Conventional enzyme-linked immunosorbent assay (ELISA) is a gold standard for screening antibodies and testing for protein or antigen presence. A significant limitation of this assay resides in the fact that only one analyte can be assessed per microplate well. Here, we describe and investigate a new technology consisting of an automated ELISA system in which up to 10 analytes can be measured within one single well, thus improving productivity, accuracy, and repeatability by reducing the amount of human labor required. Another strength of the platform is that a user can load any necessary sets/subsets of beads to perform required assays, with improved flexibility compared to manufactured-loaded arrays for multiplex analysis. We also demonstrate that this system can be used to determine the pathogenicity (i.e., presence of Shiga toxins) and serotype (i.e., Escherichia coli O157) of E. coli isolates.  相似文献

The experience of the Mississippi State University (MSU) Libraries illustrates the challenges presented by moving from consortial to local chat, as well as an evaluation of the advantages of chat reference in an academic setting. Countering the consortial, instant-messaging model prevalent in virtual reference today, the MSU Libraries have found chat offers advantages in an academic community. Moving to a local setting, it was possible for the MSU Libraries to maintain quality control over chat transactions while building relationships with faculty and students across campus.  相似文献

Degradation rates of electrical current during constant voltage operation of SOFCs with anodes made using NiO precursor powders from two different manufacturers with and without the addition of aluminum titanate (ALT) added by either mechanical mixing or anode infiltration have been quantified using a novel MATLAB algorithm. Because the algorithm has been used to quantify degradation rates for many different SOFC tests, it is thought that the method can be applied to most measured SOFC data to quantify the instantaneous cell degradation rate as a function of time for the entire SOFC performance measurement. Degradation rates determined at different times have been plotted against varying concentrations of ALT addition, facilitating the estimation of optimum ALT concentration for SOFC anodes made with NiO from a specific manufacturer. The algorithm used to determine degradation rates is available upon request to the corresponding author.  相似文献

The objectives of this study were to estimate genetic correlations among body condition scores (BCS) from various sources, dairy form, and measures of cow health. Body condition score and dairy form evaluated during routine type appraisal was obtained from the Holstein Association USA, Inc. A second set of BCS was obtained from Dairy Records Managements Systems (DRMS) and was recorded by producers that use PCDART dairy management software. Disease observations were obtained from recorded veterinarian treatments in several dairy herds in the United States. Estimated breeding values for diseases in Denmark were also obtained. Genetic correlations among BCS, dairy form, and cow health traits in the United States were generated with sire models. Models included fixed effects for age, DIM, and contemporary group. Random effects included sire, permanent environment, herd-year season for health traits, and error. Predicted transmitting abilities (PTA) for BCS and dairy form were correlated with estimated breeding values for disease in Denmark. The genetic correlation estimate between BCS from DRMS and BCS from the Holstein Association USA, Inc., was 0.85. The genetic correlation estimate between BCS and a composite of all diseases in the United States was -0.79, and PTA for BCS was favorably correlated with an index of resistance to disease other than mastitis in Denmark (0.27). Dairy form was positively correlated with a composite of all diseases in the United States (0.85) and was unfavorably correlated with an index for resistance to disease other than mastitis in Denmark (-0.29). Adjustment for protein yield PTA had a minimal affect on correlations between PTA for BCS or dairy form and disease in Denmark. Selection for higher body condition or lower dairy form with continued selection for yield may slow deterioration in cow health as a correlated response to selection for increased yield.  相似文献

Abstract

Ion selective electrodes are used to conduct real-time measurements of gaseous thermal degradation products from halon replacement experiments. Response characteristics of these ion selective electrodes are needed to address potential interferences and surface poisoning experienced under field conditions. The design and the fabrication of an in situ, converging wall-jet instrument for the measurement of the dynamic responses of ion selective electrodes are described. Various time delays associated with the generation and detection of ion activity steps have been quantitatively determined.

Dynamic response measurements for iodide steps on a Corning iodide/cyanide ion selective electrode are comparable to those obtained on the best of the earlier instrument designs, the in situ, switched wall-jet instrument. The advantages of the converging wall-jet instrument over the switched wall-jet instrument are identified.  相似文献

Tylosin and chlortetracycline (CTC) are antimicrobial chemicals that are fed to >45% of the US swine herds at therapeutic and sub-therapeutic dosages to enhance growth rates and treat swine health problems. These compounds are poorly absorbed during digestion so that the bioactive compound or metabolites are excreted. This study investigated the degradation and stabilization of swine manure that contained no additives and compared the observed processes with those of manure containing either tylosin or CTC. The batch anaerobic incubation lasted 216 days. The breakdown of insoluble organic matter through anaerobic hydrolysis reactions was faster for manure containing CTC compared with tylosin or no-antimicrobial treatments. Volatile fatty acid (VFA) accumulation, including acetate, butyrate, and propionate, was greater for CTC-containing manure compared to tylosin and no-antimicrobial treatments. The relative abundance of two aceticlastic methanogens, Methanosaetaceae and Methanosarcinaceae spp., were less for CTC manure than manure with no-antimicrobial treatment. In addition, generation of methane and carbon dioxide was inhibited by 27.8% and 28.4%, respectively, due to the presence of CTC. Tylosin effects on manure degradation were limited, however the relative abundance of Methanosarcinaceae spp. was greater than found in the CTC or no-antimicrobial manures. These data suggest that acetate and other C-1 VFA compounds would be effectively utilized during methanogenesis in the presence of tylosin.  相似文献

Depth of penetration experiments were conducted with fragment simulating projectiles launched into Celotex® in order to develop an equation for the strike velocity as a function of the FSP mass and the depth of penetration into Celotex® recovery media. A powder gun launched FSPs, designed in accordance with STANAG-2920 [NATO STANAG-2920 Ballistic Test Method for Personal Armour Materials and Combat Clothing, 2nd ed., 1999.] weighing between 0.13 g and 53.78 g at striking velocities between 198 m/s and 1524 m/s. A multiple linear regression analysis was used to determine an empirical relationship for the strike velocity to the impact parameters of depth of penetration, fragment mass, and mean presented area. Sabot launched natural fragments weighing between 2.8 g and 15.8 g at striking velocities between 532 m/s and 1084 m/s were used to validate the equation.  相似文献

Estimates of daughter fertility were computed using first artificial insemination (AI) breedings reported to the US Dairy Herd Improvement Association (DHIA) from 1995 through 1997. An animal model was used to compute estimated breeding values (EBV) of daughter groups with fixed effects of herd-year-month bred and classes of early lactation energy-corrected milk, days in milk (DIM) when bred, and parity. Standard deviations and ranges of bull EBV for daughter fertility for DIM were 9.1 and -31 to 18; standard deviations and ranges of bull EBV for daughter fertility for nonreturn were 3.8 and -11 to 10. Correlations were computed for EBV for daughter fertility with EBV for mating bull fertility and with predicted transmitting abilities (PTA) for milk, somatic cell score (SCS), and productive life for bulls (213) with minimums of 200 matings and 100 progeny with reproductive traits. None of the correlations among EBV for reproductive traits differed from 0.0. Correlations of EBV for daughter fertility with PTA for productive life were significantly positive. PTA for yield traits were not correlated with EBV for daughter differences in nonreturn or DIM. Very low correlations of EBV for daughter reproductive traits with PTA for yield indicate that, in order to improve daughter fertility, fertility must be incorporated in sire selection decisions.  相似文献

We have developed a new type of chemical microsensor based on piezoresistive microcantilever technology. In this embedded polymer microsensor, a piezoresistive microcantilever is partially "embedded" into a polymeric material. Swelling of the polymer upon analyte exposure is measured as a simple resistance change in the embedded cantilever. Arrays of these sensors, each employing a different polymeric material, provide for the identification of a wide range of chemical vapor analytes. Advantages of this system over previous "surface" piezoresistive microcantilever chemical sensors include enhanced mechanical simplicity (no mechanical approach necessary), greater resistance to shock or movement, and lower cost.  相似文献
