The lysis of group AB erythrocytes by human complement was studied by different anti-A and anti-B IgM monoclonal antibodies (mabs) in a 51Cr-release assay. The concentration of membrane-bound immunoglobulin was detected by ELISA, and the amount of C1q and C3 bound to sensitized red cells was measured by using purified, 125I-labelled molecules. We have demonstrated that there is an exponential relationship between the concentration of the sensitizing IgM mabs and C1q binding to the sensitized AB cell. The efficiency of binding was related to the number of antibodies bound; thus, anti-A sensitized cells bound 3-6 times more C1q than anti-B sensitized cells did. AB cells, on the other hand, bound similar amounts of C3 whether anti-A or anti-B was present. The lytic efficiencies of the various IgM mabs during short incubation times were different, suggesting that the complement activation rates vary widely with different antibodies on the AB cell membrane. The binding of C1q to an antibody-sensitized target activates a cascade, whose components may migrate away from the sensitizing antibody; interactions between the activation processes generated by the anti-A and anti-B antibodies may thus occur. Choosing appropriate pairs of anti-A and anti-B mabs for the simultaneous sensitization of AB cells has indeed resulted in stimulation in some and inhibition in other combinations of mabs. We recently demonstrated that chemical proteasome inhibition induced inner retinal degeneration, supporting the pivotal roles of the ubiquitin–proteasome system in retinal structural integrity maintenance. In this study, using beclin1-heterozygous (Becn1-Het) mice with autophagic dysfunction, we tested our hypothesis that autophagy could be a compensatory retinal protective mechanism for proteasomal impairment. Despite the reduced number of autophagosome, the ocular tissue morphology and intraocular pressure were normal. Surprisingly, Becn1-Het mice experienced the same extent of retinal degeneration as was observed in wild-type mice, following an intravitreal injection of a chemical proteasome inhibitor. Similarly, these mice equally responded to other chemical insults, including endoplasmic reticulum stress inducer, N-methyl-D-aspartate, and lipopolysaccharide. Interestingly, in cultured neuroblastoma cells, we found that the mammalian target of rapamycin-independent autophagy activators, lithium chloride and rilmenidine, rescued these cells against proteasome inhibition-induced death. These results suggest that Becn1-mediated autophagy is not an effective intrinsic protective mechanism for retinal damage induced by insults, including impaired proteasomal activity; furthermore, autophagic activation beyond normal levels is required to alleviate the cytotoxic effect of proteasomal inhibition. Further studies are underway to delineate the precise roles of different forms of autophagy, and investigate the effects of their activation in rescuing retinal neurons under various pathological conditions. 相似文献

Nine multiparous Holstein-Friesian cows (initially 97 d in milk), were used in a 3 × 3 lattice square design experiment with 4-wk periods. All cows received 4 kg/d concentrates and dietary treatments were based on silages offered ad libitum: perennial ryegrass (PRG); timothy (TIM); tall fescue (TF); red clover (RC); red clover/corn silage mixture [40/60 on a dry matter (DM) basis; RCC]; red clover/whole-crop oat silage mixture (40/60 on a DM basis; RCO); or red clover/whole-crop oat silage mixture (25/75 on a DM basis; ORC). The remaining treatments were based on RCO with feed intake restricted to the level of PRG (RCOr) or with a low protein concentrate (50/50 mixture of barley and molassed sugar beet pulp; RCOlp). Experiment objectives were to evaluate diet effects on N partitioning and N isotopic fractionation. Yields of milk and milk protein were consistently high for diets RC, RCC, and RCO and low for the diets based on poorly ensiled grass silages. Restriction of intake (RCOr) and inclusion of a higher proportion of whole-crop oat silage (ORC) and the low-protein concentrate (RCOlp) led to some loss of production. Diet had little effect on milk fat, protein, and lactose concentrations: low concentrations of milk protein and lactose reflect the restricted energy intakes for all treatments. The highest diet digestibilities were measured for RC and PRG, whereas increasing inclusion of the whole-crop oat silage (0, 60, and 75% of forage DM) led to a marked decrease in diet digestibility (0.717, 0.624, and 0.574 g/g, respectively). Urinary excretion of purine derivatives, an indicator for rumen microbial protein synthesis, was significantly higher for RCC than for TIM and TF. Nitrogen intake ranged between 359 and 626 g/d (treatment means). Partitioning of N intake to feces and urine was closely related to N intake, although urinary N losses were less than predicted from N intake for the 60/40 mixtures of cereal silage and red clover silage. The 15N content of milk, urine, and feces were all influenced by diet 15N content. Isotopic fractionation meant that feces and milk were enriched and urine was depleted in 15N relative to the diet. Significant relationships were observed between the extent of enrichment of urine, feces, and milk, suggesting some commonality in fractionation pathways. The trend for the lowest 15N enrichment in milk protein occurring in diets with low N-use efficiency (milk N/feed N) was contrary to expectations, possibly because of endogenous contributions to milk protein or fractionation when dietary ammonia was incorporated into microbial protein. 相似文献

The nuclear shielding constants in OCS are studied using ab initio theoretical methods and gas-phase NMR measurements. The shielding surfaces are calculated and the rovibrational effects and the resulting temperature dependence are analyzed. The temperature dependence of 13C shielding in the gas phase is determined experimentally in the range 278-373 K. 13C is the single nucleus for which the experimental data for the temperature dependence can be converted to a reference-independent scale, and good agreement of the measured and calculated ab initio results is observed. 48 preschool-aged children's knowledge of the distinctions between animate and inanimate objects was assessed by showing them stimulus films of animate and inanimate objects that moved in different ways. Responses gathered included attributions of animate or inanimate properties, justifications for attribution choices and accuracy in labeling objects as alive or not. Results indicate that 5-yr-olds and some 4-yr-olds performed near the levels of 16 college-aged adults, whereas 3-yr-olds did not, although the animate–inanimate distinction did mediate their behavior. A generalized "animistic" attitude was not found: rather, developmental changes appeared to accrue from increasing precision about the specific properties that do and do not distinguish animate from inanimate objects.
