorectal cancer prevention. The 1st study examined psychological well-being at 3 months, in relation to screening outcome, in 4,153 individuals. The 2nd study used longitudinal data to examine changes in psychological functioning from before to after screening in relation both to screening outcome and baseline indicators of vulnerability. There were few psychological differences between those who had received negative results or had polyps detected. These findings were confirmed in the longitudinal study, which also found no evidence for vulnerability to adverse effects among those who were initially most anxious or who perceived their risk of cancer to be higher. This study assessed the value of biplane transesophageal echocardiographic assessment of diastolic flow reversal in the descending aorta as an alternative to Doppler color flow imaging in determining severity of aortic regurgitation. In 45 patients undergoing cardiac operations, the severity of aortic regurgitation was assessed by semiquantitative grading of the width of the Doppler color flow regurgitant jet relative to the left ventricular outflow tract, and the presence of diastolic flow reversal was assessed with pulsed-wave Doppler measurements at three sites in the descending aorta. In four patients, the diastolic flow reversal method was the only available form of assessment because of inadequate visualization of the left ventricular outflow tract beneath a mitral valve prosthesis. Diastolic flow reversal in the descending aorta was not observed in patients without aortic regurgitation and was always present in patients with severe aortic regurgitation. Aortic valve replacement successfully eliminated descending aortic flow reversal in all 19 patients in whom it was present before valve replacement. Identification of diastolic flow reversal at multiple sites in the descending aorta with biplane transesophageal echocardiography helps to confirm the presence of severe aortic regurgitation and can serve as an alternative method of assessment when visualization of the left ventricular outflow tract is impaired.  相似文献

Intermediate phases were found to agree with previous work, but the homogeneity ranges and high-temperature phase relationships were in some cases at variance. Two studies examined whether the accessibility of performance prototypes influences performance appraisals. Pilot studies revealed students used performance prototypes when rating instructor performance. Study 1 manipulated the accessibility of these prototypes and the time delay of performance ratings. Results showed no effect of the prime on rating error and accuracy; however, discrimination accuracy decreased over time and recognition bias became more conservative. Study 2 manipulated prototype accessibility and type of rating stimuli (videotape vs. vignette). Rating accuracy and recall were higher for vignette than videotape stimuli, and only those participants exposed to the vignette exhibited priming effects. Results supported transfer-appropriate processing and implied that cognitive primes may have a stronger effect on performance ratings based on "paper-people" than videotaped stimuli. The Mean Opinion Scale (MOS) is a questionnaire used to obtain listeners' subjective assessments of synthetic speech. This paper documents the motivation, method, and results of six experiments conducted from 1999 to 2002 that investigated the psychometric properties of the MOS and expanded the range of speech characteristics it evaluates. Our initial experiments documented the reliability, validity, sensitivity, and factor structure of the P.L. Salza et al. (Acta Acustica, Vol. 82, pp. 650–656, 1996) MOS and used psychometric principles to revise and improve the scale. This work resulted in the MOS-Revised (MOS-R). Four subsequent experiments expanded the MOS-R beyond its previous focus on Intelligibility and Naturalness, to include measurement of the Prosody and Social Impression of synthetic voices. As a result of this work, we created the MOS-Expanded (MOS-X), a rating scale shown to be reliable, valid, and sensitive for high-quality evaluation of synthetic speech in applied industrial settings.  相似文献

Dilute solution viscometry was used to explore the effect of solid-state ageing on the interactions that occur between guar gum and water. The resulting data set, derived from nearly 700 independent experiments, led to a value for the Flory–Huggins interaction parameter, χ, of 0.56 ± 0.12. This value, which appeared independent of ageing, is in good agreement with the majority of published data. The effect of ageing on molar mass was also explored, using Mark–Houwink–Sakurada (MHS) theory. Absolute molar mass values were found to depend sensitively on the choice of MHS constants, but the effect of ageing was unequivocal; under all conditions, it resulted in a pronounced decrease in molar mass. In concert, these results strongly suggest that, in guar, solid-state ageing reactions are largely associated with scission of the molecular backbone. This hypothesis was then tested by infra-red and Raman spectroscopy. Although infra-red spectroscopy did reveal some subtle differences between the spectra of guar and locust bean gum (LBG), a related polysaccharides with a different galactose:mannose ratio, no equivalent effects were seen in aged guar. However, clear differences between the Raman spectra of guar and LBG were seen, demonstrating that the technique is well capable of revealing changes in galactose:mannose ratios. Examination of aged guar samples revealed no comparable effects, reinforcing the notion that, in this polysaccharide, chain scission reactions dominate such that solid-state ageing does not lead to changes in the nature of its interaction with water.  相似文献
